Total mercury in wild felids occurring in protected areas in the central Brazilian Amazon 36
LOPES,Marcelly Castello Branco; CARVALHO,Gabriel Oliveira de; BERNARDO,Robson Roney; MACEDO,Joana; LINO,Adan Santos; RAMALHO,Emiliano Esterci; KASPER,Daniele; MEIRE,Rodrigo Ornellas; TORRES,João Paulo Machado; MALM,Olaf.
ABSTRACT This is the first study to investigate mercury (Hg) contamination in felid species in the Brazilian Amazon. We collected 26 fur samples from wild felids of four species (Puma concolor, Panthera onca, Leopardus pardalis and Leopardus wiedii) occurring in the Mamirauá and Amanã sustainable development reserves, in the state of Amazonas. Samples were from museum specimens, except for five P. onca samples collected from free-living individuals. Total Hg concentrations ranged from 0.12 to 48.1 µg g-1. Concentrations of Hg did not differ significantly between museum specimens and live individuals of P. onca, but varied significantly among species, with significantly higher concentrations for P. onca and L. pardalis, which could be related to factors...

Mercury Levels in Eggs, Embryos, and Neonates of Trachemys callirostris (Testudines, Emydidae) 37
RENDÓN-VALENCIA,BEATRIZ; ZAPATA,LINA M; C. BOCK,BRIAN; PÁEZ,VIVIAN P.; PALACIO,JAIME A..
We quantified total mercury concentrations in eggshells, egg yolks, and embryos from 16 nests of the Colombian Slider (Trachemys callirostris). Nests were collected in different stages of development, but estimated time of incubation in natural substrates was not correlated with mercury levels in the eggs, suggesting that mercury was not absorbed from the substrate, but more likely passed on to the embryos during folliculogenesis by the reproductive females who had bioaccumulated the mercury from environmental sources. Mean mercury concentrations were higher in embryos than in eggshells or egg yolks, indicating that embryos also bioaccumulate mercury present in other egg tissues. Intra-clutch variation in egg yolk mercury concentrations was relatively...

Oxidative Stress, Chromium-Resistance and Uptake by Fungi: Isolated from Industrial Wastewater 52
Elahi,Amina; Rehman,Abdul.
ABSTRACT Trichosporon asahii and Rhodotorula mucilaginosa isolated from wastewater effluents were identified as chromium-resistant yeasts. Cr(VI) concentrations at 8 mM and 6 mM were inhibitory for R. mucilaginosa and T. asahii. Remarkably elevated GSH (69.88 ± 10.01) and GSSG (11.24 ± 0.96) was observed under metal stress in T. asahii as compared to R. mucilaginosa GSH (18.95 ± 3.19) and GSSG (3.7 ± 2.74) mM g-1 8 level. Statistical analysis revealed significantly higher GSH/GSSG ratio in both strains. NPSH (29.84 ± 0.54) level in T. asahii was much higher than in R. mucilaginosa (6.05 ± 0.24). Chromate reductase (ChR) was assayed and its activity was optimum at 50°C (pH 6) in T. asahii while R. mucilaginosa showed higher activity at 30°C (pH 7). Activity...

Abstract: The Paranhana River, located in Southern Brazil, is one of the major tributaries of the Sinos River basin and receives mainly industrial and domestic effluents. In the present study, water physicochemical and microbiological analyses, condition factor, micronucleus test, gill histopathology and metal bioaccumulation in the muscle of the native fish Bryconamericus iheringii collected at two sites (S1 and S2) of the Paranhana River under different degrees of anthropogenic pressures were assessed in four sampling campaigns. Data from water quality parameters, condition factor, mucous cells proliferation in fish gills and bioaccumulation of chromium and manganese in muscle evidenced higher impacts at S2, whereas a higher genotoxic potential was...

Arsenic tolerance of Microcystis novacekii (Komárek-Compère, 1974) and its arsenic decontamination potential 52
Ferreira,Fernanda Aires Guedes; Franco,Maione Wittig; Oliveira,Dirce Maria De; Magalhães,Sérgia Maria Starling; Barbosa,Francisco Antônio Rodrigues.
ABSTRACT Cyanobacteria possess metallic ion interaction properties that should be explored with the aim of recovering arsenic (As) contaminated areas. Contamination of As is an issue of worldwide concern due to the risk of human chronic intoxication and negative environmental health effects. In this study the potential decontamination of As(III) and As(V) using cyanobacteria cultures was assessed. Microcystis novacekii (Komárek-Compere, 1974) showed normal growth in concentrations of As(V) similar to those found in natural environments contaminated with As, demonstrating its resistance to As(V). Growth rates gradually decreased upon exposure to high As(V) concentrations from 600 to 5630 mg.L-1 while As(III) affected growth from 14.7 - 85.7 mg.L-1. The...

Bioaccumulation of silver and the isolation of metal-binding protein from P.diminuta 52
Ibrahim,Zaharah; Ahmad,Wan Azlina; Baba,Abu Bakar.
A silver uptake study by Pseudomonas diminuta was carried out by growing the bacteria in a chloride-free medium (CFM) containing silver ions (50 muM) in a batch culture. From the results, it was found that higher amounts of silver were accumulated inside the cell during early exponential phase compared to the amount bound at the cell surface. This suggested a possible mechanism for metal uptake during bacterial growth. In view of this, attempts were made to isolate proteins which might be associated with silver-binding properties from cultures of P.diminuta grown in the presence and absence of silver. The proteins were first extracted from the bacterial cultures by precipitation with ammonium sulfate followed by purification using isoelectric focussing and...

Uptake of free and complexed silver ions by different strains of Rhodotorula mucilaginosa 58
Gomes,Newton C.M.; Rosa,Carlos A.; Pimentel,Patrícia F.; Mendonça-Hagler,Leda C.S..
Five strains of Rhodotorula mucilaginosa were tested for the ability to accumulate free and complexed silver ions by metabolism-dependent and -independent processes. The ability to take up Ag+ was observed in both live and dead biomass, whereas silver dicyanide [Ag (CN)2-] uptake was strictly glucose dependent. In contrast to Ag (CN)2-, glucose addition inhibited by 16 to 25% the Ag+ uptake rate of living UFMG - Y02, Y27, and Y35 cells, while strains CBS 316 and UFMG-Y01 showed an improved uptake rate of about 115% and 13%, respectively. The Langmuir sorption model was used to evaluate the silver sorption capability of the R. mucilaginosa strains. The calculated q max value suggested that R. mucilaginosa strains UFMG-Y27 had the highest loading capacity....

Detection of poliovirus type 2 in oysters by using cell culture and RT-PCR 58
Vinatea,Cecília E.B.; Sincero,Thaís C.M.; Simões,Claúdia M.O.; Barardi,Célia R.M..
Shellfish are readily contaminated with viruses present in water containing sewage due to the concentrating effect of filter feeding. Enteroviruses are generally used as a model for the detection of viruses from shellfish due to their public health significance. In the present work, oysters were placed in glass aquaria containing seawater plus unicellular algae. Two experiments were performed: 1) oysters bioaccumulating four different poliovirus type 2 concentrations: 5 x 10(4), 2.5 x 10(4), 5 x 10³ and 5 x 10² PFU/mL during 20h; 2) oyster tissues directly inoculated with 6.0 x 10(5) and 1.0 x 10(5) PFU/mL. After viruses seeding, tissue samples were processed by an adsorption-elution-precipitation method. Positive controls were performed by seeding 6.0 x...

Selenium biofortification on garlic growth and other nutrients accumulation 75
Pérez,María B; Lipinski,Víctor Mario; Filippini,María Flavia; Chacón-Madrid,Katherine; Arruda,Marco Aurelio Z; Wuilloud,Rodolfo G.
ABSTRACT Selenium (Se) is an essential element for humans and has anti-cancer function. Garlic can accumulate Se, so it is an option to Se supplementation in the human diet. The aim of this research was to study Se uptake and accumulation during garlic growth. Four doses of Na2SeO4 and Na2SeO3 solution were applied in the substrate (0, 5, 10 and 15 kg ha-1 Se) for one time in August 2014, with a random plot design and 3 replicates on garlic clone Rubi INTA. Three harvests were made, in September, October and December 2014. After each harvest, leaves, bulbs and roots were separated and conditioned (peeled and chopped), lyophilized, and finally acid-digested prior to Se, Mg, Zn, Mn, Cu, Fe, P and S determination by inductively coupled plasma mass...

Variation of heavy metal concentrations (Ag, Cd, Co, Cu, Fe, Pb, V, and Zn) during the life cycle of the common cuttlefish Sepia officinalis 5
Miramand, Pierre; Bustamante, Paco; Bentley, Daniel; Koueta, Noussithe.
The developmental changes in the concentration of 8 essential and non-essential heavy metals (Ag, Cd, Cu, Co, Fe, Pb, V, Zn) in the tissues (digestive gland, cuttlebone and whole animal) of the common cuttlefish Sepia officinalis collected in the bay of the river Seine were monitored from the end of the embryogenesis until the adult reproductive stage. Compared to embryos, juveniles after hatching displayed much higher concentrations of Ag, Cu, Fe and Zn, suggesting an efficient incorporation from seawater. Conversely, the amounts of Cd, Pb and V in hatchlings remained constant suggesting that these metals are barely bioavailable for juveniles. Once the juveniles start to feed, the digestive gland appears to play a major role in the storage of all metals....

Past and present platinum contamination of a major European fluvial-estuarine system: Insights from river sediments and estuarine oysters 5
Abdou, M.; Schaefer, J.; Cobelo-garcia, A.; Neira, P.; Petit, J. C. J.; Auger, Dominique; Chiffoleau, Jean-francois; Blanc, Gerard.
Platinum Group Metals (PGM) are modern, technologically relevant elements for which (i) the anthropogenic cycle has outcompeted the natural cycles and (ii) environmental behavior, fate and impact are still widely unknown. Stripping voltammetry was used for accurate determinations of platinum (Pt) in historical records of river sediments and estuarine oysters from the Gironde fluvial-estuarine continuum (SW France) comprising the Lot River. Sediment cores from the Lot River, dated from 1952 to 2001, showed past Pt contamination due to former industrial (smelter) activities in the Lot River watershed. These samples revealed the phasing-out of a historical Pt contamination with Pt/Th (Thorium) values of 11 × 10− 5 ± 0.79 × 10− 5 for the deepest part of the...

Oligotrophy as a major driver of mercury bioaccumulation in medium-to high-trophic level consumers: A marine ecosystem-comparative study 5
Chouvelon, Tiphaine; Cresson, Pierre; Bouchoucha, Marc; Brach-papa, Christophe; Bustamante, Paco; Crochet, Sylvette; Marco-miralles, Francoise; Thomas, Bastien; Knoery, Joel.
Mercury (Hg) is a global contaminant of environmental concern. Numerous factors influencing its bioaccumulation in marine organisms have already been described at both individual and species levels (e.g., size or age, habitat, trophic level). However, few studies have compared the trophic characteristics of ecosystems to explain underlying mechanisms of differences in Hg bioaccumulation and biomagnification among food webs and systems. The present study aimed at investigating the potential primary role of the trophic status of systems on Hg bioaccumulation and biomagnification in temperate marine food webs, as shown by their medium-to high-trophic level consumers. It used data from samples collected at the shelf-edge (i.e. offshore organisms) in two...

Biological availability of sediment-bound trace metals. 5
Luoma, S.
It has been established that direct uptake of (at least some) trace metals from sediments contributes to metal concentrations in benthic organisms. Important controls on metal uptake from sediment are the concentration of the exposure, the partitioning of the metals among components of the sediment, and the redox potential of the sediment. Important problems in ecology, physiology, geochemistry and biogeochemistry remain unresolved, however, and a concentrated interdisciplinary research effort will be necessary before a complete understanding of the fate and impact of sediment-bound metals will be possible.
