Assessment of genetic diversity in Cattleya intermedia Lindl. (Orchidaceae) BABT

Orchids are valuable pot plants and Cattleya intermedia is a promising species underused in breeding programs. Recently, breeding work with this species produced superior plants that are believed to be not the true species owing to the morphological differences from wild plants. The aim of this study was to estimate the level of genetic diversity and interrelationships between wild and bred Cattleya intermedia collected at three different Brazilian states and from commercial breeders with RAPD markers. Genetic diversity in passion fruit (Passiflora spp.) evaluated by RAPD markers BABT

The objective of this study was to characterize the genetic diversity within a Passiflora collection by PCR-RAPD markers. Genetic analysis was performed in 70 accessions, representing 11 species of the genus Passiflora. The use of only five primers produced 136 reproducible polymorphic bands. The hierarchical classification showed high levels of dissimilarities between and within the species studied. A clear separation was obtained among species and accessions of P. edulis and P. edulis f. flavicarpa, which were forms of the high commercial value species. The fingerprints produced from the studied genotypes would allow the identification of improved varieties/populations in an easy, fast and inexpensive manner.

Temporal genotypic diversity of Schizaphis graminum (Rondani 1852) (Hemiptera: Aphididae) in a black oats (Avena strigosa) field BABT

The aim of this work was to analyze the clonal diversity variation in Schizaphis graminum during a complete phenological cycle of black oats (Avena strigosa). RAPD markers were used for detection of DNA polymorphisms of each clonal lineage, derived from a single clone collected weekly during a period of four months, in a crop field of black oats, Londrina, Paraná, Brazil. The monthly genotypic diversity was estimated by Shannon Information Index (H). Only four genotypes were distinguished from 122 specimens, with one of them overly predominant in all sampling dates (>60%). Induction of cassava somatic embryogenesis in liquid medium associated to floating membrane rafts BABT

The objective of this study was to examine the effect of two culture systems, liquid medium associated to floating membranes and solid medium, both supplemented with different concentrations of 2,4-D, in the induction of somatic embryogenesis of cassava (Manihot esculenta Crantz). Only 28% of the young leaf lobes (with 9 µM 2,4-D) were induced to form organized embryogenic structures (OES) with membrane rafts, compared to 50% of the explants presenting this type of tissue in solid medium with 36 µM of 2,4-D. Production of herbicide-resistant coffee plants (Coffea canephora P.) via Agrobacterium tumefaciens-mediated transformation BABT

Transgenic plants of Coffea canephora P. resistant to the herbicide ammonium glufosinate were regenerated from leaf explants after co-culture with Agrobacterium tumefaciens strain EHA105 harboring pCambia3301, a plasmid that contains the bar and the uidA genes both under control of 35S promoter. Direct somatic embryogenesis was induced on basal medium contained ¼ strength macro salts and half strength micro salts of MS medium, organic constituents of B5 medium and 30 g.L-1 sucrose supplemented with 5µM N6 - (2-isopentenyl)-adenine (2-iP). The phytohormone ethylene is involved in several physiological and developmental processes in higher plants, including ripening of fruits, abscission of organs and tissues, senescence, wound response as well as in other abiotic stresses. The enzyme 1-aminocyclopropane-1-carboxylate oxidase (ACO) catalyzes the last step of ethylene biosynthesis. The production of ethylene and the expression of a Coffea arabica ACO gene during the last stages of fruit maturation were investigated. Coffee is one of the most valuable agricultural commodities and ranks second on international trade exchanges. The genus Coffea belongs to the Rubiaceae family which includes other important plants. The genus contains about 100 species but commercial production is based only on two species, Coffea arabica and Coffea canephora that represent about 70 % and 30 % of the total coffee market, respectively. Increased synthesis of galactinol and raffinose family oligosaccharides (RFOs) has been reported in vegetative tissues in response to a range of abiotic stresses. In this work, we evaluated the transcriptional profile of a Coffea canephora galactinol synthase gene (CcGolS1) in two clones that differed in tolerance to water deficit in order to assess the contribution of this gene to drought tolerance. The expression of CcGolS1 in leaves was differentially regulated by water deficit, depending on the intensity of stress and the genotype. Produção de prolina e suscetibilidade ao glufosinato de amônio em plantas transgênicas de citrumelo Swingle PAB

O objetivo deste trabalho foi avaliar a sensibilidade de plantas transgênicas de citrumelo Swingle com elevada produção de prolina, ao herbicida glufosinato de amônio. As plantas utilizadas apresentavam a inserção do gene mutante da enzima delta1-pirrolina-5-carboxilato sintetase (P5CS), responsável pela biossíntese de prolina. A expressão do gene p5cs em plantas transgênicas causou aumento nas quantidades de prolina em tecidos foliares, em até cinco vezes, quando comparadas às plantas-controle tratadas com 200 µM de glufosinato de amônio. Analysis of the genetic diversity in Metopolophium dirhodum (Walker) (Hemiptera, Aphididae) by RAPD markers Rev. Bras. entomol.

The emergence of host-races within aphids may constitute an obstacle to pest management by means of plant resistance. There are examples of host-races within cereals aphids, but their occurrence in Rose Grain Aphid, Metopolophium dirhodum (Walker, 1849), has not been reported yet. In this work, RAPD markers were used to assess effects of the hosts and geographic distance on the genetic diversity of M. dirhodum lineages. Twenty-three clones were collected on oats and wheat in twelve localitites of southern Brazil. From twenty-seven primers tested, only four primers showed polymorphisms. Utilização de nutrientes por células de algodoeiro cultivadas em suspensão Scientia Agricola

Com o objetivo de conhecer os requerimentos nutricionais de suspensões celulares de algodoeiro foi feita uma análise da utilização de nutrientes no intervalo de 30 dias, normalmente utilizado entre subculturas. As culturas de células em suspensão de algodoeiro, genótipo 101-102B, foram estabelecidas e subcultivadas mensalmente em meio líquido. Foram analisadas o crescimento das culturas, as variações de pH do meio de cultura e a absorção de nutrientes. A matéria fresca das culturas de células foi superior a 6 vezes o valor inicial após 30 dias de cultivo. Houve aumento do pH no início da cultura, estabilizando-se em 6,2 próximo à fase estacionária.
