ABSTRACT Molecules with immune modulating activity are ubiquitously distributed in nature and their impact on aquaculture has been exploited in order to increase fish resistance to pathogens. Here, we investigated the effect of inactivated Parapoxvirus ovis (iPPVO) on blood cells and innate and acquired immune response of silver catfish (Rhamdia quelen). iPPVO inoculation had no effect on respiratory burst activity; however, following iPPVO inoculation, we observed a significant decrease on circulating monocytes concomitantly with an increased number of heterophilic granulocytes and thrombocytes, which are the main cells involved in innate immunity and provide connection with acquired immunity. Fish inoculated with a combination of bovine serum albumin... |

In the present work, immunogenicity of recombinant in vitro assembled hepatitis C virus core particles, HCcAg.120-VLPs, either alone or in combination with different adjuvants was evaluated in BALB/c mice. HCcAg.120-VLPs induced high titers of anti-HCcAg.120 antibodies and virus-specific cellular immune responses. Particularly, HCcAg.120-VLPs induced specific delayed type hypersensitivity, and generated a predominant T helper 1 cytokine pro file in immunized mice. In addition, HCcAg.120-VLPs prime splenocytes proliferate in vitro against different HCcAg.120-specific peptides, depending on either the immunization route or the adjuvant used. Remarkably, immunization with HCcAg.120-VLPs/Montanide ISA888 formulation resulted in a significant control of... |

Aluminum salts have been widely used in vaccine formulations and, after their introduction more than 80 years ago, only few vaccine formulations using new adjuvants were developed in the last two decades. Recent advances in the understanding of how innate mechanisms influence the adaptive immunity opened up the possibility for the development of new adjuvants in a more rational design. The purpose of this review is to discuss the recent advances in this field regarding the attempts to determine the molecular basis and the general mechanisms underlying the development of new adjuvants, with particular emphasis on the activation of receptors of innate immune recognition. One can anticipate that the use of these novel adjuvants will also provide a window of... |

Successful vaccine application means maximum protection with minimal number of administrations. A rational development of vaccines involves studies of the nature of the antigen as well as of the adjuvant to be used to improve the immune responses. This has provided the impetus for studies to design the degradable devices and for different approaches to antigen delivery by different routes of administration. The development of controlled release systems based on polymeric devices that permit a sustained or pulsed release of encapsulated antigens has attracted much interest. Polymeric delivery systems consist of polymers that release their content continuously in a controlled manner over a period of time. The development of a biocompatible delivery system... |

The attenuated vaccine against Schistosoma mansoni induces Th1-mediated protective immunity and we have sought to identify a role for IL-12 in this model. Elevated levels of IL-12 (p40 mRNA) were detected in the lymph nodes (LN) and the lungs of vaccinated mice, whilst treatment of vaccinated mice with anti-IL-12 antibodies decreased the ratio of IFNg:IL-4 secreted by in vitro-cultured LN cells. However, there was only marginal abrogation of the level of resistance in these mice. Soluble antigens from the lung-stage of the parasite (SLAP) appeared to be efficient stimulators of IFNg and IL-12 secretion. These antigens when used to immunise mice in conjunction with IL-12 as an... |

Fish vaccination has been increasingly exploited as a tool to control pathogen infection. The production of immunoglobulin following vaccination might be affected by several factors such as management procedures, water temperature, and the presence of xenobiotics. In the present study, we aimed to investigate the kinetics of immunoglobulin production in silver catfish (Rhamdia quelen) inoculated with inactivated Aeromonas hydrophila and kept at two different water temperatures (17.4±0.4° or 21.3±0.3°C). The effect of a second antigen inoculation and exposure of fish to sublethal concentrations of the herbicides atrazine and glyphosate at 10% of the lethal concentration (LC50-96h) on specific serum antibodies were also investigated. Antibodies to A.... |

Neosporosis is caused by the intracellular protozoan parasite Neospora caninum. This major disease-causing pathogen is responsible for inducing abortion in cattle, and these adverse events occur sporadically all over the world, including Japan. Currently, there are no vaccines on the market against infection with N. caninum. Because live and attenuated vaccines against N. caninum have had safety and effectiveness issues, development of a next-generation vaccine is urgently required. To develop a vaccine against neosporosis, my laboratory has been focused on the following: 1) understanding the host immune responses against Neospora infection, 2) identifying vaccine antigens and 3) developing an effective antigen-delivery system. The research strategy taken... |

Saponin is the generic name of steroid or triterpene glycosides, and the capacities of some saponins to stimulate both Th1 immune response and production of cytotoxic T cells are useful as vaccine components against intracellular pathogens. Because saponins have been found commonly in starfish, we assessed the potential of starfish, Asterias amurensis and Asterina pectinifera, as adjuvant sources. Crude starfish saponins had hemolytic activities (EC50=10 to 100 μg/ml) and thin layer chromatography indicated heterogeneity of their constituents. When starfish saponis were subcutaneously injected into mice with ovalbumin (OVA), OVA-specific IgG, especially IgG2a instead of IgG1 was produced in mouse blood, suggesting starfish saponins stimulated Th1 type... |

ABSTRACT: Adjuvants, such as mineral oils, are widely used in the application of herbicides by reducing the drift and evaporation of the droplets and by increasing herbicide uptake by the plant. However, little is known about how mineral oil behaves when in contact with the soil. Thus, the objective of this research was to evaluate the transport of atrazine via leaching with the addition of mineral oil in a soil agricultural under laboratory conditions. To quantify the concentration of the herbicide along the profile of the very clay soil column (30 cm), 14C-atrazine (ring-U-14C) was used with 16,667 Bq of radioactivity per column in volume of 200 mL solution and 0 (control), 1, and 2% mineral oil (v/v) was added in the application volume. Atrazine was... |

ABSTRACT: Using an adjuvant to optimize and increase the foliar activity of post emergence herbicides is an acceptable way to reduce their side effects. Field researches were conducted to detect a suitable adjuvant for two post-herbicides namely, isoproturon+tribenuron and pyroxsulam against little seed canary grass (Phalaris minor) and field bindweed (Convolvulus arvensis). Treatments were, isoproturon+tribenuron at 1,025 g a.i. ha-1 (recommended dose) and pyroxsulam at 140.6 g a.i. ha-1 (recommended dose) alone or their reduced dose (75% of the recommended dose) isoproturon+tribenuron at 769 and pyroxsulam at 105.5 g a.i. ha-1, tank mixed with three different adjuvants viz. alkylethersulphate sodium salt at 400, fatty alcohol ethoxylate at 200 and... |

ABSTRACT: Tank mixed application of herbicides with adjuvant is required for effective weed management and to reduce herbicide load on agricultural crops. Field studies were conducted to investigate the effect of herbicides at reduced rates with adjuvant on common goosefoot (Chenopodium album) and field bindweed (Convolvulus arvensis) and grain yield of wheat for two consecutive winter seasons. Treatments included post-emergence foliar application of four herbicides viz., fluroxypyr + tribenuron-methyl + clopyralid, mesosulfuron-methyl + iodosulfuron-methyl sodium, bromoxynil + MCPA, iodosulfuron-methyl sodium (pre mixed formulations) at their recommended rates 244.5, 14.2, 444.6, 10 g a.i. ha-1, respectively and 25% reduced rates with alkyl ether sulfate... |
