Sanitary quality of seeds of “pau-cigarra” (Senna multijuga) PFB - Pesquisa Florestal Brasileira

The aim of this research was to identify fungi associated with “pau-cigarra” seeds (Senna multijuga). Evaluation was carried out in nine lots composed of seeds collected from mother trees at Atlantic Rain Forest located in Antonina, Morretes and Guaraqueçaba municipalities, Paraná state, Brazil. Sanitary analysis was carried out by adopting blotter test and agar plate test. Morphological characteristics were used for fungi identification. It was observed higher fungi frequency contaminating seeds over filter paper (blotter test) compared with agar plate test. Helminthosporium sp., Alternaria sp., Ulocladium sp., Pestalotia sp., Penicillium sp., Trichoderma sp., Chaetomium sp. and Monilia sp. were the most important genus found associated with S. multijuga...

Fungos Associados aos Frutos e Sementes de Capororoca (Myrsine ferruginea) Myrsinaceae PFB - Pesquisa Florestal Brasileira

Os objetivos neste trabalho foram identificar os fungos associados aos frutos e sementes de capororoca (Myrsine ferruginea Spreng) obtidos de diferentes lotes e verificar a incidência de fungos entre frutos verdes e maduros e nas sementes obtidas destes frutos. Para a detecção de fungos utilizou-se o método de papel-filtro (PF), sendo 400 frutos e 400 sementes para cada amostra, incubadas sob 12 horas luz negra/12 horas escuro, na temperatura de 20 ºC ± 1 ºC durante sete dias, avaliando-se a incidência (%) de fungos. Pôde-se verificar a possível transmissão dos fungos Alternaria sp., Cladosporium sp., Colletotrichum sp., Fusarium sp., Macrophomina sp. e Pestalotia sp. dos frutos para as sementes. Sensitivity of brazilian-pine (Araucária angustifolia) embryos to desiccation and temperature PFB - Pesquisa Florestal Brasileira

The “recalcitrant” behavior of Araucaria angustifolia (Bert.) O. Kuntze seeds (Brazilian pine) limits the conservation of genetic resources of this important species in ex situ genebanks. As initial steps in developing storage protocols, we studied the interaction of water content and temperature on embryo survival. Embryos were excised from seeds and “flash-dried” to water contents as low as 0.29 g water/g dry mass and then exposed to 0°C, -5°C and -18°C for 4 h. Drying rate was calculated from drying time courses and expressed as (g water / g dry weight) / hour. Fungi Association with Cedar’s Seeds and Fusarium sp. and Pestalotia sp. Pathogenicities PFB - Pesquisa Florestal Brasileira

This research had as main objectives the evaluation of cedar’s seeds seed-borne fungi and the Fusariumsp. and Pestalotia sp. pathogenicities. For the fungi detection Potato Dextrose and Agar medium and Blottertest methods were used. For the pathogenicity tests, isolates of Fusarium sp. and Pestalotia sp. were used. The following fungi were observed: Pestalotia sp., Fusarium sp., Phomopsis sp., Colletotrichum sp., Macrophomina sp. and Cladosporium sp. The fungi observed in the highest frequencies were: Fusarium sp., Phomopsis sp. and Pestalotia sp. Flash-drying for embryos desiccation PFB - Pesquisa Florestal Brasileira

Seeds of several tree species from Brazilian wet forest are in dangerous situation and show recalcitrant characteristics. Therefore, it is not possible to store them by traditional methods adopted with orthodox seeds and it limits their genetic resources ex-situ conservation in seed banks. This situation encourages researches to find news technologies in order to store these species in genebanks. Therein, flash-drying method for embryos desiccation shows to be a promising technique in order to resolve seed conservation of Brazilian endogenous tree species in genebanks. This paper has an objective of to become known the flash-drying technique and to be accessible to seed  physiologists
