ABSTRACTCattleya intermediais an Atlantic Forest species endemic to Brazil that is classed as vulnerable on the list of threatened species. In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. In all treatments 100% plantlet survival was achieved. The integrated analysis of height of aerial part, number of leaves per plantlet, fresh mass, number of roots per plantlet and length of the longest root showed that the plantlets exhibited... |

Mimosa tenuiflora (Mimosaceae) or “jurema-preta” is well distributed in the northeast Brazil, being popularly used to treat skin lesions, burns and inflammation. The healing effect of the alcoholic extract prepared with its barks corroborates the popular use. This study aimed to evaluate the inflammatory response of polysaccharides extracted from M. tenuiflora barks (EP-Mt) by methanol/NaOH and ethanol precipitation. Inflammatory activity was assessed in rat models of acute inflammation (paw edema and peritonitis), by the following parameters: edema, vascular permeability, leukocyte migration, myeloperoxidase activity and pharmacological modulation of nitric oxide and prostaglandins. EP-Mt presented 3.8% yield, 41% carbohydrate and 0.34% protein. EP-Mt... |

Abstract Water deficit is a major factor limiting crop yield in rainfed areas. We hypothesized that under water deficit the decrease of photosynthetic production stimulates: carbohydrate remobilization from leaves, stems and roots to reproductive organs; and decreasing flowering intensity and pod development. The present work aims to study the effect of water deficit during bloom and grain pod-filling stages in two indeterminate soybean cultivar, Vtop and Nidera. The following physiological parameters were evaluated by means of daily CO2 assimilation rate (Ai), dynamic of carbohydrates in tissues, plant growth, grain yield and yield components. The study was conducted in a greenhouse with plants sown in tanks of 0.5 m3. Regardless of the phenological... |

Sperm-surface glycopeptides were obtained from intact sperm membranes after proteolytic release by different enzymatic treatments such as autoproteolysis, trypsin, papain and pronase. Glycopeptides were isolated, their properties and composition were examined, and their monosaccharide and amino acid constituents were characterized. The monosaccharides identified were fucose, mannose, galactose, N-acetylglucosamine, and N-acetylgalactosamine, which form part of more than one type of oligosaccharide units. Autoproteolytic treatment mainly provided O-glycosidic type oligosaccharides, while a mixture of O- and N-glycosidic oligosaccharides was obtained in variable proportions when treated with trypsin, papain or pronase. The highest degree of peptide cleavage... |

Bryothamnion seaforthii, a red alga common to the Northeastern coast of Brazil, was used to prepare the protein fraction F0/60 by ammonium sulfate precipitation. The chromatography of F0/60 on DEAE-Sephadel column resulted in two lectin fractions, PI and PII, which have antinociceptive properties in rodents. We determined the antinociceptive activity of the PII fraction and of a carbohydrate-containing fraction (CF) in mice. The CF was prepared from the dried algae, after digestion with 100 mM sodium acetate, pH 6.0, containing 5 mM cysteine, EDTA and 0.4% papain, at 60ºC. A 10% cetylpyridinium chloride was added to the filtrate, and the precipitate was dissolved with 2 M NaCl:ethanol (100:15, v/v) followed by the carbohydrate precipitation with ethanol.... |

The cell surface carbohydrates of four strains of Aeromonas caviae were analyzed by agglutination and lectin-binding assays employing twenty highly purified lectins encompassing all sugar specificities. With the exception of L-fucose and sialic acid, the sugar residues were detected in A. caviae strains. A marked difference, however, in the pattern of cell surface carbohydrates in different A. caviae isolates was observed. Specific receptors for Tritricum vulgaris (WGA), Lycopersicon esculentum (LEL) and Solanum tuberosum (STA) (D-GlcNAc-binding lectins) were found only in ATCC 15468 strain, whereas Euonymus europaeus (EEL, D-Gal-binding lectin) sites were present exclusively in AeQ32 strain, those for Helix pomatia (HPA, D-GalNAc-binding lectin) in AeC398... |

Citrus plants were grown under two thermal conditions for evaluating carbon metabolism acclimation to moderate warm temperature (30/20ºC, day/night), and its likely impact on plant growth. As reference, plants were grown at 25/20ºC, in which they were subjected to optimum temperature for photosynthesis during the diurnal period (25ºC). Higher photosynthetic rates were found at 30/20ºC as compared to 25/20ºC in both mature and young leaves, being this response associated with higher stomatal conductance. After 30 days of thermal treatment, plants grown at 30/20ºC presented higher shoot growth as compared to those at 25/20ºC. The carbohydrate concentration decreased in stem and root tissues, while it increased in leaf tissues under moderate warm conditions.... |

In general, tropical soils present low concentrations of zinc (Zn), and the deficiency of Zn is recognized as a world nutritional problem for cereal production and human beings. Therefore, the main goal of this study was to assess the effects of different methods of Zn application on the quality of corn and sorghum grains grown in Oxisol. Two experiments were set up in the experimental area of UNESP (campus of Jaboticabal, Brazil). The following nine treatments were applied: three doses of Zn by banded application (seed furrows), three doses of Zn by incorporation into soil (0-20 cm depth), foliar application, seed application, and control (no Zn applied). The treatments were arranged in randomized blocks with four replicates. The contents of Zn,... |

Abstract Hydrocolloids of Pereskia aculeata Miller (OPNH) are potential ingredients in food industry as emulsifiers and stabilizers. The extraction process of OPNH requires the removal of pigments with activated carbon. Because this step is critical to the quality of the ingredient and has an impact on costs, a new activated carbon has been developed with residues from the same process. Residues activated with NaOH and H3PO4 (300 °C, 1 h) were subjected to batch adsorption tests in model solutions of malachite green (MG), carbohydrate and protein. Residue treated with 85% H3PO4 (OPNAC) had higher productivity and MG adsorption capacity, displaying a predominantly microporous surface (MEV/BET) with chemical activation confirmed by TG/FTIR. OPNAC showed... |

The soluble and insoluble cotyledon (SPF-Co and IPF-Co) and tegument (SPF-Te and IPF-Te) cell wall polymer fractions of common beans (Phaseolus vulgaris) were isolated using a chemical-enzymatic method. The sugar composition showed that SPF-Co was constituted of 38.6% arabinose, 23.4% uronic acids, 12.7% galactose, 11.2% xylose, 6.4% mannose and 6.1% glucose, probably derived from slightly branched and weakly bound polymers. The IPF-Co was fractionated with chelating agent (CDTA) and with increasing concentrations of NaOH. The bulk of the cell wall polymers (29.4%) were extracted with 4.0M NaOH and this fraction contained mainly arabinose (55.0%), uronic acid (18.9%), glucose (10.7%), xylose (10.3%) and galactose (3.4%). About 8.7% and 10.6% of the... |

To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Reducing sugars assays are quantitative, sensitive and almost usable with all kind of polysaccharide but some compounds may interfere with them. When polysaccharide is... |

Estimates of carbohydrate assimilation within the mesozooplankton compartment are a fundamental concern in oceanography because they give a general expression of the biologically-regulated incorporation of metabolic fuel produced from inorganic nutrients by phytoplankton in the sea. A new tool is proposed for the assessment of such processes. It is based on the measurement of the activity of an enzyme, pyruvate kinase (PK), on samples of the whole mesozooplankton community. Here, the results are presented of an early attempt at such an implementation in the Bay of Biscay (north-east Atlantic) in springtime. A main area situated in front of an estuary (Gironde) has been extensively studied-the same sampling grid was covered on three occasions (two 20 day... |
