The seed coat of Phaseolus vulgaris interferes with the development of the cowpea weevil [Callosobruchus maculatus (F.) (Coleoptera: Bruchidae)] Anais da ABC (AABC)

We have confirmed here that the seeds of the common bean (Phaseolus vulgaris, L.) do not support development of the bruchid Callosobruchus maculatus (F.), a pest of cowpea [Vigna unguiculata (L.) Walp] seeds. Analysis of the testa (seed coat) of the bean suggested that neither thickness nor the levels of compounds such as tannic acid, tannins, or HCN are important for the resistance. On the other hand, we have found that phaseolin (vicilin-like 7S storage globulin), detected in the testa by Western blotting and N-terminal amino acid sequencing, is detrimental to the development of C. maculatus. O efeito da adição de quitina sobre o desenvolvimento e a sobrevivência do bruquídeo Callosobruchus maculatus (F.) foi estudado empregando-se um sistema de sementes artificiais feitas com farinha de Vigna unguiculata (feijão-de-corda). O polissacarídeo interferiu no desenvolvimento do inseto causando uma diminuição na taxa de sobrevivência. O índice de Howe mostra que as dietas contendo quitina são nutricionalmente inadequadas.

Cysteine proteinases and cystatins BABT

This review describeds the definition, localization, functions and examples of cysteine proteinases and their protein inhibitors in vertebrate, non-vertebrate animals and plants. These inhibitors are related with defense mechanisms of plant against pests. It also describes the factors involved in the specific cysteine proteinase-cystatin interaction and high degree of affinity and large specificity in this interaction which are not only represented by the compatibility between amino acid residues of the active site involved in catalysis, but also of all amino acid residues that participante in the enzyme-inhibitor interaction.

Cowpea Vicilins: Fractionation of Urea Denatured Sub-Units and Effects on Callosobruchus maculatus F. (Coleoptera: Bruchidae) Development BABT

Vicilins (7S storage globulins) isolated from cowpea (Vigna unguiculata L.) seeds which were susceptible (S) and resistant (R) to the cowpea weevil (Callosobruchus maculatus F., Coleoptera: Bruchidae) were denatured by urea and fractionated by ion-exchange chromatography. Isolated fractions were incorporated in artificial seeds for assessment of their toxicity to C. maculatus. The most acidic fractions of both susceptible (CE-31 cultivar) and resistant (IT81D-1045 line) seeds were shown to affect development and survival of the bruchid. Results indicated that vicilin polypeptides of toxic nature were expressed in both types of storage globulins although at different levels.

The presence of insulin in plants is not accepted by the scientific community in general. In this review we discuss this paradigm and retrieve information that strongly suggests that insulin is indeed found in plants. We present results, which indicate that a protein molecule with the same amino acid sequence as bovine insulin is expressed in leguminous plants. Additionally, we provide evidence that proteins associated with insulin signalling pathways in vertebrates are also found in association with insulin-like molecules in plants.
