Background To study the relationship between intracellular anabolism and astaxanthin production, the influence of intracellular protein and fatty acids on astaxanthin production by four mutant Phaffia rhodozyma strains and their variations was investigated in this research. Results First, the content of astaxanthin in cells showed a reverse fluctuation in contrast to that of protein during the whole fermentation process. Moreover, compared with the three other strains, the astaxanthin-overproducing mutant strain of the yeast P. rhodozyma, called JMU-MVP14, had the highest specific productivity of astaxanthin as 6.8 mg/g, whereas its intracellular protein and fatty acid contents were the lowest. Дано обоснование актуальности, стратегии и тактики исследований, направленных на выявление новых коммерчески перспективных видов зелёных микроводорослей для получения природных кетокаротиноидов астаксантина и кантаксантина. Приведена унифицированная схема скрининга, основанная на методе двухстадийной накопительной культуры.

Исследовано влияние концентрации азота и фосфора в среде на скорость роста и накопление астаксантина в вегетативных клетках зелёной микроводоросли Haematococcus pluvialis при выращивании методом одностадийной полупроточной культуры. Показано, что необходимым условием сохранения монадной структуры, деления клеток на протяжении длительного периода и накопления в них астаксантина является поддержание минимального уровня биогенных элементов в среде в пределах 0.2-0.4 мМ N и 0.01-0.02 мМ Р.

The microalga Haematococcus pluvialis Flotow is one of the natural sources of astaxanthin, a pigment widely used in salmon feed. This study was made to discover optimal conditions for biomass and astaxanthin production in H. pluvialis from Steptoe, Nevada (USA), cultured in batch mode. Growth was carried out under autotrophic (with NaNO3, NH4Cl and urea) and mixotrophic conditions (with 4, 8, 12 mM sodium acetate) under two photon flux densities (PFD) (35 and 85 µmol m-2 s-1). The carotenogenesis was induced by 1) addition of NaCl (0.2 and 0.8 %), 2) N-deprivation and 3) high PFD (150 µmol m-2 s-1). Effect of feeding methods on the astaxanthin production by Phaffia rhodozyma in fed-batch process BABT

The effect of feeding methods on the production of astaxanthin by the yeast Phaffia rhodozyma ATCC 24202 was studied, using continuous and pulsed fed-batch processes and low cost materials as substrates (sugar cane juice and urea). In continuous fed-batch processes, a cellular astaxanthin concentration of 383.73 µg/g biomass was obtained. But in pulsed fed-batch processes a reduction in the cellular astaxanthin concentration (303.34 µg/g biomass) was observed. Thus the continuous fed-batch processes could be an alternative to industrial production of astaxanthin, allowing an increase in the biomass productivity without losses on astaxanthin production by the yeast.
