Pathogenic threats and probiotic use in larviculture of the scallop, Pecten maximus ArchiMer

During a series of experiments, bacteriological elements in scallop larval rearing were investigated: larvae susceptibility to pathogens as a function of their age, and the use of probiotic bacteria during larviculture. Younger larvae (d5 PF) were highly more susceptible to pathogenic-challenge than their older siblings, which were challenged at an older age (d15 PF). A challenge with 104 CFU mL−1 of V. pectenicida killed 100% of d5 PF larvae 7 days following challenge, yet killed only 9% of d15 PF larvae 9 days following challenge. In contrast with the Japanese oyster Crassostrea gigas and the Manila clam Ruditapes philippinarum, Ostrea edulis seed production in the hatchery has been reported to be erratic, with sudden and unexplained larval and post-metamorphosis mortalities. Fecundity and initial larval quality have been related to broodstock conditioning, but effects on larval development and metamorphosis remain poorly understood. Larval mortalities occurring in molluscan hatcheries have often been associated with bacterial contamination. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions. For this species, an addition of chloramphenicol was found necessary. However, this chemical has now been banned in Europe. Thus either substitition products or an improvement in the rearing procedures is essential. Experimental shift of diet and DIC stable carbon isotopes: Influence on shell delta C-13 values in the Manila clam Ruditapes philippinarum ArchiMer

The influences of diet and seawater dissolved inorganic carbon (DIC) on the carbon isotope composition of shell aragonite (Cinched in the Manila clam Ruditapes philippinarum reared under laboratory conditions were investigated. Clams were exposed to two successive negative carbon isotope shifts: a first shift in diet (delta C-13(phytoplankton)) and a second shift, 35 days later, in DIC (delta C-13(DIC)). Both successive shifts induced a decrease in delta C-13(shell). These results are the first to experimentally confirm an incorporation of respired carbon derived from food and carbon from DIC into shell carbonate of adult bivalves. Influence of feeding regime and temperature on development and settlement of oyster Ostrea edulis (Linnaeus, 1758) larvae ArchiMer

Under controlled conditions of food density and temperature, larval performances (ingestion, growth, survival and settlement success) of the flat oyster, Ostrea edulis, were investigated using a flow-through rearing system. In the first experiment, oyster larvae were reared at five different phytoplankton densities (70, 500, 1500, 2500 and 3500 μm3 μL−1: ≈1, 8, 25, 42 and 58 cells μL−1 equivalent TCg), and in the second, larvae were grown at four different temperatures (15, 20, 25 and 30°C). Overall, larvae survived a wide range of food density and temperature, with high survival recorded at the end of the experiments. Ontogenic changes in the contents of dopamine, norepinephrine and serotonin in larvae and postlarvae of the bivalve Pecten maximus ArchiMer

In the present study, norepinephrine (NE), epinephrine (E), dopamine (DA) and 5-hydroxy-tryptamine (5HT) have been quantified by high performance liquid chromatography (HPLC) in Pecten maximus during larval and post-larval development. With average values ranging from 2 to 5 pg mug(-1) of protein, NE remained low through the whole larval life, while epinephrine (E) was undetected. DA and 5HT were the most abundant monoamines with significant variations between larval, pre-metamorphic and post-larval stages. Analysis of larval oyster grazing by flow cytometry ArchiMer

The ingestion of 8 algal species by oyster larvae (Crassostrea gigas Thunberg, 1793) was measured by flow cytometry (FCM). In a preliminary experiment, cell size (estimated by light scatter) and chlorophyll fluorescence of 30 algal species were evaluated to select those species which could be mixed together and still be easily discriminated by FCM. Grazing experiments were carried out over 48 h with 6 and 15-day old larvae fed on 3 algal mixtures, each containing 3 different algal species. The concentration of each species was estimated at 0, 6, 12, 24 and 48 h by FCM. From 1988 to 1989, a study has been conducted pertaining to the growth of the flat oyster Ostrea edulis, as well as Marteilia refringens and Bonamia ostreae outbreaks. Commercial mono specific farming (flat oysters), with low density or mixed (flat and hollow oysters) along with higher density, has been taking place in the Arcachon basin oceanographic sectors, where main hydrobiological parameters were acquired simultaneously. The excellent growth of the flat oyster was witnessed in mono specific farming. However this growth was average in mixed farming, probably due to higher total oyster density and interspecies competition. Testing of the Argentina oyster, Ostrea puelchana, in several French oyster farming sites ArchiMer

Because of diseases affecting fIat oyster Ostrea edulis, acclimatization of the Argentina oyster Ostrea puelchana has been tested in 1989-90 in five French oyster farming sites. Following ICES recommendations , hatchery produced juveniles, from parents imported from Argentina, were transferred to experimental si tes in summer 1989. Very high mortalities occurred, reaching 46 to 98% in March-Apri1 1990. They continued later on, resulting in the end of experiments in March 1991. Manila clam (Tapes philippinarum) culture and sediment interactions were tested by comparing two rearing areas, including an oceanic ('Le-Ferret') and a more estuarine ('Les-Jacquets') sites in the bay of Arcachon (France). The growth of a calibrated clam population (10-mm spat) was monitored in these two areas with a concomitant sediment-water interface survey over a 1.5-year period. For lack of antibiotics, King scallop larviculture faces high mortalities. In the experimental mollusc hatchery of Argenton (Brittany, France), the presence of a vibrio, Vibrio pectinicida, associated with the stocks seems to be responsible for this low survival rate. As prophylactic methods are difficult to implement in the current structures (water sterilization, egg decontamination, isolation of the stocks), the addition of chloramphenicol is the only solution to protect the stocks. However, the use of this antibiotic, considered as dangerous for man, has been prohibited in animal production throughout the whole of Europe since 1994. Comparaison de deux méthodes de prélèvement pour le dénombrement des larves de Crassostrea gigas en zone conchylicole ArchiMer

La numération des larves d'huître dans le plancton renseigne les ostréiculteurs sur la date de pose des collecteurs. La technique utilisée est très ancienne (BOURY, 1928) ; pour chaque station trois prélèvements par traits de filets pendant 10 minutes sont effectués : deux en surface (filet 200 et filet 130) et un à 1 mètre de profondeur (filet 130). Une méthode par pompage avec mesure instantanée du volume d'eau filtrée a été mise au point (prélèvements en surface et à 1 mètre de profondeur, et rétention des véligères sur filets 200). La comparaison des deux techniques met en évidence l'inadaptation de l'ancienne méthode. A flow-through rearing system for ecophysiological studies of Pacific oyster Crassostrea gigas larvae ArchiMer

While literature is relatively abundant on adult shellfish, ecophysiological studies at the larval stage are scarce because of both technical difficulties and inadequate methodology. A tool dedicated to provide basic information for larval ecophysiology was accordingly developed. Action de l'acétate de tributyle-étain sur les oeufs et les larves D de deux mollusques d'intérêt commercial : Crassostrea gigas (Thunberg) et Mytilus galloprovincialis (Lmk) ArchiMer

The Tributyl tin acetate, a component of certain antifouling paints, has deleterious effects on eggs and D shaped larvae from Crassostrea gigas at 5 p.p.b. (HIS & ROBERT, 1980). Here we have tested concentrations as low as 1 p.p.b. on eggs and on 24 hours old larvae both from Crassostrea gigas and Mytilus galloprovincialis. The same results are got ; this product kills embryos and larvae in a few days ; it prevents growth of 24 hours old larvae exposed to this toxic. Mussels larvae seem to be less sensitive.
