Two Leptospira sp. isolates were obtained by the first time from goats in Brazil and characterized by sequencing rrs, rpoB and secY genes, PFGE and typing with monoclonal antibodies. Both isolates are identical and belong to Leptospira santarosai. Analysis of the rrs and the rpoB genes sequences revealed 100% identity between the goat isolates and the Bananal reference strain. When secY sequences of the two isolates were compared to each other, it was observed that they had identical sequences. However, when compared to that of the Bananal reference strain, there were 15 mismatches along the 549 bp secY sequence. Forty-four cows from five herds infected with tuberculosis (TB) and without paratuberculosis (PTB), and 21 cows from a herd without either infection were studied. The cattle presented concordant results in both the skin test and gamma-interferon assay for TB and two commercial ELISAs for PTB. Animals were divided according to TB test results into Group A with 28 TB-infected animals, Group B with 16 TB-negative animals from infected herds, and Group C with 21 TB-negative cows from a tuberculosis-free herd (which were used as controls). Isolates from suggestive bovine tuberculosis lesions were tested by a multiplex polymerase chain reaction (m-PCR) targeting for RvD1Rv2031c and IS6110 sequences, specific for M. bovis and Mycobacterium tuberculosis complex respectively. Bovine tuberculosis, a chronic infection in cattle caused by Mycobacterium bovis, remains an economic and public health problem for several countries. Due to its economic impact on international trade, contagious nature, and implications for human health, global programs to eradicate the disease were implemented worldwide. Those programs are based on slaughtering PPD-reactive animals. Despite the National Programs in Brazil, complete eradication has not been achieved, and the disease remains, albeit at a lower prevalence. Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Moreover, bacterial growth is slow and test results are available only after three to five months of incubation Furthermore, shedding of bacilli at levels detectable by fecal culture is irregular and does not occur during the early stages of infection, which compromises the sensitivity of this methodology. Leptospires have never been recovered from goats in Brazil. Serum samples were obtained from 248 goats from Rio de Janeiro and from the seroreactive animals, urine samples were collected and processed for Leptospira isolation. A total of 52 positive reactions were observed, corresponding to 20.9% of the samples. The most prevalent reactions were to serovars Hardjo (36.5%), Shermani (30.8%), Icterohaemorrhagiae (9.6%), Grippotyphosa (9.6%), Autumnalis (5.8%), Castellonis (3.8%) and Bratislava (3.8%). Two strains of Leptospira sp. were isolated, both in the same region, but from different flocks. Presumptive identification based on serologic methods suggests those strains to be from Grippotyphosa serogroup. |

ABSTRACT: Leptospirosis is a zoonosis that is widely distributed in tropical countries. This infection is also associated with reproductive losses in livestock, which has a significant economic impact. The objective of this study is to investigate the seroprevalence and risk factors associated with leptospiral infection in commercial sheep flocks from the northwestern mesoregion of Rio Grande do Sul. Serum samples were analyzed in 319 sheep using the Microscopic Agglutination Test (MAT). At the time of sampling, an individual and herd questionnaire was carried out to investigate the risk factors. ABSTRACT: Sheep breeding is an economic activity of great value for the Brazilian livestock industry, and infectious diseases can significantly compromise its productivity. The aim of this study was to detect Leptospira spp. in mixed breed sheep from a semiarid region of the State of Paraíba, Northeast Brazil. In total, 49 blood and renal tissue samples, as well as 25 urine samples, were collected from 49 sheep in the slaughter line. ABSTRACT: This study evaluated the presence of vaginitis and the bacterial load associated with different intravaginal implants in ewes. Twenty-four Dorper and crossbred ewes were allocated into three groups and received intravaginal implant containing 0.3g progesterone (CIDR(r)), 60mg MAP or sponges without progesterone (CONTROL) for six days. Then, CIDR and MAP treated-ewes received 12.5mg dinoprost and 300IU eCG. Vaginal mucus samples were collected at four times: before device insertion, at the day of its removal, 24 and 48 hours after. The samples were cultured and the colonies were counted (CFU/mL) and identified.
