O protocolo...

Rescue of a non-viable accession and rapd analysis of recovered plants of Arachis retusa PAB
Gagliardi,Rachel Fatima; Pacheco,Georgia Pereira; Oliveira,Carlos Alberto; Carneiro,Leonardo Alves; Valls,José Francisco Montenegro; Vieira,Maria Lucia Carneiro; Mansur,Elisabeth.
In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

Influence of substrates and in vitro preconditioning treatments on ex vitro acclimatization of Arachis retusa PAB
Pacheco,Georgia; Gagliardi,Rachel Fátima; Cogliatti,Mariana Buturini; Manhães,Harlen Barreira; Carneiro,Leonardo Alves; Valls,José Francisco Montenegro; Mansur,Elisabeth.
The objective of this work was to evaluate the influence of substrate and preconditioning treatments on the acclimatization of in vitro plants of Arachis retusa. Plants were transferred to Plantmax or sand, and fertilized with Hoagland's nutrient solution. Plants maintained in sand, with or without fertilizer, showed the highest survival rates. In order to evaluate the influence of in vitro preconditioning treatments, stem segments were cultured on MS medium supplemented with different sucrose concentrations. The highest survival and developmental rates were observed in plants from two accessions cultured on MS supplemented with 1.5% and 3% sucrose. Flowering and fruit production were observed after five months.

In vitro somatic embryogenesis and adventitious root initiation have a common origin in eggplant (Solanum melongena L.) Rev. Bras. Bot.
Tarré,Erika; Magioli,Claudia; Margis-Pinheiro,Márcia; Sachetto-Martins,Gilberto; Mansur,Elisabeth; Santiago-Fernandes,Lygia D. R..
Somatic embryogenesis was induced from cotyledon explants of eggplant cultured on MS medium supplemented with 54 µM NAA. Anatomical analysis of somatic embryo initiation and development was performed during the first four weeks. Proembryo formation was observed after the second day of culture, directly from perivascular cells or via pro-embryogenic masses derived from indeterminate meristematic masses (IMMs) originated in the vascular tissue. Those IMMs also gave rise to root primordia after 10 days of culture. The origin of embryos is discussed as well as the similarities between somatic embryogenesis and adventitious root formation.

Germinability after desiccation, storage and cryopreservation of seeds from endemic Encholirium Mart. ex Schult. & Schult. f. and Dyckia Schult. & Schult. f. species (Bromeliaceae) Acta Botanica
Tarré,Erika; Pires,Bárbara Balzana Mendes; Guimarães,Ana Paula Mançano; Carneiro,Leonardo Alves; Forzza,Rafaela Campostrini; Mansur,Elisabeth.
(Germinability after desiccation, storage and cryopreservation of seeds from endemic Encholirium Mart. ex Schult. & Schult. f. and Dyckia Schult. & Schult. f. species (Bromeliaceae)). Seed storage procedures require previous determination of optimal temperature and light conditions for germination, as well as of tolerance to desiccation and low temperatures. The aim of this paper was to study the effects of desiccation, storage at low temperatures and cryopreservation on the germinability of seeds of six Encholirium and two Dyckia species, which were selected according to vulnerability criteria. Initial germinability of newly harvested seeds varied from 35 to 95%. Seeds presented photoblastic behaviour since light was necessary to induce or...

Eggplant (Solanum melongena L.): tissue culture, genetic transformation and use as an alternative model plant Acta Botanica
Magioli,Claudia; Mansur,Elisabeth.
Eggplant is an agronomically important non-tuberous solanaceous crop grown primarily for its large oval fruit. In popular medicine, eggplant is indicated for the treatment of several diseases, including diabetes, arthritis, asthma and bronchitis. Eggplant is susceptible to a number of diseases and pests capable of causing serious crop losses. This problem has been addressed by hybridizing eggplant with wild resistant Solanum species, which present a wide genetic diversity and are source of useful agronomic traits. The application of in vitro methodologies to eggplant has resulted in considerable success. Eggplant tissues present a high morphogenetic potential that is useful for developmental studies as well as for establishing biotechnological approaches...

Somatic embryogenesis and plant regeneration from callus cultures of Cleome rosea Vahl BABT
Simões,Claudia; Albarello,Norma; Callado,Cátia Henriques; Castro,Tatiana Carvalho de; Mansur,Elisabeth.
This paper describes a protocol for the efficient vegetative propagation of Cleome rosea by somatic embryogenesis. Leaf and stem explants from nursery-grown seedlings of C. rosea were cultivated on Murashige and Skoog (MS) medium supplemented with indole-3-acetic acid (IAA), a -naphthaleneacetic acid (NAA), 4-amino-3,5,6-trichloropicolinic acid (picloram) or 2,4-dichlorophenoxyacetic acid (2,4-D). Nodular calli were produced from both explant types in the presence of 4.5 and 9.0 µM 2,4-D. Embryo development and maturation were achieved when calli from stem explants were transferred to media containing a ten-fold reduction of 2,4-D concentration initially used (0.45 and 0.90 µM). Leaf-derived calli did not form embryos with the same treatments. The highest...
