Avaliação de substratos e adubos orgânicos na aclimatização de plântulas de Heliconia psittacorum PAB

O objetivo deste trabalho foi avaliar a eficiência de três substratos orgânicos - casca de arroz carbonizada, pó de casca de coco seco e verde - e dois adubos -Vitasolo® e húmus de minhoca -, na aclimatização de plântulas de Heliconia psittacorum L., provenientes da micropropagação. Aos 75 dias, avaliaram-se a altura das plantas, o diâmetro do pseudocaule, o número de folhas e a área da terceira folha. A casca de arroz foi mais eficiente que o pó de casca de coco, verde ou seco; o pó de casca de coco verde foi mais eficiente que o pó de casca de coco seco; e o húmus foi mais eficiente que o Vitasolo®.

Obtaining tetraploid plants of ruzigrass (Brachiaria ruziziensis) R. Bras. Zootec.

The objective of the present work was to induce chromosome doubling in genotypes of Brachiaria ruziziensis with potential for production of good-quality biomass and tolerance/resistance to stress caused by aluminum and spittlebugs. Germinated seeds of B. ruziziensis were immersed in 0.1% colchicine solution for 2 or 3 hours. Ploidy level was determined by flow cytometry and confirmed by chromosome count. Fertility of the tetraploid plants was assessed through evaluation of pollen viability by staining. There was no statistical difference with regard to seedling survival and number of duplicate seedlings between the treatments. Survival rate was 8%, of which 11.45% were tetraploid genotypes. Breeding programs for Brachiaria spp. use both intraspecies and interspecies crosses between sexual and apomictic plants in order to obtain new cultivars with the desired characteristics. As there are different ploidy levels both within and between species of this genus, it becomes necessary to evaluate the genotypes used in breeding programs, as a guide to breeders when adopting crossing strategies. In this work, DNA content and chromosome number were determined in order to characterise ploidy levels in Brachiaria spp. genotypes. In the analysis of 15 genotypes, DNA content varied with the ploidy levels (2x, 3x and 4x), and between species and/or taxon. Protoplast production from napier grass and pearl millet triploid hybrids Ciência e Agrotecnologia

The objective of this work was to obtain protoplasts from napier grass and pearl millet triploid hybrids as a basis for future studies on chromosomal duplication. Explants were taken from mesophyll of in vitro- and in vivo-cultured plants or from calli of two triploid hybrids (H1 and H2), which were treated with enzymatic solutions containing different concentrations of cellulase R-10 (0.5, 1.0, 1.5 and 2.0%) with an additional 0.2% macerozyme and 0.1% driselase or 1.0% pectolyase Y-23 and 0.5% hemicellulase. Enzymatic digestion was monitored once every hour for five hours. Protoplasts were obtained from in vitro and in vivo leaflets of both triploid hybrids, and in vitro leaflets were the best explant sources. The quantity of produced protoplasts varied...

Histogram score contributes for reliability of DNA content estimatives in Brachiaria spp Ciência e Agrotecnologia

Flow cytometry allows to estimate the DNA content of a large number of plants quickly. However, inadequate protocols can compromise the reliability of these estimates leading to variations in the values of DNA content the same species. The objective of this study was to propose an efficient protocol to estimate the DNA content of Brachiaria spp. genotypes with different ploidy levels using flow cytometry.
