Gel mobility shift scanning of pectin-inducible promoter from Penicillium griseoroseum reveals the involvement of a CCAAT element in the expression of a polygalacturonase gene Genet. Mol. Biol.
Ribon,Andréa de O.B.; Ribeiro,João Batista; Gonçalves,Daniel B.; Queiroz,Marisa V. de; Araújo,Elza F. de.
Previous reports have described pgg2, a polygalacturonase-encoding gene of Penicillium griseoroseum, as an attractive model for transcriptional regulation studies, due to its high expression throughout several in vitro growth conditions, even in the presence of non-inducing sugars such as sucrose. A search for regulatory motifs in the 5' upstream regulatory sequence of pgg2 identified a putative CCAAT box that could justify this expression profile. This element, located 270 bp upstream of the translational start codon, was tested as binding target for regulatory proteins. Analysis of a 170 bp promoter fragment by electrophoretic mobility shift assay (EMSA) with nuclear extracts prepared from mycelia grown in pectin-containing culture medium revealed a high...

Development of a transformation system for Penicillium brevicompactum based on the Fusarium oxysporum nitrate reductase gene BJM
Varavallo,Maurílio Antônio; Queiroz,Marisa Vieira de; Pereira,Jorge Fernando; Ribeiro,Ronney Adriano; Soares,Marcos Antônio; Ribeiro,João Batista; Araújo,Elza Fernandes de.
Penicillium brevicompactum is a filamentous fungus that presents a potential for industrial use due its efficient pectinase production. A heterologous transformation system was developed for P. brevicompactum based on the complementation of a nitrate reductase mutant. Nitrate reductase mutants were obtained by resistance to chlorate in a rate of 23.24% when compared to other mutations that lead to the chlorate resistance. One mutant named 4457-18X was chosen for the transformation experiments with the pNH24 vector containing de Fusarium oxysporum nitrate reductase gene. A frequency of approximately 3 transformants/µg DNA was obtained using the circular vector pNH24. This frequency was multiplied about 10 fold using the linearized vector with the Xba I...

Seroprevalence for equine infectious anaemia in Equidae seized in the municipality of Petrópolis, State of Rio de Janeiro, Brazil, 2015/2018 Ciência Rural
Cruz,Astrid Paola Mattheis; Aquino,Maria Helena Consedey de; Helayael,Michel José Sales Abdalla; Silva,Márcio Roberto; Ribeiro,João Batista; Hott,Marcos Cicarini; Magalhães Júnior,Walter Coelho Pereira de; Mendonça,Juliana França Monteiro de; Souza,Guilherme Nunes de.
ABSTRACT: Equine infectious anaemia (EIA) is a globally distributed disease caused by a lentivirus. EIA is a notifiable disease that should be reported to the Official Veterinary Service (SVO). Outbreaks are controlled by sacrificing the EIA-positive animals. The study analysed the prevalence of EIA in 165 Equidae seized on public roads in the municipality of Petrópolis, Rio de Janeiro, Brazil, from 2015 to 2018. After the seizure and identification of the Equidae, blood was collected to diagnose EIA. A prevalence of 11.8% was observed among the Equidae seized, and the main risk factor was the region of seizure. A kernel-smoothed map showed areas with the highest density of EIA-positive Equidae in the centre and to the east of the study area. In this...
