犬のE.canisおよびA.ρhagocytophilum感染状況を明らかにするため,従来法である蛍光抗体法(IFA),および組換え主要膜表面蛋白質を抗原とした酵素抗体法を原理とする新規の特異抗体検出キットを組合せて全国的血清疫学調査を行った. IFAによるスクリーニング検査では1,206検体中64頭(5.3%)がE.canis 20倍以上の抗体価を示したが,これら64頭は特異抗体検出キットではすべてE.canis陰性であり,E. canis以外の近縁種に感染していたと考えられた.いっぽうIFAでA.ρhagocytophilumに対して20倍以上の抗体価を示したものは1,206検体中3頭(0.2%)であった.うち1頭は特異抗体検出キットでもA.phagocytophilum陽性を示し,日本にもA.phagocytophilumに感染した犬 がいることが示唆された.しかし,抗体陽性犬に臨床症状は認められず,病原性との関連は不明であった. Anaplasma phagocytophilum causes human granulocytic anaplasmosis (HGA) and tick-borne fever in ruminants. A closely related and potentially novel Anaplasma sp. in Japan was recently characterized. The aims of the study were to provide molecular evidence for the presence of these 2 species in Japan, and to develop a reliable PCR method based on the nucleotide differences within the citrate synthase (gltA) gene. DNA samples from 182 ixodid ticks (134 Ixodes persulcatus, 35 Haemaphysalis douglasii and 13 I. ovatus) collected from 2 sites in Hokkaido, Japan, were screened for A. phagocytophilum and its closely related Anaplasma sp. Anaplasma marginale and A. phagocytophilum are intracellularrickettsial pathogens causing bovine anaplasmosis and human granulocyticanaplasmosis, respectively. Effective vaccines for the control ofanaplasmosis are not available despite attempts using differentapproaches, such as attenuated strains, infected erythrocyteand tick cell-derived purified antigens. Recent reports demonstrated that A. marginale and A. phagocytophilum co-exist in geographic areas, that concurrent infections may occur in ruminants and ticks and that there are similarities and differences at molecular level between both species. The aim of this article is to make a comparison between the main characteristics of Anaplasma marginale and Anaplasma phagocytophilum. |
