Yeast lipases: enzyme purification, biochemical properties and gene cloning Electron. J. Biotechnol.
Vakhlu,Jyoti.
Lipases are placed only after proteases and carbohydrases in world enzyme market and share about 5% of enzyme market. They occur in plants, animals and microorganisms and are accordingly classified as plant, animal and microbial lipases. Wherever they exist, they function to catalyze hydrolysis of triglycerides to glycerol and fatty acid. Like carbohydrases and proteases, lipases of microbial origin enjoy greater industrial importance as they are more stable (compared to plant and animal lipases) and can be obtained in bulk at low cost. Majority of yeast lipases are extracelluar, monomericglycoproteins with molecular weight ranging between ~33 to ~65 kD. More than 50% reported lipases producing yeast, produce it in the forms of various isozymes. These...

The exopolysaccharides biosynthesis by Candida yeast depends on carbon sources Electron. J. Biotechnol.
Gientka,Iwona; Bzducha-Wróbel,Anna; Stasiak-Rózañska,Lidia; Agnieszka Bednarska,Aleksandra; Blazejak,Stanislaw.
Background: The exopolysaccharides (EPS) produced by yeast exhibit physico-chemical and rheological properties, which are useful in the production of food and in the cosmetic and pharmaceutical industries as well. The effect was investigated of selected carbon sources on the biosynthesis of EPS by Candida famata and Candida guilliermondii strains originally isolated from kefirs. Results: The biomass yields were dependent on carbon source (sucrose, maltose, lactose, glycerol, sorbitol) and ranged from 4.13 to 7.15 g/L. The highest biomass yield was reported for C. guilliermondii after cultivation on maltose. The maximum specific productivity of EPS during cultivation on maltose was 0.505 and 0.321 for C. guilliermondii and C. famata, respectively. The...

Production and stability studies of the bioemulsifier obtained from a new strain of Candida glabrata UCP 1002 Electron. J. Biotechnol.
Asfora Sarubbo,Leonie; Moura de Luna,Juliana; de Campos-Takaki,Galba Maria.
Evaluation of both tenso-active and emulsifying activities indicated that a biosurfactant was produced by the newly isolated and promising strain Candida glabrata isolated from mangrove sediments. The extracellular water-soluble emulsifying agent was isolated and identified as a heteropolymer. The maximum of bioemulsifier production was observed when the strain was grown on soluble and insoluble substrates cotton seed oil plus glucose, reaching values of 10.0 g/l after 144 hrs at 200 rpm. The cell-free culture broth containing the examined agent lowered the surface tension of the medium to 31 mN/m. Stable and compact emulsions with emulsifying activity of 75% of cotton seed oil were detected. The emulsification capacity remained practically unaltered...

SFORCIN,J. M.; FERNANDES JÚNIOR,A.; LOPES,C. A. M.; FUNARI,S. R. C.; BANKOVA,V..
Recently, propolis has been attracting the attention of researchers due to various biological activities and therapeutic properties. In Brazil, propolis is produced all year long, so there may be some seasonal variations. This work was carried out in order to compare propolis collected during the four seasons by its in vitro antimicrobial activity on yeast pathogens isolated from human infections. Propolis was produced by africanized honeybees in Botucatu, São Paulo State, collected throughout a year and pooled by season. Hydroalcoholic solutions of propolis were prepared with each pool and diluted in agar, using serial concentrations of propolis from each pool. A determination of minimal inhibitory concentration (MIC) was performed. The results show that...

PINTO,ANA CAROLINA C.; ROCHA,DEBORA A.S.; MORAES,DANIEL C. DE; JUNQUEIRA,MARIA L.; FERREIRA-PEREIRA,ANTONIO.
Abstract: Candida albicans is the most frequent fungal species that causes infections in humans. Fluconazole is the main antifungal used to treat Candida infections, and its prolonged and indiscriminate use for the last decades are the most established causes which originated resistant strains. Fungal drug resistance is associated to alterations in ERG11 gene and overexpression of multidrug resistance (MDR) transporters belonging to two families: ATP-binding cassette (ABC) and Major Facilitator Superfamily (MFS). To evaluate the role of MFS transporters in azoles resistance of C. albicans clinical strains, this study aimed to analyze four Candida albicans clinical isolates from the University Hospital in Juiz de Fora (Minas Gerais/Brazil), selected in our...

Biofilms: microbes and disease BJID
Aparna,Madhu Sharma; Yadav,Sarita.
Bacteria that attach to surface aggregate in a hydrated polymeric matrix of their own synthesis to form biofilms. These represent microbial societies with their own defense and communication system. Transitioning from acute to chronic infection is frequently associated with biofilm formation.Bacteria in biofilms are innately more resistant to antimicrobial agents. The presence of indwelling medical devices increases the risk for biofilm formation and subsequent infection. The current antibiotic therapies are of limited effectiveness in resolving biofilms infection. This review attempts to discuss the stages in biofilm formation, their pathogenic mechanisms, effect of antimicrobial agents, detection and eradication of the biofilms.

Emergence of resistant Candida in neutropenic patients BJID
Nucci,Marcio; Colombo,Arnaldo L..
Problems with resistance to antifungal drugs have emerged due to an increase in the incidence of systemic fungal infections and widespread use of antifungal agents. Accordingly, efforts have been made to develop adequate fungal susceptibility tests. The ideal test should have high intra and inter-laboratory reproducibility, good correlation with the clinical outcome, and should be easy to perform. While no such test has yet been developed, advances have been made. Over the past decade, many reports of fungal resistance have been published, most of them in AIDS patients. Though the frequency of resistant strains is still low in neutropenic cancer patients, and is mostly limited to Candida glabrata and Candida krusei, resistance to Candida albicans has also...

Successive mycological nail tests for onychomycosis: a strategy to improve diagnosis efficiency BJID
Meireles,Tereza Elizabeth Fernandes; Rocha,Marcos Fábio Gadelha; Brilhante,Raimunda Sâmia Nogueira; Cordeiro,Rossana de Aguiar; Sidrim,José Júlio Costa.
Onychomycosis is a fungal infection of nails caused by dermatophytes, yeasts and moulds, accounting for about 50% of onychopathies. A high frequency of onychomycosis caused by Candida species has been reported during the last few years in northeast Brazil, as well as in other regions of the world. A clinical diagnosis of onychomycosis needs to be confirmed through laboratory exams. We evaluated the importance of serial repetition of direct microscopic exams and fungal culture for the diagnosis of onychomycosis in the city of Fortaleza, Ceará, in northeast Brazil. We first made a retrospective study of 127 patients with onychomycosis, identifying the fungi that had been isolated from fingernails and toenails. We then made a prospective study of 120...
