Expression of manganese peroxidase by Lentinula edodes and Lentinula boryana in solid state and submerged system fermentation Anais da ABC (AABC)
HERMANN,KATIA L.; COSTA,ALESSANDRA; HELM,CRISTIANE V.; LIMA,EDSON A. DE; TAVARES,LORENA B.B..
The production of ethanol from lignocellulosic biomass is referred as a second generation biofuel, whose processing is one of the most promising technologies under development. There are few available studies on the use of enzymes produced by fungi as active for the biodegradation of lignocellulosic biomass. However, the manganese peroxidase (MnP) enzyme presents high potential to degrade lignin and the basidiomycetes are the major producers of this oxidase. Thus, this study aimed at evaluating the ability of fungi Lentinula edodes and Lentinula boryana to produce this enzyme when cultivated in submerged fermentation system (SS) and also in solid-state fermentation system (SSF) containing Eucalyptus benthamii sawdust with or without corn cob meal. In the...

Interference of a commercial catalase preparation in laccase and peroxidase activities BABT
Ballaminut,Nara; Yamanaka,Renata; Machado,Katia Maria Gomes.
The influence of commercial catalase preparations (fungal and bovine origin) on laccase and peroxidase activity assays was evaluated using enzymatic extracts obtained from several basidiomycetes grown under different culture conditions. No hydrogen peroxide was detected in the extracts. Inhibition of laccase activity by 40 to 80% was related to the catalase source. In addition, oxidation of the substrate (ABTS) by fungal catalase in the absence of the enzymatic extract from basidiomycetes was observed. The results demonstrated the need for the evaluation of interference of the commercial catalase preparation when its use was required in the reaction mixture.

Halotolerance, ligninase production and herbicide degradation ability of basidiomycetes strains BJM
Arakaki,R.L.; Monteiro,D.A.; Boscolo,M.; Dasilva,R.; Gomes,E..
Fungi have been recently recognized as organisms able to grow in presence of high salt concentration with halophilic and halotolerance properties and their ligninolytic enzyme complex have an unspecific action enabling their use to degradation of a number of xenobiotic compounds. In this work, both the effect of salt and polyols on growth of the basidiomycetes strains, on their ability to produce ligninolytic enzyme and diuron degradation were evaluated. Results showed that the presence of NaCl in the culture medium affected fungal specimens in different ways. Seven out of ten tested strains had growth inhibited by salt while Dacryopinax elegans SXS323, Polyporus sp MCA128 and Datronia stereoides MCA167 fungi exhibited higher biomass production in medium...

Characterization of fungal inoculum used in soil bioremediation BJM
Ballaminut,Nara; Matheus,Dácio R..
Studies have indicated the capacity of basidiomycetes to degrade recalcitrant organopollutants. However, the age of the fungal inoculum to obtain a more effective degradation has not been defined. The criterion used is total colonization of the substrate. Psilocybe castanella CCB444 and Lentinus crinitus CCB274 have been evaluated in soils containing hexachlorobenzene. In the present study, the physiological conditions of the fungal inocula were characterized on solid substrate (sugarcane bagasse, starch and soy flour). Colonization of the substrate, loss of organic matter, pH variation, organic carbon, total nitrogen, fungal biomass and enzymatic activity were evaluated over 30 days of incubation. Colonization of the substrate was almost complete after 20...
