Successive mini-cuttings collection in Piptocarpha angustifolia mini-stumps: effects on maturation, adventitious root induction and root vigor Agronomy
Stuepp, Carlos André; Wendling, Ivar; Koehler, Henrique Soares; Zuffellato-Ribas, Katia Christina.
The low percentage of seed germination and adventitious root induction in cuttings of Piptocarpha angustifolia has limited its silvicultural use. Thus, we studied the effect of tissue maturation on successive collections of mini-cuttings grown in a semi-hydroponic system. Likewise, we evaluated potential multiplication via the mini-cutting technique by assessing the survival and productivity of mini-stumps as well as the formation of adventitious roots and the root vigor. We made 32 sprout collections during two years in a clonal mini-garden. From these collections, mini-cutting experiments were installed throughout several seasons. Mini-cuttings of 8±1 cm long were prepared, planted in boxes with vermiculite and carbonized rice husk (1:1 v/v) and placed...

The southern State of Rio Grande do Sul (RS) is the main rice producer in Brazil with a 60% participation of the national production and 86% participation of the region. Rice culture irrigation system is done by flooding, which leads to soil salinization, a major environmental constraint to production since it alters the plants’ metabolism exposed to this type of stress. The indica cultivar, widely used in RS, has a higher sensitivity to salinity when compared to that of the japonica cultivar in other physiological aspects. Current research analyzes enzymes expression involved in salt-subjected indica and japonica rice cultivars’ respiration. Oryza sativa L. spp. japonica S.Kato (BRS Bojuru, IAS 12-9 Formosa and Goyakuman) and Oryza sativa L. spp. indica...

Dynamics in global DNA methylation and endogenous polyamine levels during protocorm-like bodies induction of Cattleya tigrina A. Richard Biological Sciences
Almeida, Vivian; Fraga, Hugo Pacheco de Freitas; Navarro, Bruno Bachiega; Guerra, Miguel Pedro; Pescador, Rosete.
Cattleya tigrina is endemic to the Atlantic forest biome and classified as vulnerable in the Red Book of Brazilian Flora. In vitro techniques comprise valuable tools for the conservation of endangered plant species. The aim of the present study was to evaluate the morphological features, global DNA methylation levels and free polyamines during protocorm-like bodies (PLBs) induction of C. tigrina. Along with that, an efficient protocol for in vitro propagation of this species is proposed. The first evidences of PLBs induction in C. tigrina occurred at seven days in culture, starting from the basal portion of the leaf abaxial surface. A hypomethylation marked the beginning of cell differentiation, followed by an increased global DNA methylation at 35 days in...
