Inactivated Parapoxvirus ovis as inducer of immunity in silver catfish (Rhamdia quelen) Anais da ABC (AABC)
PAVAN,TATIANA R.; NIED,CRISTIAN O.; NORO,MIRELA; ANZILIERO,DENIZ; FRANDOLOSO,RAFAEL; KREUTZ,LUIZ CARLOS.
ABSTRACT Molecules with immune modulating activity are ubiquitously distributed in nature and their impact on aquaculture has been exploited in order to increase fish resistance to pathogens. Here, we investigated the effect of inactivated Parapoxvirus ovis (iPPVO) on blood cells and innate and acquired immune response of silver catfish (Rhamdia quelen). iPPVO inoculation had no effect on respiratory burst activity; however, following iPPVO inoculation, we observed a significant decrease on circulating monocytes concomitantly with an increased number of heterophilic granulocytes and thrombocytes, which are the main cells involved in innate immunity and provide connection with acquired immunity. Fish inoculated with a combination of bovine serum albumin...

Clinical and laboratory aspects of common variable immunodeficiency Anais da ABC (AABC)
Kokron,Cristina M.; Errante,Paolo R.; Barros,Myrthes T.; Baracho,Gisele V.; Camargo,Maristela M.; Kalil,Jorge; Rizzo,Luiz V..
Common variable immunodeficiency (CVID) is an immunological disorder characterized by defective antibody production, recurrent infections, most notably of the respiratory tract, autoimmune phenomena and cancer. Some CVID patients may also present disturbances of the cellular immune response such as a decrease in the number and proportion of different lymphocyte populations, diminished lymphoproliferative response to mitogens and antigens, altered production of cytokines, and deficient expression of cell-surface molecules. Most Brazilian CVID patients included in this study show a decrease in T and B lymphocyte counts in the peripheral blood. Furthermore, their lymphocytes are more susceptible to apoptosis following activation than normal individuals, and...

Antibodies specific to infectious bronchitis in broilers in Ceará state, Brazil Arq. Bras. Med. Vet. Zootec.
Cardoso,W.M.; Gomes,L.P.; Romão,J.M.; Salles,R.P.R.; Teixeira,R.S.C.; Sobral,M.H.N.R.; Câmara,S.R.; Oliveira,W.F..
The experiment was carried out to determine the antibody levels to infectious bronchitis virus (IBV) in 1120 broilers of two broiler flocks, both from the same parental flock and free from previous vaccination. Forty chicks of each line were alloted to the control group and the sera were tested by indirect ELISA. The vaccination program consisted on the administration of commercial vaccines against IBV at 10 and 25 days of age. Chicks with low levels of maternal antibodies (Mab) did not show significant titers to the first vaccinal stimulus. They presented a vaccinal response to the second vaccinal stimulus reaching the top around GMT 1100 at 45 days. Chicks with high Mab titers did not show significant titers to the primary and secondary vaccinal stimuli,...

Antibody conjugate radioimmunotherapy of superficial bladder cancer BABT
Perkins,Alan; Hopper,Melanie; Murray,Andrea; Frier,Malcolm; Bishop,Mike.
The administration of antibody conjugates for cancer therapy is now proving to be of clinical value. We are currently undertaking a programme of clinical studies using the monoclonal antibody C595 (IgG3) which reacts with the MUC1 glycoprotein antigen that is aberrantly expressed in a high proportion of bladder tumours. Radioimmunoconjugates of the C595 antibody have been produced with high radiolabelling efficiency and immunoreactivity using Tc-99m and In-111 for diagnostic imaging, and disease staging and the cytotoxic radionuclides Cu-67 and Re-188 for therapy of superficial bladder cancer. A Phase I/II therapeutic trail involving the intravesical administration of antibody directly into the bladder has now begun.

Production of antisera to synthetic decapeptide of the Cooh-Terminus of rat glut2 BABT
Vestri,Sandro; Nikami,Hideki; Saito,Masayuki; Machado,Ubiratan F..
In the present study, the production of an anti-GLUT 2 antibody is reported. The antibody (S-5096) was raised in New Zealand rabbits immunized with an immunogen prepared by cross-linking the synthetic peptide and a carrier protein. A COOH-terminal decapeptide of rat GLUT 2 predicted sequence was synthesized and coupled to keyhole limpet hemocyanin, using the glutaraldehyde method. Subcellular membrane fractions were prepared from renal cortex (C) and medulla (M), and subjected to Western blotting analysis using the antiserum in a 1:200 dilution and, subsequently, [125I]protein A. As the results showed, the S-5096 anti-serum showed clear blots in kidney samples, stronger in cortex than in medulla as expected, whereas white adipose tissue and heart samples...

Effect of aging and oral tolerance on dendritic cell function BJMBR
Simioni,P.U.; Fernandes,L.G.R.; Gabriel,D.L.; Tamashiro,W.M.S.C..
Oral tolerance can be induced in some mouse strains by gavage or spontaneous ingestion of dietary antigens. In the present study, we determined the influence of aging and oral tolerance on the secretion of co-stimulatory molecules by dendritic cells (DC), and on the ability of DC to induce proliferation and cytokine secretion by naive T cells from BALB/c and OVA transgenic (DO11.10) mice. We observed that oral tolerance could be induced in BALB/c mice (N = 5 in each group) of all ages (8, 20, 40, 60, and 80 weeks old), although a decline in specific antibody levels was observed in the sera of both tolerized and immunized mice with advancing age (40 to 80 weeks old). DC obtained from young, adult and middle-aged (8, 20, and 40 weeks old) tolerized mice were...

Rabies infection and specific effect of vaccination in mice selected for high and low immunobiological parameters BJMBR
Queiroz-da-Silva,L.H.; De-Franco,M.; Sant'Anna,O.A.
Innate and acquired resistance to rabies infection was investigated in mice genetically selected for high (H) or low (L) antibody responsiveness from selections I, III and IV and in mice selected for maximal (AIRmax) or minimal (AIRmin) acute inflammatory reaction. These mouse lines were infected intramuscularly with different virus dilutions and the LD50 was determined. The HIII and HIV mouse lines were more susceptible than the LIII and LIV lines and the HI line showed a discrete but higher resistance than the LI line. Analysis of the interline (H x L) F1 hybrids from selections III and IV indicated different dominance effects on the "resistant" and" susceptible" phenotypes when the route of vaccination was changed. No differences were observed between...

Aging reduces the primary humoral response and the in vitro cytokine production in mice BJMBR
Simioni,P.U.; Costa,E.H.; Tamashiro,W.M.S.C..
Aging is accompanied by a decrease in several physiological functions that make older individuals less responsive to environmental challenges. In the present study, we analyzed the immune response of female BALB/c mice (N = 6) of different ages (from 2 to 96 weeks) and identified significant age-related alterations. Immunization with hapten-protein (trinitrophenyl-bovine serum albumin) conjugates resulted in lower antibody levels in the primary and secondary responses of old mice (72 weeks old). Moreover, young mice (2, 16, and 32 weeks old) maintained specific antibodies in their sera for longer periods after primary immunization than did old mice. However, a secondary challenge efficiently induced memory in old mice, as shown by the increased antibody...

Development of a caspase-3 antibody as a tool for detecting apoptosis in cells from rainbow trout (Oncorhynchus mykiss) Electron. J. Biotechnol.
Rojas,Verónica; Guzmán,Fanny; Valenzuela,Cristián; Marshall,Sergio H; Mercado,Luis.
Background: Apoptosis is an active cell death process mediated by caspases activation, in which different extrinsic or intrinsic signalling pathways result in direct activation of effector caspases. Caspase-3 is considered to be the most important of the executioner caspases, which cause the morphological and biochemical changes detected in apoptotic cells. Different bacterial and virus pathogens have developed different strategies to survive inside the host and overcome natural protections, one of them is inducing apoptotic death in infected cells. We have demonstrated previously that Piscirickettsia salmonis activates this process in monocytes/macrophages from salmonid RTS11 cell line both by morphological and caspase detection assays; nevertheless,...

Vaccines for finfish aquaculture: What do we need to know to make them work? Electron. J. Biotechnol.
Dixon,Brian.
Aquaculture still faces serious economic impacts due to the loss of animals to disease. A conservative estimate of 5% losses due to disease means that the finfish aquaculture industry loses over $1 billion annually on a global scale. One proven way to prevent costly disease outbreaks is to vaccinate fish against common or known pathogens. Current vaccination schemes still result in losses, however, and this may be due in part to vaccine design. Vaccines are currently designed using state of the art knowledge of immune responses, which is based primarily on mammalian studies. Just how applicable is this information to fish immunity and vaccine design, however? This review discusses what is currently known for teleost fish about two key processes that drive...

FISH IMMUNE SYSTEM Gayana
Olabuenaga,Susana E..
The defence against infections is the essential function of the immune system of all Vertebrates. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. It can be divided into two types: innate or non specific and adquired or specific, both with a humoral and cellular compartments. Their relative importance can change according to the age of fish and is always under the influence of different factors. The most relevant components are mentioned here. There are two factors in fishes which strongly influence the immune response and antibody production, seasonal changes and temperature. The main lymphoid organs are described here: the thymus, the kidney and the spleen, as well...

Impact of co-blocking the costimulatory signals on immune-related genes after high-risk rabbit corneal allograft using 2nd-generation DNA sequencing technology Genet. Mol. Biol.
Zhao,Hai-Xia; Li,Xin-Yu; Guan,Wen-Ying; Han,Xiao-Tong.
Abstract The aim of this study was to evaluate the impact and mechanism of co-blocking of costimulatory signals CD28-B7-CD40-CD40L during immune allograft rejection. Forty-eight recipient rabbits were prepared as a high-risk corneal allograft model. After surgery, the animals were randomly divided into: control group, MR1 group, anti-B7 group, and co-blocking group (n=12, each group). Subconjunctival injection was first performed on the allograft surgery day until post-surgery day five. Four weeks later, or when immune rejection occurred, the cornea was sampled to detect and analyze the gene spectrum. The survival time in the co-blocking group was significantly longer than that in the other three groups (p < 0.05). Gene expression analysis revealed that...
