Mondeguer, Florence; Allard, Pierre-marie; Souard, Florence; Elie, Nicolas; Guitton, Yann; Sibat, Manoella; Siano, Raffaele.
During their life cycle, Alexandrium minutum and Scrippsiella donghaienis can produce resistant and revivable cysts that can accumulate in marine sediments for hundreds of years. Our working hypothesis assumes that these two genera have "recorded" their adaptation to ecosystem changes in these cells preserved in the form of cysts. From these revived cysts, toxic and non-toxic dinoflagellate cultures (Alexandrium minutum and Scrippsiella donghaienis) were analyzed by a metabolomic approach to investigate the possible adaptive responses of phytoplankton to these changes in coastal ecosystems. The strains were obtained from isotopically dated sediments, 1986 and 1996 for "old" and 2006 for "modern". In total, 84 samples were obtained. Each experimental...

Belin, Catherine; Chapelle, Annie; Delmas, Daniel; Nezan, Elisabeth; Siano, Raffaele.

Innovative metabolomic workflows to study growth kinetics dinoflagellate cysts revived from modern and ancient sediments ArchiMer
Mondeguer, Florence; Souard, Florence; Guitton, Yann; Allard, Pierre-marie; Elie, Nicolas; Sibat, Manoella; Latimier, Marie; Quere, Julien; Wolfender, Jean-luc; Siano, Raffaele; Hess, Philipp.
Aquatic ecosystems worldwide have been negatively affected by eutrophication, many of them driven by increasing nutrient inputs from untreated domestic sewage and industrial and agricultural wastewater. During their life cycle, Alexandrium minutum and Scrippsiella donghaienis can produce resistant and revivable cysts that can accumulate in the sediments of the coastal marine environment for hundreds of years. As a working hypothesis, we presumed that these two genera have preserved their adaptation to such ecosystem changes in the form of cysts in the old sediments. From revivified cysts, some cultures of toxic and non-toxic dinoflagellates (A. minutum and S. donghaienis) were analyzed by a metabolomic approach to study the possible adaptive responses of...

Human-induced river runoff overlapping natural climate variability over the last 150 years: Palynological evidence (Bay of Brest, NW France) ArchiMer
Lambert, Clement; Penaud, Aurelie; Vidal, Muriel; Klouch, Khadidja; Gregoire, Gwendoline; Ehrhold, Axel; Eynaud, Frederique; Schmidt, Sabine; Ragueneau, Olivier; Siano, Raffaele.
For the first time a very high resolution palynological study (mean resolution of 1 to 5 years) was carried out over the last 150 years in a French estuarine environment (Bay of Brest; NW France), allowing direct comparison between the evolution of landscapes, surface water, and human practices on Bay of Brest watersheds, through continental (especially pollen grains) and marine (phytoplanktonic microalgae: cysts of dinoflagellates or dinocysts) microfossils. Thanks to the small size of the watersheds and the close proximity of the depositional environment to the mainland, the Bay of Brest represents an ideal case study for palynological investigations. Palynological data were then compared to published palaeo-genetic analyses conducted on the same core...

Species specific gene expression dynamics during harmful algal blooms ArchiMer
Metegnier, Gabriel; Paulino, Sauvann; Ramond, Pierre; Siano, Raffaele; Sourisseau, Marc; Destombe, Christophe; Le Gac, Mickael.
Harmful algal blooms are caused by specific members of microbial communities. Understanding the dynamics of these events requires comparing the strategies developed by the problematic species to cope with environmental fluctuations to the ones developed by the other members of the community. During three consecutive years, the meta-transcriptome of micro-eukaryote communities was sequenced during blooms of the toxic dinoflagellate Alexandrium minutum. The dataset was analyzed to investigate species specific gene expression dynamics. Major shifts in gene expression were explained by the succession of different species within the community. Although expression patterns were strongly correlated with fluctuation of the abiotic environment, and more...

Diversity of cultured photosynthetic flagellates in the northeast Pacific and Arctic Oceans in summer ArchiMer
Balzano, S.; Gourvil, P.; Siano, Raffaele; Chanoine, M.; Marie, D.; Lessard, S.; Sarno, D.; Vaulot, Daniel.
During the MALINA cruise (summer 2009), an extensive effort was undertaken to isolate phytoplankton strains from the northeast (NE) Pacific Ocean, the Bering Strait, the Chukchi Sea, and the Beaufort Sea. In order to characterise the main photosynthetic microorganisms occurring in the Arctic during the summer season, strains were isolated by flow cytometry sorting (FCS) and single cell pipetting before or after phytoplankton enrichment of seawater samples. Strains were isolated both onboard and back in the laboratory and cultured at 4 °C under light/dark conditions. Overall, we isolated and characterised by light microscopy and 18 S rRNA gene sequencing 104 strains of photosynthetic flagellates which grouped into 21 genotypes (defined by 99.5% 18 S rRNA...

A new potentially toxic Azadinium species (Dinophyceae) from the Mediterranean Sea, A. dexteroporum sp. nov. ArchiMer
Percopo, Isabella; Siano, Raffaele; Rossi, Rachele; Soprano, Vittorio; Sarno, Diana; Zingone, Adriana.
A new photosynthetic planktonic marine dinoflagellate, Azadinium dexteroporum sp. nov., is described from the Gulf of Naples (South Tyrrhenian Sea, Mediterranean Sea). The plate formula of the species, Po, cp, X, 4, 3a, 6, 6C, 5?S, 6'" and 2, is typical for this recently described genus. Azadinium dexteroporum is the smallest rep-resentative of the genus (8.5m average length, 6.2m average width) and shares the presence of a small antapical spine with the type species A.spinosum and with A.polongum. However, it differs from all other Azadinium species for the markedly asymmetrical Po plate and the position of the ventral pore, which is located at the right posterior end of the Po plate. Another peculiarity of A.dexteroporum is the pronounced concavity of...

ArchiMer
Chapelle, Annie; Le Bec, Claude; Siano, Raffaele; Caradec, Florian; Andrieux, Francoise.
Variable inter and intraspecies alkaline phosphatase activity within single cells of revived dinoflagellates ArchiMer
Girault, Mathias; Siano, Raffaele; Labry, Claire; Latimier, Marie; Jauzein, Cecile; Beneyton, Thomas; Buisson, Lionel; Del Amo, Yolanda; Baret, Jean-christophe.
Adaptation of cell populations to environmental changes is mediated by phenotypic variability at the single-cell level. Enzyme activity is a key factor in cell phenotype and the expression of the alkaline phosphatase activity (APA) is a fundamental phytoplankton strategy for maintaining growth under phosphate-limited conditions. Our aim was to compare the APA among cells and species revived from sediments of the Bay of Brest (Brittany, France), corresponding to a pre-eutrophication period (1940’s) and a beginning of a post-eutrophication period (1990’s) during which phosphate concentrations have undergone substantial variations. Both toxic marine dinoflagellate Alexandrium minutum and the non-toxic dinoflagellate Scrippsiella acuminata were revived from...

Action Indice Composition. Livrable n° AIV. Rapport final, 16 juillet 2014. ArchiMer
Delmas, Daniel; Siano, Raffaele.
Studies on phytoplankton community diversity cannot be limited microphytoplankton (cells>20 ìm), it is indeed necessary to take into account also the nano (3-20ìm) picophytoplankton (<3 ìm) components. In the frame of this work aimed at studying temporal phytoplankton community diversity two complementary methodologies of phytoplankton sample analysis, pigment and genetic (metarbarcoding) diversity, have been used as alternative to routine morphological analysis carried out at light microscopy. The REPHY station (047-P-016 Concarneau Large) off-shore the Concarneau Bay has been sampled during the annual highest phytoplankton production (March-July 2012) with a frequent sample interval strategy (twice per week). Results obtained by both pigment and...

The multiannual dynamic of the cyst-forming and toxic marine dinoflagellate Alexandrium minutum was studied over a time scale of about 150 years by a paleoecological approach based on ancient DNA (aDNA) quantification and cyst revivification data obtained from two dated sediment cores of the Bay of Brest (Brittany, France). The first genetic traces of the species presence in the study area dated back to 1873 ± 6. Specific aDNA could be quantified by a newly-developed real-time PCR assay in the upper core layers, in which the germination of the species (in up to 17–19 year-old sediments) was also obtained. In both cores studied, our quantitative paleogenetic data showed a statistically significant increasing trend in the abundance of A. minutum ITS1 rDNA...

Marine plankton support global biological and geochemical processes. Surveys of their biodiversity have hitherto been geographically restricted and have not accounted for the full range of plankton size. We assessed eukaryotic diversity from 334 size-fractionated photic-zone plankton communities collected across tropical and temperate oceans during the circumglobal Tara Oceans expedition. We analyzed 18S ribosomal DNA sequences across the intermediate plankton-size spectrum from the smallest unicellular eukaryotes (protists, > 0.8 micrometers) to small animals of a few millimeters. Eukaryotic ribosomal diversity saturated at similar to 150,000 operational taxonomic units, about one-third of which could not be assigned to known eukaryotic groups....

Evolutionary processes and cellular functions underlying divergence in Alexandrium minutum ArchiMer
Le Gac, Mickael; Metegnier, Gabriel; Chomerat, Nicolas; Malestroit, Pascale; Quere, Julien; Bouchez, Olivier; Siano, Raffaele; Destombe, Christophe; Guillou, Laure; Chapelle, Annie.
Understanding divergence in the highly dispersive and seemingly homogeneous pelagic environment for organisms living as free drifters in the water column remains a challenge. Here, we analyzed the transcriptome wide mRNA sequences, as well as the morphology of 18 strains of Alexandrium minutum, a dinoflagellate responsible for Harmful Algal Blooms worldwide, to investigate the functional bases of a divergence event. Analysis of the joint site frequency spectrum (JSFS) pointed toward an ancestral divergence in complete isolations followed by a secondary contact resulting in gene flow between the two diverging groups, but heterogeneous across sites. The sites displaying fixed SNPs were associated with a highly restricted gene flow and a strong...

Bacteria enhance the production of extracellular polymeric substances by the green dinoflagellate Lepidodinium chlorophorum ArchiMer
Roux, Pauline; Siano, Raffaele; Collin, Karine; Bilien, Gwenael; Sinquin, Corinne; Marchand, Laetitia; Zykwinska, Agata; Delbarre-ladrat, Christine; Schapira, Mathilde.
High biomasses of the marine dinoflagellate Lepidodinium chlorophorum cause green seawater discolorations along Southern Brittany (NE Atlantic, France). The viscosity associated to these phenomena has been related to problems in oyster cultivation. The harmful effect of L. chlorophorum might originate from the secretion of Extracellular Polymeric Substances (EPS). To understand whether the EPS are produced by L. chlorophorum or its associated bacteria, or if they are a product of their interaction, batch cultures were performed under non-axenic and pseudo-axenic conditions for three strains. Maximum dinoflagellate cell abundances were observed in pseudo-axenic cultures. The non-sinking fraction of polymers (Soluble Extracellular Polymers, SEP), mainly...
