Two α -galactosidase (P1 and P2) and one invertase present in the culture of Aspergillus terreus grown on wheat straw for 168 h at 28ºC were partially purified by gel filtration and hydrophobic interaction chromatographies. Optimum pH and temperatures for P1, P2 and invertase preparations were 4.5-5.0, 5.5 and 4.0 and 60, 55 and 65ºC, respectively. The K M app for Ï -nitrophenyl-α -D-galactopyranoside were 1.32 mM and 0.72 mM for P1 and P2, respectively, while the K M app value for invertase, using sacarose as a substrate was 15.66 mM. Abstract A thermohalophilic fungus, Aspergillus terreus AUMC 10138, isolated from the Wadi El-Natrun soda lakes in northern Egypt was exposed successively to gamma and UV-radiation (physical mutagens) and ethyl methan-sulfonate (EMS; chemical mutagen) to enhance alkaline cellulase production under solid state fermentation (SSF) conditions. Most of the studies regarding cyclosporin 'A' production through fungi concentrate around Tolypocladium inflatum. This is mainly due to lower reported production of this drug in other fungi. The present study was therefore conducted to explore indigenous isolates of Aspergillus terreus for synthesis of this drug and defining a production medium for obtaining high yield of cyclosporin 'A'. For this purpose carbon and nitrogen sources were optimized for the selected best strain of A. terreus. In attempt to immobilize the mycelial fungi on a new hydrophobic matrix, Aspergillus terreus mycelia were immobilized into polytetrafluoroethylen (PTFE) and used for selected conversion of progesterone to 11alpha-hydroxyprogesterone. The immobilized cells exhibited higher yields than the free cells. Different parameters that can affect the cell activity were measured. The optimum pH was 6 and the optimum temperature was 30ºC. Higher transformation activities were achieved with matrix contained 40% PTFE. No considerable loss in yield was observed with storage of the cells at 4ºC for 30 days. Utilization of the immobilized cells in repeated batch processes indicated that the cells retained about 84% of their activity after reuse for eight successive cycles. |
