Uma das dificuldades de se conduzir estudos citogenéticos em espécies vegetais nativas do cerrado é a ocorrência de baixos índices metafásicos no meristema radicular. O incremento dos índices mitótico e metafásico em células de pontas de raiz tem sido obtido em várias espécies com a sincronização do ciclo celular por meio de hidroxiuréia (HU). Neste trabalho foram determinadas as condições ótimas para a sincronização eficiente do ciclo em células meristemáticas de raízes de baru (Dipteryx alata Vog.). Foram testadas quatro concentrações de HU (0,00; 1,25; 2,50; 5,00 e 10,00 mM), três tempos de exposição das raízes (10, 15 e 20 h) e dois tempos de recuperação do ciclo (5 e 10 h). The objective of this work was to compare the meiotic behavior and pollen grain viability of three species of Crotalaria. Slides for meiotic analysis were prepared by the air-drying technique. Pollen grain viability was measured by three staining procedures (Alexander's solution, tetrazolium chloride and fluorescein diacetate) and in vitro germination in a sucrose solution. Eight bivalents were observed, confirming previous reports on populations from other regions of Brazil, as well as from other countries. Abstract: The objective of this work was to evaluate, by genomic in situ hybridization (GISH), pairing configurations as potential indicators of recombination between chromosomes of different parental genomes, in two interspecific hybrids (elephant grass x pearl millet) artificially polyploidized. Anthers from young flower buds were used in the chromosomal preparations. The genomic probe was prepared with pearl millet DNA and labeled with biotin-16-dUTP by the nick translation reaction. Blocking DNA was prepared with genomic elephant grass DNA. The homoeologous intergenomic pairing, observed in the two hybrids, indicates the possibility of recombination between chromosomes of the parental genomes. |
