Paralytic shellfish poison accumulation yields and feeding time activity in the Pacific oyster (Crassostrea gigas) and king scallop (Pecten maximus) ArchiMer

Pacific oysters and king scallops placed individually in a recirculating flume were fed for 2 weeks with a constant concentration (120 cell ml(-1)) of a toxic strain of Alexandrium minutum. Fluorescence at the outlet of each experimental unit was measured continuously, and biodeposits were recovered twice daily to evaluate feeding time activity (FTA) and rates of organic filtration (OFR), ingestion (OIR) and organic absorption (OAR). Effect of environmental and nutritional factors on growth and azaspiracid production of the dinoflagellate Azadinium spinosum ArchiMer

Azadinium spinosum, a small dinoflagellate isolated from the North Sea, is a producer of azaspiracids (AZAs), a group of biotoxins associated with human illness following ingestion of contaminated shellfish. Using batch and continuous cultures of A. spinosum, the present study investigated the effects of different environmental and nutritional factors (salinity, temperature, photon flux density, aeration, culture media, nitrogen sources, phosphate source, and N/P ratios) on growth, maximum cell concentration, and AZA cell quota. Paralytic phycotoxin uptake by scallops (Pecten maximus) ArchiMer

It is difficult to apply sanitary standards for mollusc contamination by paralytic shellfish poisoning to animals not consumed whole. This problem is illustrated by the 1990 embargo on a Japanese shipment of frozen scallop muscle and gonad: the sanitary threshold applied to digestive gland (in Japan) does not guarantee that muscle and especially gonad are toxin-free. Accordingly, we performed experimental contaminations of scallops (Pecten maximus) from Port-en-Bessin (Normandy, France) using a toxic Japanese strain of Alexandrium tamarense. Distribution verticale de Dinophysis spp au cours d'un cycle de 48 heures dans le port d'Antifer (Normandie) ArchiMer

Time-related Dinophysis spp vertical profiles have been studyed during a 48 hours cycle in a submerged cylindric tank. A coïncidental Gymnodinium sp. (Scrippsiella trochoidea and Prorocentrum triestinum as companion species) red tide was believed to have strong effect on Dinophysis distribution in the upper layer during the tirst 24 hours cycle of the period of investigation (August 30-31, 1991). Between 24 and 48 hours, the sharp decrease of Dinophysis densities as well as the graduai decline observed in the red tide and in the vertical thermal gradient have resulted in a migration of dominant species : D. sacculus and D. cf. acuminata toward subsuperficiallevels (5 to 7 m.d) with a higher range in cells vertical movements. L'objectif général de l'étude est de simuler les épisodes de contamination de certains bivalves (huîtres et moules) par les toxines d'Alexandrium minutum et A. calenella à partir d'un module expérimental adapté, et en vue de construire un modèle de contamination applicable à ces espèces. Ce modèle sera ensuite susceptible d'être couplé aux modèles biologiques locaux (estuaire de Penzé et étang de Thau) de déterminisme des blooms, ainsi qu'aux modèles hydrodynamiques développés par différentes équipes (Ifremer, Université de Montpellier). La première année (2001-2002) était consacrée à la simulation des contaminations en estuaire de Penzé (Alexandrium minulum) et au développement de cultures en masse d'A. calenella de Thau en bioréacteurs.

Le maximum saisonnier de Dynophysis cf acuminata à Antifer (Normandie, France) ArchiMer

Since 1987 Antifer oil tanker terminal (Normandy, France) was weIl known, as a preferential area for summer highest blooms of Dinophysis cf acuminata, a toxic dinoflagellate. Three oceanographic cruises were made in August 1989, before, in the course of and after the seasonal max. of this species. Results obtained from these cruises (hydrological parameters and phytoplankton) as weIl as from a daily survey at a reference station in Antifer harbour, corrobora te the prevailing role of coastal up flowing of Seine estuarine waters, pushed to the coast by a dominant S - SW wind, on intensity and duration of Dinophysis bloom. Such observations are in agreement with those of Summer 1987 at reference station.

Etude de la décontamination de moules toxiques (toxines diarrhéiques) en laboratoire et en milieu naturel ArchiMer

Two studies of depuration, one in laboratory conditions and the other one in an oyster's pond were performed using 2 sets of contaminated mussels with diarrhetic toxins at a high and a low level (3 and 1 M.U.). Depuration kinetics were followed both by the adult mouse test and okadaïc acid analysis. Results are - in the pond, high toxic mussels need about 20 days to become eatable (toxicity level : 0.5 M.U.g-1 digestive glands), -this time decreases when the initial toxicity is low but it increases when mussels are underfed, - a good correlation is observed between the total toxicity and okadaïc acid concentration.

The relationship between azaspiracid shellfish poisoning and a small dinoflagellate, Azadinium spinosum, has been shown recently. The organism produces AZA1 and -2, while AZA3 and other analogues are metabolic products formed in shellfish. We evaluated whether mussels were capable of accumulating dissolved AZA1 and -2, and compared the toxin profiles of these mussels at 24 h with profiles of those exposed to live or lysed A. spinosum. We also assessed the possibility of preparative production of AZA metabolites by exposing mussels to semi-purified AZA1. Application of a digital pattern recognition system to Dinophysis acuminata and D-sacculus complexes ArchiMer

An image discrimination technique was developed to improve specific identification of some toxic Dinophysis cells (marine dinoflagellates involved in diarrhoeic shellfish poisoning), especially the ''acuminata'' and ''sacculus'' groups, which can be present at different ratios in natural sea-water samples collected during toxic episodes. This work was performed with image analysis software SAMBA (TITN Alcatel) using preserved cells directly observed through an inverted microscope and recorded with a video camera before further processing. The growth potential study of waters of the Vilaine bay was realized from march to september 1987 and for three sampling stations. The growth of depleted test cells of Skeletonema costatum was estimated and compared from existing nutrients pools for aIl water samples. As a first result it appears that inorganic nitrogen could stimulate the growth during spring while no clear relationship is observed between the algal growth and other hydrological parameters during spring or summer periods. Nevertheless these first results need further corroboration.

The dragonet Callionymus lyra, a target species used for evaluation of biological effects of chemical contaminants on French coasts ArchiMer

An oceanographic cruise in 1991 along the English Channel and Atlantic coasts of France indicated the value of dragonet Callionymus lyra Linnaeus, 1758 as a target species for monitoring biological effects. Ethoxyresorufin-O-deethylase measurements in this species provided a first assessment of the effects of hydrocarbons (PCB, PAH, dioxins) on the marine environment. The specific behavior of dragonet was defined by biochemical characterization. Since AZA poisoning has been reported following consumption of contaminated shellfish it is important to have these toxins available for toxicological studies, and a sustainable production of AZAs as calibrants in monitoring programs without having to rely on natural events. Influence de la Chloration sur les Premiers Stades Larvaires de Palaemon serratus (Pennant). ArchiMer

In order to estimate the effects of chlorine used in cooling systems of electric power plants on zooplankton, some experiments have been realized with the four first larval stages of the prawn (P. serratus Pennant). Chlorine injection was performed by constant adding at 16 and 20 degree C with times exposures of 5, 15 and 25 minutes. If there is no detectable variations in sensitivity of different stages, toxic effects of oxydants issued from chlorination can be observed (for both the two acclimatation temperatures) at 15 minutes exposure with a LD 50 : 32 mg/l of chlorine.

The occurrence of new phytoplankton species in a coastal area may be explained by the import of shellfish containing whole live algal cells in their digestive tracts. Indeed, shellfish containing toxic algal cells can induce both primary contaminations in safe areas ( initially free from toxic microalgae), and secondary contaminations of other shellfish growing in the same area via the faeces of the imported animals.
