Preparation, characterization and in vitro antimicrobial activity of liposomal ceftazidime and cefepime against Pseudomonas aeruginosa strains BJM
Torres,Ieda Maria Sapateiro; Bento,Etiene Barbosa; Almeida,Larissa da Cunha; Sá,Luisa Zaiden Carvalho Martins de; Lima,Eliana Martins.
Pseudomonas aeruginosa is an opportunistic microorganism with the ability to respond to a wide variety of environmental changes, exhibiting a high intrinsic resistance to a number of antimicrobial agents. This low susceptibility to antimicrobial substances is primarily due to the low permeability of its outer membrane, efflux mechanisms and the synthesis of enzymes that promote the degradation of these drugs. Cephalosporins, particularty ceftazidime and cefepime are effective against P. aeruginosa, however, its increasing resistance has limited the usage of these antibiotics. Encapsulating antimicrobial drugs into unilamellar liposomes is an approach that has been investigated in order to overcome microorganism resistance. In this study, antimicrobial...

Grandisin induces apoptosis in leukemic K562 cells BJPS
Cortez,Alane Pereira; Menezes,Elizabeth Gomes Paulino; Benfica,Polyana Lopes; Santos,Alexandre Pereira dos; Cleres,Larissa Moreira; Ribeiro,Higor de Oliveira; Lima,Eliana Martins; Kato,Massuo Jorge; Valadares,Marize Campos.
Abstract In this study, the potential antileukemic activity of grandisin, a lignan extracted from Piper solmsianum, was evaluated against the leukemic line K562. The cytotoxicity of grandisin (0.018 to 2.365 µM) was evaluated in K562 and normal peripheral blood lymphocytes by Trypan Blue Exclusion and MTT methods after 48h exposure to the drug. In both methods, cellular viability was concentration-dependent and the IC50 values were lower than 0.85µM. Analysis of K562 cells after treatment with grandisin showed that the cell cycle was arrested in the G1 phase with a 12.31% increase, while both S and G2 phases decreased. Morphological studies conducted after the exposure of K562 to grandisin revealed changes consistent with the apoptosis process, which was...

In vitro skin penetration of clobetasol from lipid nanoparticles: drug extraction and quantitation in different skin layers BJPS
Silva,Luís Antônio Dantas; Taveira,Stephânia Fleury; Lima,Eliana Martins; Marreto,Ricardo Neves.
Clobetasol propionate (CP) is a potent topical corticosteroid that causes several cutaneous and systemic side effects. In the present work, CP was encapsulated in nanostructured lipid carriers (NLCs) to increase drug retention in the outer skin layers and improve the safety of topical therapy. NLCs were prepared using a microemulsion technique with a mixture of lecithin, taurodeoxycholate, stearic acid, and oleic acid. In vitro penetration studies were performed in a modified Franz-type diffusion cell, and porcine ears were used as a model of human skin. A simple and sensitive liquid chromatographic method was developed and validated for clobetasol determination in different skin layers. NLCs presented uniform size distribution, high zeta potentialand...

Development and physical evaluation of Maytenus ilicifolia effervescent granules using factorial design BJPS
Cunha-Filho,Marcilio Sérgio Soares da; Gustmann,Pricila Castilho; Garcia,Felipe Sanches; Lima,Eliana Martins; Sá-Barreto,Lívia Cristina Lira de.
The medicinal plant Maytenus ilicifolia is a commonly used phytomedicine for the treatment of gastritis. The high dose required and low density of these extracts make necessary a daily intake of several capsules, hindering adherence to the medication. The purpose of this work was to develop a suitable dosage form for the administration of Maytenus ilicifolia using effervescent granules. A 23 factorial design was used to study the physical characteristics of the granules (particle size distribution, repose angle, Carr index, scanning electron microscopy and disintegration time). Moisture stability was also determined. According to the experimental design, granule size is the most important factor in determining the flow characteristics of effervescent...
