Lab-Scale production of Bacillus atrophaeus' spores by solid state fermentation in fifferent types of bioreactors BABT

Studies were conducted to evaluate Bacillus atrophaeus spores' production by solid-state fermentation (SSF) using sugarcane bagasse as support and soybean molasses as substrate at lab-scale in column bioreactors (forced aeration), plastic bags and Erlenmeyer flasks (aeration by diffusion). Different moisture contents (84%, 86% and 88%; 89%, 91% and 93%) and aeration rates (30mL/min, 45mL/min, 60mL/min and 90mL/min) were studied. The best condition for spore production (3.3x10(10) CFU.g-¹dry matter) in column bioreactor was 80% of initial humidity and no aeration. Development of a Low Cost Bioprocess for Endotoxin Production by Bacillus thuringiensis var israelensis Intended for Biological Control of Aedes aegypti BABT

Aedes aegypti is the vector of Dengue disease, responsible for 20,000 deaths/year worldwide. Bacillus thuringiensis var israelensis - Bti releases selective and effective toxins (crystal proteins) against A. aegypti larvae. We present a low cost bioprocess for toxin production, accomplished by a selected Brazilian strain Bti (BR-LPB01) and employment of low cost substrates. Soybean meal and sugarcane molasses lead to high toxic effectiveness after 2L bioreactor fermentation (LD50=26ng/mL), near to the reference strain IPS82 (LD50=17.3 ng/mL). L-Lysine is an essential aminoacid added as supplement for animal feed. The aim of this work was to produce an L-Lysine enriched bran using Brazilian agroindustrial byproducts. Both the raw material costs and purification steps were minimized. Firstly, medium composition for the growth of Corynebacterium glutamicum ATCC 21799 was optimized targeting enhanced L-Lysine production - salt, vitamins and nitrogen sources concentrations were tested and selected. Respirometry kinetics of phenol oxidation by Comamonas testosteroni Pb50 under various conditions of nutritional stress BABT

Kinetics of phenol biodegradation using suspended biomass of Comamonas testosteroni Pb50 (monoculture) was measured under conditions of nutrient abundance, limitation, and prolonged cell starvation in a fed-batch reactor, with phenol being the sole carbon and energy source. The pre-washed cells were applied for measurement of the phenol and oxygen uptake rates at varied starting phenol concentrations with the kinetic parameters calculated using the Haldane model. The results revealed that nutrient limitation significantly suppressed the maximum value of exogenous respiration rate while the endogenous respiration rate, affinity and tolerance to phenol increased. Formulated products containing a new phytase from Schyzophyllum sp. phytase for application in feed and food processing BABT

A new formulated product containing high yield of phytase from Schizophyllum sp., an important mushroom used for medicinal studies, was developed for application in feed industries and for future use in food processing. The enzyme presented a high activity yield 55.5 U/mL and 6240 U/gds in liquid and solid formulated product, respectively. It showed a good shelf-life in concentrated product, retaining 67.8% of its activity after 60 days of storage at room temperature and 90% of the activity was maintained in the liquid formulation after the same period. Isolation and screening of microorganisms with potential for biotransformation of terpenic substrates BABT

The objective of the present work was to isolate and select strains with potential to perform the biotransformation of terpenic substrates. Microorganisms obtained from a collection culture and also isolated from a natural source of terpene substrate were tested. Seventeen strains were selected by their resistance to terpenes in potato dextrose agar containing up to 1% of limonene or α-pinene and β-pinene (1:1). Subsequently, 10 strains were selected by their capacity of using these terpenes as sole carbon source in a mineral medium. The biotransformation capacity of these strains was tested and the products obtained were identified by GC-MS.

This work presents a statistical model of survival analysis for three pathogenic bacterial strains (Escherichia coli, Listeria monocytogenes and Staphylococcus aureus), when treated with neutralized and non-neutralized filtered supernatants broth from cultures of Lactobacillus acidhophilus, Lactobacillus rhamnosus and Lactobacillus sake. Survival analysis is a method employed to determine the period of time from an initial stage up to the occurrence of a particular event of interest, as death or a particular culture growth failure. Bovine mastitis in the metropolitan area of Curitiba: antibiotic resistance and antimicrobial control of the infection BABT

A study from cows with mastitis was performed and Staphylococcus aureus was the predominant pathogen in 46.4 % among 153 studied strains from 276 milk samples of infected cows. Antibiotic resistance of 71 S. aureus isolates was determined in order to search resistant strains to antibiotics of clinical interest, as well as to determine their degree of multi-resistance. It was found that 60% of the S. aureus strains presented resistance to β-lactams, but none to oxacillin, teicoplamin or vancomycin. On the other hand, with the aim of reducing the use of current antibiotics and their associated resistance, a new formulation was introduced. Biofiltration of a styrene/acetone vapor mixture in two reactor types under conditions of styrene overloading BABT

This aim of study was to compare the performance of a biofilter (BF) and trickle bed reactor (TBR) under increased styrene loading with a constant acetone load, 2 gc/m3/h. At styrene loading rates up to 30 gc/m3/h, the BF showed higher styrene removal than TBR. However, the BF efficiency started to drop beyond this threshold loading and could never reach steady state, whereas the TBR continued to yield a 50% styrene removal. The acetone removal remained constant (93-98%) in both the reactors at any styrene loading. Once the overloading was lifted, the BF recovered within 26 min, whereas the TBR efficiency bounced back only to 95%, gradually returning to complete removal only in 10 h.

Simple models for the continuous aerobic biodegradation of phenol in a packed bed reactor BABT

This paper proposes the use of a preliminary, phenol removal step to reduce peak loads arriving at a conventional effluent plant. A packed bed reactor (PBR) using polyurethane foam, porous glass and also cocoa fibres as the inert support material was used. Experiments have been carried out where the flow-rates, plus inlet and outlet phenol concentrations were measured. A simple, plug-flow model is proposed to represent the results. Zero, first order, Monod and inhibited kinetics rate equations were evaluated. It was found that the Monod model gave the best fit to the experimental data and allowed linear graphs to be plotted. The Monod saturation constant, K, is approximately 50 g m-3, and ka is around 900 s-1.

Relation between citric acid production by solid-state fermentation from cassava bagasse and respiration of Aspergillus niger LPB 21 in semi-pilot scale BABT

The aim of this work was to study the relation between citric acid production and respiration of Aspergillus niger LPB 21 in solid-state fermentation of cassava bagasse. The experiments were carried out in horizontal drum bioreactor coupled with a gas chromatography system. Fermentation was conduced for 144 h with initial substrate moisture of 60% using heat-treated cassava bagasse as sole carbon source. Alpha amylase from a fungal culture grown on oil cakes and its properties BABT

Solid-state fermentation was carried out for the production of alpha-amylase using Aspergillus oryzae. Different oil cakes such as coconut oil cake (COC) sesame oil cake (SOC), groundnut oil cake (GOC), palm kernel cake (PKC) and olive oil cake (OOC) were screened to be used as substrate for the enzyme production and also compared with wheat bran (WB). GOC was found to be the best producer of the enzyme among these. Combination of WB and GOC (1:1) resulted higher enzyme titres than the individual substrates. The study of xylanase production was conducted by Streptomyces viridosporus T7A in submerged (SmF) and solid-state fermentation (SSF), using agro-industrial residues and sub-products. Napier grass, sugarcane bagasse and soybean bran were used as carbon source, substrate/support, and nitrogen source, respectively. In SmF, Napier grass (1% v/w) supplemented with soybean bran, hydroxyethylcellulose and B complex vitamins were used. The objective of this work was to study the production of biomass with copper bioaccumulation in submerged fermentation using sugarcane molasses. Candida pelliculosa BARU 05 isolated from Baru (Dipteryx alata) was selected for its good capacity to accumulate the copper. The dependent variables evaluated were the amount of phycocyanin obtained and its purity, demonstrating that there might be a restructuring of phycobilisomes, especially when the culture was subjected to red light, which increased the purity level up to 33% with a reduction of 16% in phycocyanin content, but with higher photosynthetic efficiency compared to natural light.
