Analysis of the genetic diversity of Eucalyptus cladocalyx (sugar gum) using ISSR markers Agronomy
Ballesta, Paulina; Mora, Freddy; Contreras-Soto, Rodrigo Ivan; Ruiz, Eduardo; Perret, Sandra.
Eucalyptus cladocalyx F. Muell is a tree endemic to southern Australia and is distributed across four isolated regions: Kangaroo Island, southern Flinders Ranges, and two geographical zones in Eyre Peninsula. E. cladocalyx is capable of growing under extreme environmental conditions, including dry and saline soils. The objective of this study was to analyze genetic diversity in 45 half-sib families planted in northern Chile that are distributed across five different zones (provenances). Genetic variability was assessed using ISSR (Inter Simple Sequence Repeat) molecular markers. The results showed low levels of genetic diversity within populations (He = 0.113 to 0.268) in contrast with other Eucalyptus species. In addition, there was a significant genetic...

Molecular characterisation of Aspergillus flavus isolates from peanut fields in India using AFLP BJM
Singh,Diwakar; Radhakrishnan,T.; Kumar,Vinod; Bagwan,N.B.; Basu,M.S.; Dobaria,J.R.; Mishra,Gyan P.; Chanda,S.V..
Aflatoxin contamination of peanut, due to infection by Aspergillus flavus, is a major problem of rain-fed agriculture in India. In the present study, molecular characterisation of 187 Aspergillus flavus isolates, which were sampled from the peanut fields of Gujarat state in India, was performed using AFLP markers. On a pooled cluster analysis, the markers could successfully discriminate among the ‘A’, ‘B’ and ‘G’ group A. flavus isolates. PCoA analysis also showed equivalent results to the cluster analysis. Most of the isolates from one district could be clustered together, which indicated genetic similarity among the isolates. Further, a lot of genetic variability was observed within a district and within a group. The results of AMOVA test revealed that...

Carrasco,Basilio; Díaz,Carole; Moya,Mario; Gebauer,Marlene; García-González,Rolando.
The genetic characterization of 29 elite Japanese plum cultivars (Prunus salicina) and 4 Prunus cultivars was carried out by analyzing 97 Simple Sequence Repeat (SSR) alleles and 232 binary Inter Simple Sequence Repeat (ISSR) loci. A high level of genetic variability was found for these two molecular markers among the Japanese plum cultivars compared to other Prunus species. On average, the variability found by analyzing the SSR alleles were Na = 12.1, Ne = 5.2, Ho = 0.9, He = 0.8 and F= -0.127, whereas ISSR yielded values of h = 0.15 and I = 0.27. The genetic relationship among cultivars was estimated with Principal Coordinate Analysis (PCA) and a Bayesian clustering approach using the software program Structure. This program identified two subgroups...

Genetic diversity in two variants of Orobanche gracilis Sm. [var. gracilis and var. deludens (Beck) A. Pujadas] (Orobanchaceae) from different regions of Spain Electron. J. Biotechnol.
Román,Belén; Hernández,Rosabell; Pujadas-Salvá,Antonio José; Cubero,José Ignacio; Rubiales,Diego; Satovic,Zlatko.
The pattern of genetic variation among populations of two Orobanche gracilis Sm. taxa (var. gracilis and var. deludens (Beck) A. Pujadas) from Northern and Southern Spain growing on different hosts was analysed using RAPD markers. The diversity analysis within populations revealed a higher level of diversity in the populations from the North when compared to the Southern ones. The results of principal co-ordinate analysis (PCoA) based on Dice distances among samples clearly established the separation of samples according to the taxonomical variety and the geographical origin of each population. The Southern populations of both var. gracilis and var. deludens were more differentiated among them than those of var. gracilis from the North. The analysis of...

Pinarkara,Emine; Kayis,Seyit A; Hakki,Erdogan E; Sag,Ayla.
Cannabis sativa L. is a multiple-use plant. However, its cultivation is strictly controlled due to its psychoactive nature and usage in producing drugs such as marijuana, and hashish. In this study, psychoactive type Cannabis samples, which were seized from 29 different locations of Turkey, were used. Interests were to identify the genetic relatedness of the seized samples and to partition molecular variance between and within populations. Randomly Amplified Polymorphic DNAs were employed for analysis based on single plant material and bulked samples of them. Data were analysed via cluster and principal coordinate analyses (PCoA). Analysis of molecular variance (AMOVA) was performed to obtain variations between and within populations. Cannabis accessions...

Structure and genetic relationships between Brazilian naturalized and exotic purebred goat domestic goat (Capra hircus) breeds based on microsatellites Genet. Mol. Biol.
Oliveira,Joelliton Domingos de; Igarashi,Maria Luiza Silveira de Paiva; Machado,Théa Mírian Medeiros; Miretti,Marcos Mateo; Ferro,Jesus Aparecido; Contel,Eucleia Primo Betioli.
The genetic relationships and structure of fourteen goat (Capra hircus) populations were estimated based on genotyping data from 14 goat populations (n = 410 goats) at 13 microsatellite loci. We used analysis of molecular variance (AMOVA), principal component analysis (PCA) and F statistics (F IS, F IT and F ST) to evaluate the genetic diversity (Ho, He and ad) of the goats. Genetic distances between the 14 goat populations were calculated from allelic frequency data for the 13 microsatellite markers. Moderate differentiation was observed for the populations of the undefined breeds (including the Anglo-Nubian-M breed), the naturalized Brazilian breeds (Moxotó, Canindé), the exotic purebred breeds (Alpine, Saanen, Toggenbourg and Anglo-Nubian) and the...

Human population genetic structure detected by pain-related mu opioid receptor gene polymorphisms Genet. Mol. Biol.
López Soto,Eduardo Javier; Catanesi,Cecilia Inés.
Several single nucleotide polymorphisms (SNPs) in the Mu Opioid Receptor gene (OPRM1) have been identified and associated with a wide variety of clinical phenotypes related both to pain sensitivity and analgesic requirements. The A118G and other potentially functional OPRM1 SNPs show significant differences in their allele distributions among populations. However, they have not been properly addressed in a population genetic analysis. Population stratification could lead to erroneous conclusions when they are not taken into account in association studies. The aim of our study was to analyze OPRM1 SNP variability by comparing population samples of the International Hap Map database and to analyze a new population sample from the city of Corrientes,...

Genetic diversity of Bemisia tabaci (Genn.) Populations in Brazil revealed by RAPD markers Genet. Mol. Biol.
Lima,L.H.C.; Campos,L.; Moretzsohn,M.C.; Návia,D.; Oliveira,M.R.V. de.
Bemisia tabaci (Genn.) was considered a secondary pest in Brazil until 1990, despite being an efficient geminivirus vector in beans and soybean. In 1991, a new biotype, known as B. tabaci B biotype (=B. argentifolii) was detected attacking weed plants and causing phytotoxic problems in Cucurbitaceae. Nowadays, B. tabaci is considered one of the most damaging whitefly pests in agricultural systems worldwide that transmits more than 60 different plant viruses. Little is known about the genetic variability of these populations in Brazil. Knowledge of the genetic variation within whitefly populations is necessary for their efficient control and management. The objectives of the present study were to use RAPD markers (1) to estimate the genetic diversity of B....

Liu,Yan-Qun; Qin,Li; li,Yu-Ping; Wang,Huan; Xia,Run-Xi; Qi,Yong-Hong; li,Xi-Sheng; Lu,Cheng; Xiang,Zhong-Huai.
The genetic diversity and genetic structure of three Chinese silkworm species Bombyx mori L., Antheraea pernyi Guérin-Meneville and Samia cynthia ricini Donovan were comparatively assessed based on RAPD markers. At the species level, A. pernyi and B. mori showed high levels of genetic diversity, whereas S. cynthia ricini showed low level of genetic diversity. However, at the strain level, A. pernyi had relatively highest genetic diversity and B. mori had lowest genetic diversity. Analysis of molecular variance (AMOVA) suggested that 60% and 72% of genetic variation resided within strains in A. pernyi and S. cynthia ricini, respectively, whereas only 16% of genetic variation occurred within strains in B. mori. In UPGMA dendrogram, individuals of A. pernyi...

Morphological and genetic differentiation among Chilean populations of Bufo spinulosus (Anura: Bufonidae) RChHN
Méndez,Marco A.; Soto,Eduardo R.; Correa,Claudio; Veloso,Alberto; Vergara,Eliseo; Sallaberry,Michel; Iturra,Patricia.
Bufo spinulosus has a wide and fragmented range distribution in Chile (18° to 33° S) along altitudinal and latitudinal gradients. Genetic variation was estimated using RAPD (Random Amplified Polymorphic DNA) markers in 10 populations from northern and central Chile. Morphometric and genetic information was analyzed as a function of geographical origin. The correlation between genetic and morphometric differentiation was analyzed by the Mantel test. An increase in body size as a function of latitude was observed. Specimens from El Tatio had the smallest body size and the greatest morphometric divergence. The AMOVA applied to genetic data indicated that 57.85 % of the variance is explained by interregional differences and that 30.12 % of the...

Bressan,Eduardo de Andrade; Briner Neto,Thiago; Zucchi,Maria Imaculada; Rabello,Ronaldo José; Veasey,Elizabeth Ann.
Traditional growers of the Vale do Ribeira, São Paulo State, grow and make use of several D. alata landraces. This study assessed the genetic diversity of 16 landraces, using isozymatic and morphological markers and comparing them with 19 commercial varieties of D. alata. Their distribution in different levels of organization such as households and communities of the Vale do Ribeira was evaluated. Isozymatic analyses were performed with polyacrylamide (six systems) and starch gels (one system), while the morphological analyses were carried out with 24 traits. Due to the polyploid nature of this species, the isozymatic bands were scored as binary data. Morphological traits were also scored as binary data. Principal coordinates and cluster analyses were...
