Responses and adaptations of the adenylate energy charge and digestive enzyme activities to tidal emersion of Crassostrea gigas populations in Marennes-Oleron Bay ArchiMer
Moal, Jeanne; Samain, Jean-francois; Le Coz, Jean-rene; Daniel, Jean-yves.
Crassostrea gigas oysters cultivated in Marennes-Oleron Bay (France), face seasonal variations in the environment: salinity, turbidity, phytoplanktonic production, and the tidal rhythm of emersion. The oysters are maintained at different depths to reduce competition for space (e.g. by settlement of mussel spat). The physiological responses of two groups of oysters to different periods of emersion were studied using a seasonal sampling strategy: one group was on a flat part of the area, and the effect of emersion was studied in a short time survey (less than three hours emersion at low tide); the other was located in a very narrow place with a steep slope. This group was subdivided into three, according to bathymetric position, to study the long term effect...

Model for Rapid Detection of Sublethal Effects of Pollutants. II. An in situ Example: Anomalous Metabolism of the Hyponeustonic Copepod Anomalocera patersoni Coinciding with an Oil Spill An unusual correlation has been obsemed between the amylase and trypsin content of a natural zooplankton population and of the hyponeustonic copepod Anomalocera patersoni and the oil spill of the "Amoco Cadiz" in 1978. This anomaly has also been obsewed in Artemia sp. cultures (San Francisco Bay strain) poisoned by copper. This phenomenon could originate from a general physiological perturbation. Such a non-specific perturbation could explain the anormal correlation between parameters which are usually independent and specifically regulated. The determination of such...

Effect of sediment nearness on the metabolic enzyme activity and energy state of the oyster Crassostrea gigas ArchiMer
Le Moullac, Gilles; Fleury, Pierre-gildas; Le Coz, Jean-rene; Moal, Jeanne; Samain, Jean-francois.
This study was designed to assess in situ the effects of sediment nearness on Crassostrea gigas metabolism. One year-old oysters were reared for 5 months (April-August) in plastic bags on metallic frames at 10 and 60 cm off the sediment at the beginning of April. The management of the energetic resources (storage, consumption) was estimated with respect to biochemical changes of proteins, lipids and carbohydrates. In order to evaluate the effect of the sediment nearness at cellular level, the metabolic rate was evaluated by assessing pyruvate kinase (PK) and phosphoenolpyruvate carboxykinase (PEPCK) enzyme activities and electron transport system (ETS) activity. The metabolic pathways were assessed by measuring ATP and intermediary metabolites such as...

Possible processes of nutritive adaptations for zooplankton : a demonstration on Artemia ArchiMer
Samain, Jean-francois; Moal, Jeanne; Daniel, Jean-yves; Le Coz, Jean-rene.
The ingestion rate and the resulting assimilation calculated by means of a law taking into account activities of the digestive enzymes (amylase and trypsin) on the carbohydrates and proteins ingested, are determined in two experiments on Artemia. They are fed the same species of phytoplankton (Tetraselmis suecica), at the same concentration but with two chemical compositions. In one experiment, the cells are rich in carbohydrates and poor in proteins, and in the second one they are poor in carbohydrates, rich in proteins. Different observed ingestion rates induce a balance in proteins, and a large difference in the carbohydrates ingested. When digestive enzymes are taken into account, the assimilated carbohydrates and proteins are similar (explaining the...

Signification biologique des teneurs protéiques et des activités de l'amylase et des protéases chez Artemia salina L. aspects d'application a l'étude de la nutrition ArchiMer
Samain, Jean-francois; Boucher, Jean; Buestel, Dominique; Borne, M.; Le Coz, Jean-rene.
The relations between the activity of the digestive enzymes amylase and proteases were studied on Artemia salina L. For this purpose, a method for measuring the ingestion was developed compatible with the speed of response of the digestive enzymes. It is demonstrated that the proteic density variations in Artemia of the same size are in relation to the food content of their digestive tract. Therefore the measurement of the size and weight of proteins (proteic density) allows the evaluation of the ingestion. With the aid of this method, a daily ingestion rhythm, with bimodal character, could be demonstrated. The influence on the ingestion, of the concentration of food available (Tetraselmis suecica Butcher) was studied in function of the size of the...

Oyster adenytate energy charge: response to levels of food ArchiMer
Moal, Jeanne; Le Coz, Jean-rene; Samain, Jean-francois; Daniel, Jean-yves; Bodoy, Alain.
Adenylatc Encrgy Charge (AEC) values of Crassostrea gigas (9 and 18 months old) cultured in ponds in Marennes-Oléron Bay (Atlantic coast, France) are reported. Two ponds were loaded with two different initial oyster biomasses in order to obtain different food conditions. The growth of oysters was different in the two ponds which corroborated the effectivences of attaining different food conditions. AEC level was different depending on the pond and was therefore related to food availability per unit oyster biomass. Well-fed oysters exhibited greater AEC values than poorly-fed oysters, throughout thc study. Information on nutritional situations for wild or cultured populations can therefore be obtained by AEC. In cach pond, the seasonal AEC variations were...

Nutritional value of six Pavlovophyceae for Crassostrea gigas and Pecten maximus larvae ArchiMer
Ponis, Emanuele; Probert, I; Veron, Benoit; Le Coz, Jean-rene; Mathieu, Michel; Robert, Rene.
Four unidentified new strains of Pavlovophyceae [Pavlova sp. AC 250 (M), Pavlova sp. AC 248 (Psh), Pavlova sp. AC 251 (Pth), Pavlova sp. AC 538 (Psm)] and two known species [Pavlova pinguis (Ppi), Rebecca salina (ex Pavlova salina, Rsa)] were characterized (productivity, size, dry weight, ash, gross composition, fatty acids, sterols) and their nutritional value in bispecific diets were evaluated both on Crassostrea gigas and Pecten maximus larvae. All microalgae exhibited poor food value for C gigas larvae. P. pinguis and R. salina did not sustain any growth, like the control starved larvae. No exo-toxicity of P. pinguis and R. salina was detected. These two species were weakly ingested by C. gigas larvae, while the four other pavlovophytes were...

Effect of reproduction on escape responses, metabolic rates and muscle mitochondrial properties in the scallop Placopecten magellanicus ArchiMer
Kraffe, E; Tremblay, R; Belvin, S; Le Coz, Jean-rene; Marty, Y; Guderley, H.
In scallops, gametogenesis and spawning can diminish the metabolic capacities of the adductor muscle and reduce escape response performance. To evaluate potential mechanisms underlying this compromise between reproductive investment and escape response, we examined the impact of reproductive stage (pre-spawned, spawned and reproductive quiescent) of the giant scallop, Placopecten magellanicus, on behavioural (i.e., escape responses), physiological (i.e., standard metabolic rates and metabolic rates after complete fatigue) and mitochondrial capacities (i.e., oxidative rates) and composition. Escape responses changed markedly with reproductive investment, with spawned scallops making fewer claps and having shorter responses than pre-spawned or...

Bi-parental families were produced in hatchery and tested in the field in 2001. Two sets of 5 families were constituted, selected on their high (R) and low (S) survival. These two sets were reared in Brittany from March to November 2002. Samplings were performed twice a month to obtain data on biometry, survival, reproductive cycle, biochemical composition, adenylate energy charge, hemolymph parameters (ions and defense system) and muscle strength. "R" and "S" oysters exhibited different reproductive effort and spawning strategy. "R" oysters allocated less energy in gonad than "S" ones and presented a complete spawning at the end of August contrary to the "S" which spawns partially. Mortality started in July when the seawater temperature reached 19 degree...

Effet de la qualité de l'eau sur la croissance larvaire de Pecten maximus en écloserie : observations préliminaires ArchiMer
Samain, Jean-francois; Cochard, Jean-claude; Chevolot, Lionel; Daniel, Jean-yves; Jeanthon, C.; Le Coz, Jean-rene; Marty, Y.; Moal, Jeanne; Prieur, D.; Salaun, Michele.
Some positive growth differences can be observed, all year long, in Pecten maximus larvae cultivated in an old fishpond, compared with the water which flows into it periodically. These waters are filtered at 1 µm through CUNO cartridges. Moreover, the larvae are cultivated in the same light, temperature and food (cultured phytoplankton) conditions. This difference is significant in terms of larvae size, dry weight, organic matter, ashes, total proteins, glycogen, and lipids. A series of preliminary observations on the chemical composition of these two waters after filtration (amino acids, fatty acids, organic mater, nutritive salts), on the bacterial flora (total, heterotrophic organisms, vibrio), on the particular material using electron microscopy, on...

Influence of phytoplankton diet mixtures on microalgae consumption, larval development and settlement of the Pacific oyster Crassostrea gigas (Thunberg) ArchiMer
Rico Villa, Benjamin; Le Coz, Jean-rene; Mingant, Christian; Robert, Rene.
Microalgae commonly used as feed for bivalves, Pavlova lutheri (P), Isochrisis affinis galbana (T) and Chaetoceros calcitrans forma pumilum (Cp), were fed to Pacific oyster Crassostrea gigas to assess their nutritional value for larval development and metamorphosis during two experiments. Monospecific, bispecific and trispecific diets were firstly evaluated during 3 weeks from D larvae to young postlarvae. Then bispecific diets, based on different T and Cp proportions, were assessed during a similar period. Concurrently, ingestion was studied through the whole larval and postlarval development for each diet and/or diet mixture. Because lipids are assumed to be a key nutrient for bivalves, biochemical analysis was undertaken on the second set of trials...

Biological, physiological, immunological and nutritional assessment of farm-reared Litopenaeus stylirostris shrimp affected or unaffected by vibriosis ArchiMer
Mugnier, Chantal; Justou, Carole; Lemonnier, Hugues; Patrois, Jacques; Ansquer, Dominique; Goarant, Cyrille; Le Coz, Jean-rene.
Shrimp aquaculture in New Caledonia is subject to seasonal mortalities during grow-out due to highly virulent Vibrio nigripulchritudo (Vn). To understand the mechanisms affecting shrimp resistance and leading to significant mortality, a shrimp ecophysiology and immunology survey was conducted on two farms, the first considered as a "control" farm (HC), the second affected by the disease (DF). Mortality observed during the survey at DF started 50 days after stocking and was typical of this disease. The main observations regarding shrimp were: (a) growth was not affected by the disease and was faster in the DF than in the HC pond; (b) disease did not affect one sex more than the other, or a specific part of the population in terms of weight; (c) the physical...

Sterol production and phytosterol bioconversion in two species of heterotrophic protists, Oxyrrhis marina and Gyrodinium dominans ArchiMer
Chu, F; Lund, E; Littreal, P; Ruck, K; Harvey, E; Le Coz, Jean-rene; Marty, Y; Moal, Jeanne; Soudant, P.
The kinetics and efficiency of sterol production and bioconversion of phytosterols in two heterotrophic protists Oxyrrhis marina and Gyrodinium dominans were examined by feeding them two different algal species (Rhodomonas salina and Dunaliella tertiolecta) differing in sterol profiles. R. salina contains predominantly brassicasterol (99%) and < 2% cholesterol. The major sterols in D. tertiolecta are ergosterol (45-49%), 7-dehydroporiferasterol (29-31%) and fungisterol (21-26%). O. marina fed R. salina metabolized dietary brassicasterol to produce 22-dehydrocholesterol and cholesterol. O. marina fed D. tertiolecta metabolized dietary sterols to produce cholesterol, 22-dehydrocholesterol, brassicasterol and stigmasterol. G. dominans fed either R. salina...

The purpose of this study is to examine the response of the oyster Crassostrea gigas to sulphide at different biological scales. A first experiment was designed to measure for 20 h the clearance (CR) and oxygen consumption (OC) rates of oysters exposed at different concentrations of sulphide (0, 3, 6, 12, 20, and 40 mu M). The second experiment was carried out to evaluate the metabolic adaptations to chronic sulphide exposure for 10 days by measuring PK and PEPCK enzyme activities, succinate and alanine content, the adenylate energy charge (AEC), and the activity of the electron transport system (ETS)in whole oysters at above 20 mu M. PK and PEPCK mRNAs and enzyme activities have been measured in adductor muscle. When exposed to sulphide above 20 mu M, CR...

Biological characteristics of European hake (Merluccius merluccius) sperm ArchiMer
Groison, Anne-laure; Suquet, Marc; Cosson, Jacky; Le Coz, Jean-rene; Jolivet, Aurelie; Garren, Francois.
Very little is known on European hake's reproductive biology and especially on biological characteristics of its sperm despite a growing interest in its aquaculture potential. This study reports, for the first time some hake sperm characteristics. After activation, the swimming phase lasts 3 min (8 min when activated with 50% sea water (SW) but lower initial velocity). The initial flagellar beat frequency (BF), velocity and percentage of motile cells decreased after 100 s. After 2 days at 4 degrees C, the mean Adenylate Energy Charge (AEC) level was 0.71. Sperm stored at 4 degrees C still showed motility after 9 days for 2 individuals. When cryopreserved, the motility recovery index of thawed spermatozoa ranged from 11.8% to 29.6%.

Changes in fatty acid and sterol composition during oogenesis in the pearl oyster Pinctada margaritifera ArchiMer
Vahirua Lechat, I; Laure, F; Le Coz, Jean-rene; Bianchini, J; Bellais, M; Le Moullac, Gilles.
The fatty acid and sterol composition of the oyster Pinctada margaritifera during oogenesis and in eggs was analysed. No major differences were observed during oogenesis, but the egg composition was significantly different from that of gonads. The amount of saturated fatty acids was the highest in eggs and the C16:0 predominant (P < 5%); by contrast, the amount of 22:6(n-3) was significantly lower (P < 5%) than in gonads. No major differences were observed for the polar lipid (PL) composition during oogenesis. The main free sterols in gonads and eggs were cholesterol and brassicasterol. Among free sterols, the proportion of cholesterol diminished continuously from the beginning to the end of gonad maturation, and this decrease persisted in eggs after...
