One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer

Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. Bonamia ostreae-induced mortalities in one-year old European flat oysters Ostrea edulis: experimental infection by cohabitation challenge ArchiMer

Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Après le naufrage de l'Erika, la question s'est rapidement posée de l'impact des hydrocarbures sur la santé des coquillages, car la pollution concernait des zones de production conchylicoles. Le suivi zoosanitaire en place dans ces zones permettait d'avoir des données de référence avant impact. Il a été choisi différents sites exposés de référence, qui ont fait l'objet d'analyse en toxicologie et en pathologie. Nous disposons aujourd'hui d'o utils diagnostiques sensibles et spécifiques (PCR, HIS ... ) et d'un protocole de transmission de l' infection qui permettent d'assurer un suivi des productions larvaires en écloserie de bivalves pour les espèces considérées comme sensibles aux infections à herpèsvirus, mais également pour de nouvelles espèces. En effet, des mortalités massives (100 %) ont été rapportées dans une écloserie de coquille St-Jacques en Bretagne et ont pu être associées à la détection de OsHV -1. Les résultats obtenus concernant cette espèce, Pecten maximus, complètent et reflètent l'ensemble des travaux réalisés.

Molecular detection and quantification of the protozoan Bonamia ostreae in the flat oyster, Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. A 4-year bacteriological survey (2003-2007) of four molluscs cultivated in France and faced with mortality episodes was performed by the French shellfish pathology network. The more abundant bacteria isolated during 92 mortality episodes, occurring mainly in Pacific oyster Crassostrea gigas, were identified by genotyping methods. Marteilia refringens is a protozoan parasite recognized as a significant pathogen of the European flat oyster Ostrea edulis. The life cycle of this species is still poorly known, although there is evidence of the need for intermediate host(s). In the present study, we have used molecular approaches to identify this parasite in samples of the dwarf oyster Ostrea stentina after reports of massive mortality along the Tunisian coasts. La cellule de veille zoosanitaire de La Tremblade s'occupe de trois zones situées au sud de la Loire: la Baie de Bourgneuf, la région de Marennes-Oléron-Charentes, et le bassin d'Arcachon. Ces régions produisent principalement des huîtres creuses. Le naissain d'huîtres creuses produit à Arcachon et en Charente représente près de 80% de la production d'huître creuse, qui sera élevée dans toutes les régions ostréicoles françaises. Un effort particulier avec des prélèvements systématiques a donc été mis en place sur Crassostrea gigas dans ces trois trois zones, notamment sur les larves et le naissain d'Arcachon et de Charente. Molecular characterisation of an Australian isolate of Bonamia exitiosa ArchiMer

An Australian (New South Wales) isolate of Bonamia was characterised at the molecular level by sequencing the 18S-ITS-1 region of the small subunit rRNA operon obtained from flat oysters Ostrea angasi shown to be infected by histological examination. Sequence data alignment with homologous genes from 2 other isolates of Bonamia (New Zealand and France) revealed high levels of nucleotide identity with both isolates. However, the Australian Bonamia is shown to be more closely related to the New Zealand isolate, suggesting the existence of an oceanic subgroup of Bonamia.

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer

The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. The paramyxean parasite Marteilia refringens infects several bivalve species including European flat oysters Ostrea edulis and Mediterranean mussels Mytilus galloprovincialis. Sequence polymorphism allowed definition of three parasite types ‘M’, ‘O’ and ‘C’ preferably detected in oysters, mussels and cockles respectively.Transmission of the infection from infected bivalves to copepods Paracartia grani could be experimentally achieved but assays fromcopepods to bivalves failed.
