Mesenchymal stem cell therapy promotes the improvement and recovery of renal function in a preclinical model Genet. Mol. Biol.
Urt-Filho,Antônio; Oliveira,Rodrigo Juliano; Hermeto,Larissa Correa; Pesarini,João Renato; David,Natan de; Cantero,Wilson de Barros; Falcão,Gustavo; Marks,Guido; Antoniolli-Silva,Andréia Conceição Milan Brochado.
Abstract Acute renal failure (ARF) is an extremely important public health issue in need of novel therapies. The present study aimed to evaluate the capacity of mesenchymal stem cell (MSC) therapy to promote the improvement and recovery of renal function in a preclinical model. Wistar rats were used as the experimental model, and our results show that cisplatin (5mg/kg) can efficiently induce ARF, as measured by changes in biochemical (urea and creatinine) and histological parameters. MSC therapy performed 24h after the administration of chemotherapy resulted in normalized plasma urea and creatinine levels 30 and 45d after the onset of kidney disease. Furthermore, MSC therapy significantly reduced histological changes (intratubular cast formation in...

The cytotoxic and growth inhibitory effects of palladium(II) complexes on MDA-MB-435 cells Genet. Mol. Biol.
Campanella,Nathália Cristina; Demartini,Mariana da Silva; Torres,Claudia; Almeida,Eduardo Tonon de; Gouvêa,Cibele Marli Cação Paiva.
The antitumorigenic potential of two palladium(II) complexes, [Pd(ca2-o-phen)Cl2 ] - C1 and [Pd(dmba)(dppp)Cl] - C2, was evaluated, using MDA-MB-435 cells, a human breast adenocarcinoma cell-line that does not express the estrogen receptor α (ER-). Growth inhibition and induced alterations in cell-morphology were analyzed. The sulforhodamine B test showed that, compared to control cells, both C1 and C2 significantly inhibited (p < 0.5) cell growth. The maximum effect with both was achieved with 1 µM complexes, after 24 h of treatment. No further cell-growth inhibition was achieved by increasing concentration or incubation time. Cell morphology was analyzed after staining with hematoxylin-eosin (HE). The morphological changes noted in the treated cells...

Abstract Chemotherapy is one of the major approaches for the treatment of cancer. Therefore, the development of new chemotherapy drugs is an important aspect of medicinal chemistry. Chemotherapeutic agents include isocoumarins, which are privileged structures with potential antitumoral activity. Herein, a new 3-substituted isocoumarin was synthesized from 2-iodo-3,5-dimethoxy-benzoic acid and oct-1-yne in a cross-coupling Sonogashira reaction followed by a copper iodide-catalyzed intramolecular cyclization as key step using MeOH/Et3N as the solvent system. The present study also evaluated the leukometry, phagocytic activity, genotoxic potential and cell death induction of three different doses (5 mg/kg, 10 mg/kg and 20 mg/kg) of this newly synthesized...

A meta-analysis of ABCG2 gene polymorphism and non-small cell lung cancer outcomes Genet. Mol. Biol.
Fu,Lei; Wang,Rong; Yin,Ling; Shang,Xiaopu; Zhang,Runtong; Zhang,Pengjun.
Abstract We aimed to analyze the correlation between ABCG2 gene polymorphisms of 34 GG/(GA + AA) loci, 421 CC/(AC + AA) loci, and non-small cell lung cancer (NSCLC) therapeutic effects via meta-analysis. With key words, the databases PubMed and EMBASE were searched for clinical studies on ABCG2 polymorphism and NSCLC. RR and 95% CIs were used to compute combined effects, followed by heterogeneity testing. Publication bias was examined using the funnel plot method. Review Manager 5.3 software was used for the meta-analysis. Ten studies were included. No evidence of heterogeneity exists in these studies. The results indicate that two polymorphic loci of ABCG2 gene (34 G>A, and 421 C>A) had no relationship with the curative effect of chemotherapy for...

Inositol metabolism in Trypanosoma cruzi: potential target for chemotherapy against Chagas' disease Anais da ABC (AABC)
OLIVEIRA,MECIA M.; EINICKER-LAMAS,MARCELO.
Chagas' disease is a debilitating and often fatal disease caused by the protozoan parasite Trypanosoma cruzi. The great majority of surface molecules in trypanosomes are either inositol-containing phospholipids or glycoproteins that are anchored into the plasma membrane by glycosylphosphatidylinositol anchors. The polyalcohol myo-inositol is the precursor for the biosynthesis of these molecules. In this brief review, recent findings on some aspects of the molecular and cellular fate of inositol in T. cruzi life cycle are discussed and identified some points that could be targets for the development of parasite-specific therapeutic agents.

Rodrigues,B.A.; Faraco,C.; Oliveira,R.; Cheuiche,S.; Teixeira,F.S.; Kirst,C.; Faraon,A.; Sousa,E.M..
An osteosarcoma involving the pelvis was diagnosed in a 9-year-old neutered Labrador bitch. The clinical signs manifested by the patient before surgical removal of the tumor included urinary difficulty, dyschezia, and inconstant non-weight-bearing on the hindlimbs. These signs were linked to the localization of the tumor, which was identified as a firm and painful mass of the size of an orange in the caudal abdomen. The favorable short term clinical results after surgery and carboplatin chemotherapy showed that the association of these approaches can be employed to manage discomfort inflicted by tumor and, therefore improve the quality of life of patient of bone cancer.

Chagas disease: Present status of pathogenic mechanisms and chemotherapy Biol. Res.
Maya,Juan Diego; Orellana,Myriam; Ferreira,Jorge; Kemmerling,Ulrike; López-Muñoz,Rodrigo; Morello,Antonio.
There are approximately 7.8 million people in Latin America, including Chile, who suffer from Chagas disease and another 28 million who are at risk of contracting it. Chagas is caused by the flagellate protozoan Trypanosoma cruzi. It is a chronic disease, where 20%-30% of infected individuals develop severe cardiopathy, with heart failure and potentially fatal arrhythmias. Currently, Chagas disease treatment is more effective in the acute phase, but does not always produce complete parasite eradication during indeterminate and chronic phases. At present, only nifurtimox or benznidazole have been proven to be superior to new drugs being tested. Therefore, it is necessary to find alternative approaches to treatment of chronic Chagas. The current treatment...

Hypogammaglobulinemia: a diagnosis that must not be overlooked BJMBR
Pimenta,F.M.C.A.; Palma,S.M.U.; Constantino-Silva,R.N.; Grumach,A.S..
Humoral immunological defects are frequent and important causes of hypogammaglobulinemia, leading to recurrent infections, autoimmunity, allergies, and neoplasias. Usually, its onset occurs in childhood or during the second and third decades of life; however, the diagnosis is made, on average, 6 to 7 years afterwards. As a consequence, antibody defects can lead to sequelae. Here we describe the clinical-laboratory characteristics, treatment, and prognoses of patients with hypogammaglobulinemia. An observational, cross-sectional, and retrospective study of patients attending the recently established outpatient group of Clinical Immunology between 2013 and 2018 was carried out. Patients with IgG levels below 2 standard deviations from the mean values for the...

Possible benefit of consolidation therapy with high-dose cytarabine on overall survival of adults with non-promyelocytic acute myeloid leukemia BJMBR
Azevedo,M.C.; Velloso,E.D.R.P.; Buccheri,V.; Chamone,D.A.F.; Dorlhiac-Llacer,P.E..
In adults with non-promyelocytic acute myeloid leukemia (AML), high-dose cytarabine consolidation therapy has been shown to influence survival in selected patients, although the appropriate doses and schemes have not been defined. We evaluated survival after calculating the actual dose of cytarabine that patients received for consolidation therapy and divided them into 3 groups according to dose. We conducted a single-center, retrospective study involving 311 non-promyelocytic AML patients with a median age of 36 years (16-79 years) who received curative treatment between 1978 and 2007. The 131 patients who received cytarabine consolidation were assigned to study groups by their cytarabine dose protocol. Group 1 (n=69) received <1.5 g/m2 every 12 h on 3...

Salinomycin enhances doxorubicin sensitivity through reversing the epithelial-mesenchymal transition of cholangiocarcinoma cells by regulating ARK5 BJMBR
Yu,Z.; Cheng,H.; Zhu,H.; Cao,M.; Lu,C.; Bao,S.; Pan,Y.; Li,Y..
Chemotherapy response rates in patients with cholangiocarcinoma remain low, primarily due to the development of drug resistance. Epithelial-mesenchymal transition (EMT) of cancer cells is widely accepted to be important for metastasis and progression, but it has also been linked to the development of chemoresistance. Salinomycin (an antibiotic) has shown some potential as a chemotherapeutic agent as it selectively kills cancer stem cells, and has been hypothesized to block the EMT process. In this study, we investigated whether salinomycin could reverse the chemoresistance of cholangiocarcinoma cells to the chemotherapy drug doxorubicin. We found that combined salinomycin with doxorubicin treatment resulted in a significant decrease in cell viability...

Changes in circulating endothelial progenitor cells predict responses of multiple myeloma patients to treatment with bortezomib and dexamethasone BJMBR
Wang,L.; Du,F.; Zhang,H.M.; Zhang,W.J.; Wang,H.X..
Four cycles of chemotherapy are required to assess responses of multiple myeloma (MM) patients. We investigated whether circulating endothelial progenitor cells (cEPCs) could be a biomarker for predicting patient response in the first cycle of chemotherapy with bortezomib and dexamethasone, so patients might avoid ineffective and costly treatments and reduce exposure to unwanted side effects. We measured cEPCs and stromal cell-derived factor-1α (SDF-1α) in 46 MM patients in the first cycle of treatment with bortezomib and dexamethasone, and investigated clinical relevance based on patient response after four 21-day cycles. The mononuclear cell fraction was analyzed for cEPC by FACS analysis, and SDF-1α was analyzed by ELISA. The study population was...
