Genetic characterization of Xylella fastidiosa isolated from citrus and coffee plants Scientia Agricola

The Citrus Variegated Chlorosis and the Coffee Leaf Scorch are some of the many destructive diseases caused by Xylella fastidiosa, a gram-negative bacterium limited to the xylem of affected plants. As its genetic characterization is still not well established, different isolates of X. fastidiosa from citrus and coffee were evaluated through RAPD (Random Amplified Polymorphic DNA) technique to characterize and classify these isolates based on similarity coefficients. Sixteen isolates of X. fastidiosa were used on this trial, obtained from citrus, coffee and almond. The genetic polymorphism evaluation was performed using six arbitrary 10-base primer pairs. Isolation of Rhodocyclus gelatinosus from poultry slaughterhouse wastewater BABT

Four cultures of photosynthetic bacteria isolated from poultry slaughterhouse wastewater were identified as Rhodocyclus gelatinosus based on the following properties: reddish color of cultures in synthetic medium, presence of motility, slightly curved Gram-negative rods morphology, gelatin liquefying activity, utilization of citrate as carbon source and production of bacteriochlorophyl a and carotenoids of the spirilloxanthin alternative series. R. gelatinosus may represent a source of nutrients and pigments with application in poultry feed.

Chemical composition of Rhodocyclus gelatinosus biomass produced in poultry slaughterhouse wastewater BABT

Rhodocyclus gelatinosus R1 grew photoautotrophically in poultry slaughterhouse wastewater inside glass columns (90x670 mm) during 7 days at 31 ± 4ºC, under anaerobiosis and lightness supplied by daylight plus 3 (100W) incandescent and 4 (40W) fluorescent lamps. The culture was centrifuged (4,500xg/20 min) and lyophilized to originate a bacterial biomass with 7.1% moisture content. Chemical composition investigation showed 67.6% crude protein, 27.6% total carbohydrate, 0.6% lipids and 4.2% ash (dry weight). Amino acid composition of the biomass was similar to others described in the literature for Rhodocyclus gelatinosus and for other photosynthetic bacteria. Bioassay assessment of metarhizium anisopliae (metchnikoff) sorokin (deuteromycota: hyphomycetes) against Oncometopia facialis (signoret) (hemiptera: cicadellidae) BJM

Citrus Variegated Chlorosis (CVC) is an economically important, destructive disease in Brazil and is caused by Xylella fastidiosa and transmitted by sharpshooter insects. In this study, the efficacy of the fungus Metarhizium anisopliae in controlling the sharpshooter Oncometopia facialis was studied by bioassay conditions. In the bioassay, insects were sprayed with a suspension containing 5 X 10(7) conidia mL-1. Adults captured in the field were treated in groups of 10 in a total of 11 replications per treatment. Significant differences between the natural mortality and the mortality of insects treated with the fungus were observed 6 days after inoculations (P<0.05).
