Bacteries associees aux production de Brachionus plicatilis ArchiMer
Nicolas, Jean-louis; Joubert, M.
Bacteria associated with semi-continuous culture of Brachionus plicatilis , which is used as the first food of marine fish larvae, were counted on Zobell 2216 E medium and identified. The numbers of bacteria were very high in both Brachionus (about 10 super(11) bact. per ml of digestive content) and in water (from 1.6.10 super(6) to 4.7.10 super(7) bact. per ml). The cultures were studied at driving a time when mortalities occurred frequently. The introduction of a Pseudomonas) phenon at a proportion of about 50% to the dominant microflora persisted in time and was little influenced by diet. It has been the cause of the repression of the Vibrio, Aeromonas group which was found in an unusually small proportion. It induced a lack of microflora diversity and...

Microbiological analysis of critical points in the chicken industry BABT
Cansian,Rogério Luis; Floriani,Salete Teresa Radeski; Valduga,Eunice.
This work is focused on identifying microbial contamination in the scalding asepsis and cooling processes as well as in fresh sausages obtained. Salmonella was identified in two scald water samples but was absent in the water from chiller and in the final product, which might be explained in terms of chlorine addition and temperature reduction. The analysis revealed that MPN of Escherichia coli was in the range of <1-11/mL in the scald water and <1-64/mL in the chiller. Aeromonas sp. count showed 5 to 3.5x10¹ CFU/mL in the scald water and 9 to 3.7x10² CFU/mL in the chiller, probably due to the psycrophile characteristics of Aeromonas and evisceration process. The analysis of fresh sausages chicken revealed a sharp increase in the Aeromonas count...

The occurrence of Aeromonas spp. in the bottled mineral water, well water and tap water from the municipal supplies BABT
Scoaris,Denise de Oliveira; Bizerra,Fernando Cezar; Yamada-Ogatta,Sueli Fumie; Abreu Filho,Benício Alves de; Ueda-Nakamura,Tânia; Nakamura,Celso Vataru; Dias Filho,Benedito Prado.
The aim of this work was to study the occurrence of Aeromonas sp in the bottled mineral water, well water and tap water from the municipal supplies. Positive samples were found for Aeromonas spp. 12.7% from the mineral water, 8.3% from the artesian water and 6.5% from the tap water. The recovery of Aeromonas spp. was significantly higher in the bottled mineral and artesian water than in the tap water from municipal supplies. The occurrence of the Aeromonas spp. did not correlate significantly with the contamination indicator bacteria (i.e. total coliforms) in the artesian water samples. However, a significant correlation was found between Aeromonas spp. and total coliforms in the both mineral water and tap water samples. The presence or absence of a...

A broad pH range and processive chitinase from a metagenome library BJMBR
Thimoteo,S.S.; Glogauer,A.; Faoro,H.; de Souza,E.M.; Huergo,L.F.; Moerschbacher,B.M.; Pedrosa,F.O..
Chitinases are hydrolases that degrade chitin, a polymer of N-acetylglucosamine linked β(1-4) present in the exoskeleton of crustaceans, insects, nematodes and fungal cell walls. A metagenome fosmid library from a wastewater-contaminated soil was functionally screened for chitinase activity leading to the isolation and identification of a chitinase gene named metachi18A. The metachi18A gene was subcloned and overexpressed in Escherichia coli BL21 and the MetaChi18A chitinase was purified by affinity chromatography as a 6xHis-tagged fusion protein. The MetaChi18A enzyme is a 92-kDa protein with a conserved active site domain of glycosyl hydrolases family 18. It hydrolyses colloidal chitin with an optimum pH of 5 and temperature of 50°C. Moreover, the enzyme...

Total protein electrophoresis and RAPD fingerprinting analysis for the identification of Aeromonas at the species level BJM
Delamare,Ana Paula Longaray; Artico,Liane de Oliveira; Grazziotin,Felipe Gobbi; Echeverrigaray,Sergio; Costa,Sérgio Olavo Pinto da.
Fifteen well-defined strains of Aeromonas of thirteen species were analyzed by SDS protein electrophoretic analysis (SDS-PAGE) and random amplified polymorphic DNA analysis (RAPD). The comparison between the patterns obtained by both methods allowed differentiating all the strains. Clusters formed by the unweighted pair group method with arithmetic averages applied to protein data correlates with the genetic and biochemical information about the species. The results show that protein fingerprinting has the potential to differentiate Aeromonas species, but the low qualitative variation indicates that this technique is not efficient for the characterization of strains within a species. Conversely, RAPD fingerprinting allows the identification of strains but...

Antimicrobial susceptibility, hemolysis, and hemagglutination among Aeromonas spp. isolated from water of a bovine abattoir BJM
Bizani,Delmar; Brandelli,Adriano.
The presence of Aeromonas spp. in food has been demonstrated. They are often introduced from water, which is the natural habitat of many species and is thought to be the main source of contamination. The occurrence of Aeromonas spp. was investigated in 70 water samples of a bovine abattoir. Aeromonas spp. were present in 21.4% of water samples. A. hydrophila was isolated from 11.4% of supplying water samples and from 25.7% of the water drained from carcasses, whereas A. sobria was sole isolated from 5.7% of supplying water samples. Although greater number of positive samples were from water drained from carcasses, only A. hydrophila isolates were present. This suggests that the supplying water is the source of contamination. The antibiotic susceptibility...

The effect of organic osmoprotectors on Aeromonas trota and A. hydrophila grown under high sodium chloride concentrations BJM
Delamare,Ana Paula L.; Dalcin,Thais; Müller,Gabriela; Costa,Sergio Olavo Pinto da; Echeverrigaray,Sergio.
The effect of organic solutes on the growth of Aeromonas trota and A. hydrophila was evaluated. Proline and glutamic acid were not effective as osmoprotectors, but betaine exerts osmoprotection allowing the growth of both strains in inhibitory concentrations of NaCl. Growth kinetics suggests that the halotolerance difference between the strains is associated with the synthesis of osmolytes rather than betaine uptake.
