Retrospective evaluation of the clinical characteristics associated with Corynebacterium species bacteremia BJID
Yanai,Mitsuru; Ogasawasa,Maiko; Hayashi,Yuta; Suzuki,Kiyozumi; Takahashi,Hiromichi; Satomura,Atsushi.
ABSTRACT Objectives: Corynebacterium spp. are becoming recognized as pathogens that potentially cause various infections. We aimed to evaluate the clinical characteristics associated with Corynebacterium spp. bacteremia. Patients and methods: We retrospectively reviewed the medical records of all adult patients who had positive blood cultures for Corynebacterium spp. in a single university hospital between January 2014 and December 2016. Patients were divided into a bacteremia group and a contamination group based on microbiological test results and clinical characteristics. Patients’ characteristics, antimicrobial susceptibility of isolated species, antimicrobials administered, and patient outcomes were evaluated. Results: Corynebacterium spp. were...

Community acquired pneumonia among pediatric outpatients in Salvador, Northeast Brazil, with emphasis on the role of pneumococcus BJID
Nascimento-Carvalho,Cristiana M.C.; Lopes,Antônio A.; Gomes,Maria Daniela B.S.; Magalhães,Morgana P.; Oliveira,Juliana R.; Vilas-Boas,Ana Luisa; Ferracuti,Roberta; Brandileone,Maria Cristina C.; Guerra,Maria Luiza L.S.; Alves,Noraney N.; Athayde,Ledilce A.; Caldas,Renilza M.; Barberino,Maria Goreth M. Pneumonia is one of the leading causes of hospitalization and death among children in developing countries, and mortality due to pneumonia has been associated with S. pneumoniae infection. This investigation was designed to describe the antimicrobial susceptibility and serotype patterns of pneumococcal strains recovered from the blood of children with community-acquired pneumonia (CAP) and to assess the clinical findings of pneumococcal bacteremic patients with pneumonia. In a 26 month prospective study, blood cultures were obtained as often as possible from children (<16 years of age) diagnosed with CAP in two emergency rooms. Antimicrobial drug susceptibility tests and serotyping were performed when pneumococcus was identified. We studied 3,431 cases...

Direct identification from positive blood broth culture by matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF MS) BJID
Barberino,Maria Goreth; Silva,Marcio de Oliveira; Arraes,Ana Carolina Palmeiras; Correia,Luís Cláudio; Mendes,Ana Verena.
ABSTRACT Bloodstream infections (BSIs) are among the most concerning bacterial infections. They are one of the leading causes of morbidity and mortality, and occur in 30-70% of critical care patients. The prompt identification of the causative microorganism can help choosing the appropriate antimicrobial therapy that will lead to better clinical outcomes. Blood culture is one of the most relevant tests for microbiological diagnosis of bacterial infections. The introduction of the MALDI-TOF microbiological diagnosis significantly decreased the time of identifying microorganisms. However, it depends on the growth on solid culture medium. In this study, 538 bottles of positive blood cultures were evaluated to test the accuracy of an in house modified...

Polymerase chain reaction and blood culture in blood donors screened by ELISA test for Chagas' disease BJPS
Kinoshita-Yanaga,Andréa Tieko; Bértoli,Marta; Martins,Vanessa Aparecida; Mizutani,Angélica Sayuri; Toledo,Max Jean de Ornelas; Araújo,Silvana Marques de; Gomes,Mônica Lúcia.
The objective of this study was to evaluate, through blood culture and PCR, the results of the ELISA for Chagas' disease in the screening of blood donors in the public blood-supply network of the state of Paraná, Brazil, and to map the epidemiological profile of the donors with respect to their risk of infection by Trypanosoma cruzi. The negative and positive results of the ELISA were confirmed by blood culture and PCR for 190/191 individuals (99.5%). For one individual (0.5%), the ELISA was inconclusive, blood culture and IIF were negative, and IHA and PCR positive. Three individuals (1.6%) were positive for T. cruzi on all the tests. Donors were predominantly female, and natives of Paraná, of rural origin, had observed or been informed of the presence of...

Polymerase Chain Reaction and blood culture for diagnosis of canine sepsis Ciência Rural
Silveira,Marcelo Marques da; Cândido,Stéfhano Luis; Santos,Karin Rinaldi dos; Maia,Maerle Oliveira; Souza,Roberto Lopes de; Sousa,Valéria Régia Franco; Almeida,Arleana do Bom Parto Ferreira de; Dutra,Valeria; Nakazato,Luciano.
ABSTRACT: Sepsis is characterized by the presence of organ dysfunction secondary to the dysregulated systemic inflammatory response associated with an infection, and has high mortality rates. Traditional diagnostic techniques based on non-microbiological isolation are time-consuming and may delay treatment. Thus, this study aimed to compare bacterial and fungal broad-range polymerase chain reaction (PCR) and blood culture for diagnosis of sepsis in dogs. Blood samples from 88 dogs with suspected sepsis were analyzed by blood culture, and PCR to detect bacterial and fungal DNA. On blood culture, 20 (22.7%) samples tested positive for bacterial isolates; however, none tested positive for fungi. Through PCR analysis, bacterial DNA was detected in 46 (52.3%)...

Trypanosomatids in dogs belonging to individuals with chronic Chagas’ disease living in Botucatu town and surrounding region, São Paulo State, Brazil J. Venom. Anim. Toxins incl. Trop. Dis.
Lucheis,S. B.; Da Silva,A. V.; Araújo Jr,J. P.; Langoni,H.; Meira,D. A.; Marcondes-Machado,J..
Among domestic animals, dogs are considered to be the major reservoirs of trypanosomatids and, due to their proximity to man, the presence of these parasites in dogs is an alert to actions aiming at triatomine control. Fifty dogs (26 males and 24 females), aged from 2 months to 15 years, belonging to 30 chronic Chagas’ disease individuals from 15 different municipalities in the western region of São Paulo State, Brazil, were subjected to blood collection for the following tests: artificial xenodiagnosis, blood culture, and Polymerase Chain Reaction (PCR). Forty-three (86%) out of 50 dogs were positive to at least one of the tests performed; 34 (68%) were positive to xenodiagnosis, 30 (60%) to blood culture, and 25 (50%) to PCR for T. cruzi and/or T....

Role of the concentration process in the recovery of Candida albicans from blood Rev. Microbiol.
Freitas,Ana Lúcia Peixoto de; Barth,Afonso Luis.
The lysis-centrifugation system (Isolator TM) is recognized as the standard method for recovery of Candida spp. from blood. In this study, the effect of the concentration process of this system was compared with conventional methods of blood culture using liquid and biphasic media. The tests were performed in vitro using Candida albicans in counts of 100, 10, 1, and 0.5 cells/ml of blood. Cultures onto chocolate agar were performed with the sediment obtained after centrifugation of IsolatorTM tube and with liquid and biphasic media after their incubation for 24 h at 35oC. Gram stain prepared from the conventional methods were also evaluated in the first 24 h. It was possible to detect Candida albicans in blood, regardless both the number of cells or...
