Bacillus subtilis as a tool for vaccine development: from antigen factories to delivery vectors Anais da ABC (AABC)
Ferreira,Luís C.S.; Ferreira,Rita C.C.; Schumann,Wolfgang.
Bacillus subtilis and some of its close relatives have a long history of industrial and biotechnological applications. Search for antigen expression systems based on recombinant B. subtilis strains sounds attractive both by the extensive genetic knowledge and the lack of an outer membrane, which simplify the secretion and purification of heterologous proteins. More recently, genetically modified B. subtilis spores have been described as indestructible delivery vehicles for vaccine antigens. Nonetheless both production and delivery of antigens by B. subtilis strains face some inherent obstacles, as unstable gene expression and reduced immunogenicity that, otherwise, can be overcome by already available gene technology approaches. In the present review we...

Crambe affected by biological and chemical seed treatments Bragantia
Cattanêo,Alex Júnior; Stangarlin,José Renato; Bassegio,Doglas; Santos,Reginado Ferreira.
ABSTRACT Crambe has great potential for cultivation in tropical regions, but its biotic factors may be a limiting aspect for its production. Thus, we aimed in this study to assess the effect of pre‐sowing seed treatments on seedling emergence, root system, production components, and oil. The experiment was conducted during two years in Cascavel, Paraná, Brazil. The experimental design consisted of randomized blocks with six treatments, including a control treatment (no treatment), biological treatments (Trichoderma asperellum and Bacillus subtilis), and chemical treatments (glutamic acid, thiamethoxam and zinc), with four replications. Emergence and growth crambe, as well as production components and oil in the first year were higher due to the contrast in...

Antibiosis and dark-pigments secretion by the phytopathogenic and environmental fungal species after interaction in vitro with a Bacillus subtilis isolate BABT
Machado,Alexandre Paulo; Vivi,Viviane Karolina; Tavares,José Roberto; Gueiros Filho,Frederico José; Fischman,Olga.
In this work, different reactions in vitro between an environmental bacterial isolate and fungal species were related. The Gram-positive bacteria had terminal and subterminal endospores, presented metabolic characteristics of mesophilic and acidophilic growth, halotolerance, positive to nitrate reduction and enzyme production, as caseinase and catalase. The analysis of partial sequences containing 400 to 700 bases of the 16S ribosomal RNA gene showed identity with the genus Bacillus. However, its identity as B. subtilis was confirmed after analyses of the rpoB, gyrA, and 16S rRNA near-full-length sequences. Strong inhibitory activity of environmental microorganisms, such as Penicillium sp, Aspergillus flavus, A. niger, and phytopathogens, such as...

Growth, sporulation and production of bioactive compounds by Bacillus subtilis R14 BABT
Carvalho,André Lacerda Ulysses de; Oliveira,Fábio Henrique Portella Corrêa de; Mariano,Rosa de Lima Ramos; Gouveia,Ester Ribeiro; Souto-Maior,Ana Maria.
The physiology of B. subtilis R14 was investigated in minimal medium under excess-oxygen and oxygen-limited conditions. Growth and efficient sporulation could be achieved in excess-oxygen culture on medium with readily metabolizable carbon and nitrogen sources, which allowed high growth rate and high biomass yield. A short transition phase between the exponential growth and sporulation could be attained by formulating a medium with a well-balanced C/N ratio. Under oxygen-limitation, but in the presence of essential nutrients (i.e. excess-nutrient cultivation), B. subtilis R14 produced bioactive compounds, which showed activity against several phytopathogenic bacteria. Under anaerobic condition, the organism did not grow neither through fermentation nor...

One-step purification and characterization of cellulase-free xylanase produced by alkalophilic Bacillus subtilis ash BJM
Sanghi,Ashwani; Garg,Neelam; Gupta,V.K.; Mittal,Ashwani; Kuhad,R.C..
The present study describes the one-step purification and characterization of an extracellular cellulase-free xylanase from a newly isolated alkalophilic and moderately thermophilic strain of Bacillus subtilis ASH. Xylanase was purified to homogeneity by 10.5-fold with ~43% recovery using ion-exchange chromatography through CM-Sephadex C-50. The purified enzyme revealed a single band on SDS-PAGE gel with a molecular mass of 23 kDa. It showed an optimum pH at 7.0 and was stable over the pH range 6.0-9.0. The optimum temperature for enzyme activity was 55 ºC. The purified xylanase did not lose any activity up to 45 ºC, however, it retained 80% and 51% of its activity after pre-incubation at 55 ºC and 60 ºC, respectively. The enzyme obeyed Michaelis-Menton...

Fusarium culmorum affects expression of biofilm formation key genes in Bacillus subtilis BJM
Khezri,Maryam; Jouzani,Gholamreza Salehi; Ahmadzadeh,Masoud.
Abstract It is known that there is correlation between biofilm formation and antagonistic activities of Bacillus subtilis strains; but, the mechanism of this correlation is not clear. So, the effect of the plant pathogen (Fusarium culmorum) on the biofilm formation in a B. subtilis strain with high antagonistic and biofilm formation activities was studied. The expression of sinR and tasA genes involved in the biofilm formation was studied in both single culture of bacterium (B) and co-culture with F. culmorum (FB) using real-time PCR. The results revealed that the expression of the sinR gene in both B and FB conditions was continuously decreased during the biofilm formation period and, after 24 h (B4 and FB4), it reached 1% and 0.3% at the planktonic phase...

Avaliação da biodegradação de parafinas e da produção de biosurfactante por Bacillus subtilis na presença de petróleo BJM
Queiroga,Carmen Lucia; Nascimento,Lídia Regina; Serra,Gil Eduardo.

Antimicrobial activity of surfactants produced by Bacillus subtilis R14 against multidrug-resistant bacteria BJM
Fernandes,Paulo André Vicente; Arruda,Isabel Renata de; Santos,Antônio Fernando Amatto Botelho dos; Araújo,Ana Albertina de; Maior,Ana Maria Souto; Ximenes,Eulália Azevedo.
Lipopeptides represent a class of microbial surfactants with increasing scientific, therapeutic and biotechnological interests. The genus Bacillus is a producer of these active compounds, and among them B. subtilis produces surfactin, the most potent biosurfactant known. These compounds can act as antibiotics, antivirals, antitumorals, immunomodulators and enzyme inhibitors. In this work, the antimicrobial activity of biosurfactants obtained by cultivation of B. subtilis R14 was investigated against multidrug-resistant bacteria. During cultivation in defined medium, the surface tension of the medium was reduced from 54 mN/m in the beginning of the microbial growth to 30 mN/m after 20 hours. A crude surfactant concentration of 2.0 g/L was obtained after 40...

Lipopeptide production by Bacillus subtilis R1 and its possible applications BJM
Jha,Sujata S.; Joshi,Sanket J.; S.J.,Geetha.
Abstract The possible application of a bacterial strain - Bacillus subtilis R1, isolated from an oil contaminated desert site in India, as biocontrol agent and its biosurfactant in microbial enhanced oil recovery are discussed. The biosurfactant production in minimal medium was carried out at different temperatures and salt concentrations, where it produced an efficient biosurfactant at 30-45 °C and in presence of up to 7% salt. It significantly reduced the surface tension from 66 ± 1.25 mN/m to 29 ± 0.85 mN/m within 24 h. In order to enhance the biosurfactant production, random mutagenesis of B. subtilis R1 was performed using chemical mutagen - ethyl methanesulfonate. Majority of the isolated 42 mutants showed biosurfactant production, but the difference...
