Integrin-linked kinase is involved in lactoferrin-induced anchorage-independent cell growth and survival in PC12 cells OAK
Ishii, Toshiaki; Uto, Tomoka; Mori, Kaori; Fujikawa, Ryu; 石井, 利明.
Aims: Bovine lactoferrin (bLf) causes anchorage-independent cell growth in PC12 cells. The present study investigated the mechanisms involved in bLf-induced anchorage-independent cell growth and survival in PC12 cells. Main methods: The number of adherent cells and suspended cells was estimated separately by using a methyl thiazol tetrazolium (MTT) assay, and the sum of both optical density (O.D.) (570 nm) values was used as a measure of the total number of cells. Key findings: Integrin-linked kinase (ILK) plays an important role in integrin and growth factor signaling pathways. Stable transfection of PC12 cells with a dominant negative kinase-deficient mutant of ILK (DN-ILK) inhibited bLf-induced anchorage-independent cell growth. The ILK activity in the...

Stochastic modeling for a better approach of the in vitro observed growth of colon adenocarcinoma cells BABT
Ruiz-Gómez,Miguel J.; Ruiz-Gómez,Antonio; Martínez Morillo,Manuel.
The definition of a stochastic model that reflects the cell growth and the use of computer software could be very useful in modelling the cell behaviour due to the possibility to introduce alterations in biology parameters to obtain different growth patterns without the use of laboratory material. Human colon adenocarcinoma cells were cultured and a growth curve was made by daily count of the cell number. Pielou modelling was applied for stochastic simulation of deterministic growth, making stochastic the cell division, the death rates and the transition time between division and death, by using different probabilities. A greater growth was produced when the cell division rate increased, considering the density dependence constant. In contrast, a lower...

LncRNA SNHG12 promotes cell growth and inhibits cell apoptosis in colorectal cancer cells BJMBR
Wang,J.Z.; Xu,C.L.; Wu,H.; Shen,S.J..
Several long non-coding RNA (lncRNA) might be correlated with the prognosis of colorectal cancer (CRC) and serve as a diagnostic and prognostic biomarker. However, the exact expression pattern of small nucleolar RNA host gene 12 (SNHG12) in colorectal cancer and its clinical significance remains unclear. The level of SNHG12 was detected by qRT-PCR in CRC tissues and CRC cells. MTT assay and colony formation assay were performed to examine the cell proliferation of CRC cells transfected with pcDNA-SNHG12 or si-SNHG12. Flow cytometry technology was used to detect cell cycle and cell apoptosis of CRC cells transfected with pcDNA-SNHG12 or si-SNHG12. The protein level of cell cycle progression-related molecules, including cyclin-dependent kinases (CDK4, CDK6),...

Effect of hyperoxia on the intestinal IgA secretory component in neonatal rats and on intestinal epithelial cells in vitro BJMBR
Li,J.J..
Oxygen therapy is essential for the treatment of some neonatal critical care conditions but its extrapulmonary effects have not been adequately investigated. We therefore studied the effects of various oxygen concentrations on intestinal epithelial cell function. In order to assess the effects of hyperoxia on the intestinal immunological barrier, we studied two physiological changes in neonatal rats exposed to hyperoxia: the change in intestinal IgA secretory component (SC, an important component of SIgA) and changes in intestinal epithelial cells. Immunohistochemistry and Western blot were used to detect changes in the intestinal tissue SC of neonatal rats. To detect intestinal epithelial cell growth, cells were counted, and...

Dietary sodium intake induced myenteric neuron hypertrophy in Wistar rats BJMBR
De Souza,R.R.; Gama,E.F.; Silva,R.D.A.; Heimann,J.C.; Maifrino,L.B.M.; Liberti,E.A..
In the present study we investigated the effect of salt intake on myenteric neuron size of the colon of adult male Wistar rats. The animals were placed on either a high-salt (HS; 8%; 12 animals) or a low-salt diet (LS; 0.15%; 12 animals) for 15 or 52 weeks and blood pressure was measured. The sizes of myenteric neurons of the distal colon from both groups were measured. No difference in neuron size was observed between the HS and LS groups after 15 weeks. After 52 weeks on HS, neuron size was increased (P<0.005) when compared with the LS group. The rats also presented hypertension, which was significantly different at 52 weeks (142 ± 11 vs 119 ± 7 mmHg). These results suggest that a long time on an HS diet can significantly increase myenteric nerve...
