Somatic, Biochemical and Hepatic Alterations in Wild Type Mice Chronically Fed High Fat Diet International Journal of Morphology
Neves,Renata Heisler; Barros Alencar,Alba Cristina Miranda de; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto; Machado-Silva,José Roberto; Gomes,Delir Corrêa.
This study evaluated whether a high fat diet (HFC group) induces overweight, hepatic steatosis and plasma lipoproteins level alteration compared to standard chow diet (SC group). Female mice were submitted to each diet over 6 months. Body mass and food intake were evaluated weekly throughout the experiment. Total cholesterol, TG, LDL-c, HDL-c and VLDL-c were analyzed. Mice were sacrificed to remove liver, spleen, heart and intestine. The volume of the organs was determined according to the submersion method. Fixed livers were embedded in paraffin and stained with hematoxylin and eosin and Masson's trichrome. The analysis used a video microscope system and a test-system with 42 test-points. The volume density was estimated for hepatocytes, steatosis and...

Beneficial Effects of Angiotensin II AT1 Blocker on Cardiovascular Adverse Remodeling Due to Nitric Oxide Synthesis Blockade International Journal of Morphology
Fernandes-Santos,Caroline; Mendonça,Leonardo de Souza; Mandarim-de-Lacerda,Carlos Alberto.
We studied with morphological tools the effects of different doses of Losartan upon the cardiovascular remodeling in nitric oxide deficient rats. At 15 weeks of age, thirty Wistar rats were separated in six groups: control (C), L-NAME (LN), and four groups were LN was given plus Losartan at different doses (1, 5, 20 and 40 mg/kg/day). The L-NAME was given for 9 weeks, the Losartan administration starting on the 2nd week of experiment. We studied the heart, thoracic aorta and superior mesenteric artery with light microscopy and stereology. The blood pressure (BP) increased since the first week of L-NAME administration, the Losartan treatment at doses of 20 and 40 mg/kg/day was efficient to reduce BP after the 7th week of treatment. The cardiac adverse...

Quantitative Morphology Update: Image Analysis International Journal of Morphology
Fernandes-Santos,Caroline; Souza-Mello,Vanessa; Faria,Tatiane da Silva; Mandarim-de-Lacerda,Carlos Alberto.
Quantitative morphology is a reliable tool in developmental, clinical and aging biology. Quantitative data of histological improvement and/or impairment due to dietary manipulations or pharmacological interventions draws the attention of countless researchers worldwide. Morphometry allows a wide range of two-dimension analysis to be performed, whereas image segmentation allows the measurement of a single structure or the determination of the intensity of its color after adequate immunostaining or even the area occupied by a specific sort of cell. When it comes to unbiased analysis, consideration should be given to suitable tissue perfusion, fixation and embedding, avoiding artifacts. Furthermore, tissue shrinkage should be taken into account. In order to...

Lean vs. Obese Mice: The Ventral Prostate Revisited International Journal of Morphology
Martins,Fabiane Ferreira; del Sol,Mariano; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto.
Obese mice (C57BL/6J-ob/ob) do not express leptin and develops hyperphagia, decreased energy expenditure, obesity, hyperglycemia, hyperinsulinemia, hypothermia, and infertility. Obesity causes reproductive dysfunction with negative impacts on prostatic structure and fertility. We aimed to compare the structure and molecular aspects of the ventral prostate between of lean and obese (ob/ob) mice. Three months old male lean and obese mice had their prostates dissected and prepared for light microscopy and immunofluorescence. In comparison to the lean mouse, the obese mouse showed a substantial structural modification in the ventral prostate starting with an atrophy of the prostate ventral lobe. Histologically, the acini showed a reduction in size, and in the...

ENALAPRIL AND VERAPAMIL ATTENUATE THE AORTIC WALL REMODELING IN NITRIC OXIDE DEFICIENCY International Journal of Morphology
Meirelles Pereira,Leila Maria; Gonçalves Bezerra,Daniele; Mandarim-de-Lacerda,Carlos Alberto.
Forty mature male rats (Wistar strain) were maintained alive for 40 days, separated in four groups of five rats each (Control, L-NAME, L-NAME + Enalapril, L-NAME + Verapamil). Three aortic wall parameters were analyzed, as follows: the Q A(SMN) (the number of the tunica media smooth muscle nuclear profiles per area), the S V(lamellae) (surface density of the aortic lamellae), and the aortic wall thickness (AWT). Rats of the L-NAME group showed high blood pressure and cardiac hypertrophy. The AWT, the Q A(SMN) and the S V(lamellae) showed the same tendency among the groups. The increased AWT, or the reduced Q A(SMN) and the S V(lamellae) in l-NAME rats were not completely prevented by the administration of enalapril or verapamil. Hypertension causes the AWT...

The Microfibril-Elastin Fiber System Distribution in Left Atrioventricular Valve of the Rat International Journal of Morphology
Rocha,Vinícius Novaes; Neto-Ferreira,Rodrigo; Mandarim-de-Lacerda,Carlos Alberto; Carvalho,Jorge José de.
The microfibril-elastin fiber system, an important constituent of the extracellular matrix, was studied in the rat left atrioventricular valve to investigate the interrelationship of oxytalan, elaunin and elastic fibers in left atrioventricular valve morphology. The elastin fibers forms continuous bundles observed along the length of the valve in atrial and ventricular layers and oriented parallel to endothelium. The elaunin and oxytalan fibers are distributed in the thickest fiber bundles along the length of the valve. The thinner fibers which radiated towards both the atrial and spongiosa layers, either as isolated or arborescent fiber bundles were identified as oxytalan fibers. With transmission electron microscopy elastic fibers were seen mainly in the...

Both Hepatic Lipogenesis and Beta-Oxidation are Altered in Offspring of Mothers Fed a High-Fat Diet in the First Two Generations (F1 and F2) International Journal of Morphology
Lannes,Wilian Rodrigues; Miranda,Andressa Cabral de; Souza-Mello,Vanessa de; Barbosa-da-Silva,Sandra; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto.
The high fat (HF) fed mothers may program susceptibility in offspring to chronic diseases and affect subsequent generations. The present study evaluated the liver structure in adulthood, focusing on the F1 and F2 generations. Females C57BL/6 (F0) were fed standard chow (SC) or HF diet (8 weeks) prior to mating and during the gestation and lactation to provide the F1 generation (SC-F1 and HF-F1). All other mothers and offspring fed SC. At 3 months old, F1 females were mated to produce the F2 generation (SC-F2 and HF-F2). The liver was kept in several fragments and prepared for histological analysis or frozen for biochemical and molecular analyzes. The F1 and F2 offspring were studied at 3 months old. HF-F1 had higher body mass (BM) compared to SC-F1 (P=...

Origin and Development of the Coronary Arteries International Journal of Morphology
Silva-Junior,Geraldo de Oliveira; Miranda,Sandro Wilson da Silva; Mandarim-de-Lacerda,Carlos Alberto.
The formation of the coronary vasculature is a fundamental event in heart development and involves a series of carefully regulated temporal events that include vasculogenesis and angiogenesis. This review focuses the knowledge concerning the formation of the coronary arteries available so far and some molecular mechanisms involved in this process. Understanding coronary embryogenesis is important for interventions regarding adult cardiovascular diseases as well as those necessary to correct heart congenital defects. The insight of the coronary artery development as a result of ingrowth changed the understanding of several congenital coronary artery variations and anomalies described in gross anatomy.
