Molecular key to seven Brazilian species of Trichogramma (Hymenoptera: Trichogrammatidae) using sequences of the ITS2 region and restriction analysis Neotropical Entomology

Using the ITS2 sequences of the ribosomal DNA together with some restriction enzymes, a simple and precise molecular key to some Brazilian species of Trichogramma was created. This key is very easy to make and quick results can be obtained on the identification of this minute parasitoid (0.25 mm). The methodology presented is easily implemented and can be used to detect possible contaminations under laboratory rearings.

Effect of Wolbachia on the Survival and Reproduction of Trichogramma kaykai Pinto & Stouthamer (Hymenoptera: Trichogrammatidae) Neotropical Entomology

Parthenogenetic inducing Wolbachia infections are particularly common in the genus Trichogramma, but the influence of the microrganism on parasitoids' fitness has received relatively little attention. The aim of this study was to determine the effects of the bacteria on the survival and reproduction of Trichogramma kaykai Pinto & Stouthamer, comparing an infected line (thelytokous) with an antibiotically cured (arrhenotokous) line, both reared on Trichoplusia ni Hübner eggs. Cured wasps produced significantly more progeny and more daughters than Wolbachia-infected wasps. Detecção de Wolbachia em uma População Telítoca de Trichogramma atopovirilia Oatman & Platner (Hymenoptera: Trichogrammatidae) via PCR com o Primer Específico wsp Neotropical Entomology

Identificou-se pela primeira vez no Brasil a presença de Wolbachia em Trichogramma através de um PCR com o primer específico wsp. A população de Trichogramma atopovirilia Oatman & Platner foi coletada na Embrapa Milho e Sorgo, em ovos de Helicoverpa zea Boddie (Lepidoptera: Noctuidae). Os resultados mostraram através da amplificação de bandas de DNA, a confirmação da presença de Wolbachia na população em estudo. A presença dessa a-proteobactéria irá contribuir para a escolha da população correta a ser utilizada em programas de controle biológico aplicado contra lepidópteros pragas.

Host Deprivation Effect on Reproduction and Survival of Wolbachia-Infected and Uninfected Trichogramma kaykai Pinto & Stouthamer (Hymenoptera: Trichogrammatidae) Neotropical Entomology

The effect of temporary host deprivation on the reproduction and survival of Trichogramma kaykai Pinto & Stouthamer was determined by comparing a Wolbachia-infected (thelytokous) and an arrhenotokous line of T. kaykai when reared on Trichoplusia ni Hübner (Lepidoptera: Noctuidae) eggs. The progeny produced by both arrhenotokous and thelytokous females decreased with increasing periods of host deprivation, but arrhenotokous females produced significantly more progeny than thelytokous females regardless of the period of host deprivation (0 to 10 days). They also produced more daughters. Host deprivation did not affect brood size within arrhenotokous or thelytokous lines. Molecular Tool for Identification of Closely Related Species of Trichogramma (Hymenoptera: Trichogrammatidae): T. rojasi Nagaraja & Nagarkatti and T. lasallei Pinto Neotropical Entomology

The sequence of the ITS2 (internally transcribed spacer 2) region of the rDNA was used to identify two closely related Trichogramma species: T. rojasi and T. lasallei, the second species recently reported in Brazil. The DNA of the samples was extracted using Chelex 100. The PCR product was linked to a pGEM-T® vector and both were then placed into Petri dishes to grow white colonies, which demonstrate the correct ligation between the vector and the DNA. From these colonies, 10µl were purified and sequenced using a special kit, and the sequences aligned using the ESEE 3.0 software.
