Assessment of the efficiency of SimPlate™ total plate count color indicator (TPC CI) to quantify mesophilic aerobic microorganisms in pasteurized milk BJM

The SimPlate™ TPC CI system is a rapid method to count mesophilic aerobic microorganisms (MAM) in foods, based on the use of resazurine to indicate bacterial growth. Its efficiency in pasteurized milk was evaluated using 142 pasteurized milk samples (38 type A, 43 type B and 61 type C) collected in Londrina, PR. The standard plating method, using Plate Count Agar (PCA) was used for comparison. The plates of both systems were incubated at 35ºC and read after 24h and 48h. The occurrence of false-positive and false-negative wells and the predominant microorganisms in them were also evaluated. The results were compared by simple correlation and mean variance analyses. Use of Readycult™ - LMX for enumeration of total coliforms and Escherichia coli in milk BJM

Readycult™ - LMX is a rapid method to test for the presence of total coliform bacteria and Escherichia coli in water, giving results in 24h. It is based on reactions of specific microbial enzymes or indicator nutrients of the medium. The goal of this paper was to study the use of Readycult™ - LMX to detect total coliform and E. coli in milk. One hundred twenty five samples of pasteurized and raw milk, collected in Londrina, PR, Brazil, were tested simultaneously by the most probable number (MPN) method using Brilliant Green Bile Lactose Broth (2%) (BRILA) and Readycult™ - LMX. ABSTRACT: Raw milk samples were collected from cooling tanks (after they cooled for 48 h) in five dairy farms and the corresponding bulk tank (bulk milk transportation, BMT) when they arrived to the industry. Routine physical chemical analyzes and quantification of psychrotrophic ( Pseudomonas spp. and P. fluorescens ) and aerobic mesophilic (AM) populations were performed. Only relative density and titratable acidity values for samples of milk from three farms were in agreement to the quality parameters required by law. In the BMT, only the protein content has not reached the minimum value established by law, and counting was performed for AM (>105 colony forming units (CFU) mL-1) and psychrotrophic bacteria (2.8x106CFU mL-1).
