Factors affecting the production and regeneration of protoplasts from Colletotrichum lindemuthianum Ciência e Agrotecnologia

The present work reports factors affecting the production and regeneration of protoplasts from Colletotrichum lindemuthianum. The usefulness of protoplast isolation is relevant for many different applications and has been principally used in procedures involving genetic manipulation. Osmotic stabilizers, lytic enzymes, incubation time and mycelial age were evaluated in terms of their effects on protoplast yield. The optimal condition for protoplast production included the incubation of young mycelia (48 h) in 0.6 mol l-1 NaCl as the osmotic stabilizer, with 30 mg ml-1 Lysing Enzymes from Trichoderma harzianum for 3 h of incubation. Colletotrichum lindemuthianum (teleomorfo Glomerella cingulata f. sp. phaseoli) apresenta ampla variabilidade genética, demonstrada por suas características morfológicas. Com este trabalho, objetivou-se caracterizar, por meio de marcadores morfológicos, diferentes isolados de C. lindemuthianum e identificar marcadores morfológicos com uso potencial em análises genéticas. Antifungal activity and ultrastructural alterations in Pseudocercospora griseola treated with essential oils Ciência e Agrotecnologia

Pseudocercospora griseola, the etiologic agent of angular leaf spot of common bean (Phaseolus vulgaris), is an important disease in all bean-producing regions worldwide and may cause extremely high yield losses. The control of this disease is made more difficult by the pathogen's genetic variability and the inefficiency of fungicides. In this study, of 26 essential oils tested at different concentrations, 25 demonstrated efficiency in affecting the germination of strains 63-31 and 63-63 of the pathogen, reaching inhibition levels of between 80% and 100%. ABSTRACT: Early blight is one of the most important fungal diseases of potato. The objective of this study was to evaluate the in vitro reaction of potato genotypes to the severity of early blight and compare the species Alternaria solani and Alternaria grandis , etiologic agents of disease, through the evaluation of physiological characteristics. Twenty-two genotypes of the Potato Breeding Program at UFLA, two cultivars, Aracy and Bintje, patterns of resistance and susceptibility, respectively were evaluated. Phenotyping for angular leaf spot severity and its implication in breeding common bean for resistance Scientia Agricola

ABSTRACT: Phenotyping for severity of angular leaf spot (ALS) in common bean is important to identify new sources of resistance and select progenies in conventional and molecular marker-assisted breeding. In this study, three phenotyping methods for ALS severity were assessed and its implications in breeding for resistance to Pseudocercospora griseola are discussed. Reaction of 144 common bean lines to P. griseola was evaluated. Three different experiments were conducted in the greenhouse (V2 and V3 stages) and in the field. Common bean lines were inoculated with a mixture of spores of pathogen races 63-63 and 63-23.
