Bioethanol strains of Saccharomyces cerevisiae characterised by microsatellite and stress resistance BJM
Reis,Vanda Renata; Antonangelo,Ana Teresa Burlamaqui Faraco; Bassi,Ana Paula Guarnieri; Colombi,Débora; Ceccato-Antonini,Sandra Regina.
Abstract Strains of Saccharomyces cerevisiae may display characteristics that are typical of rough-type colonies, made up of cells clustered in pseudohyphal structures and comprised of daughter buds that do not separate from the mother cell post-mitosis. These strains are known to occur frequently in fermentation tanks with significant lower ethanol yield when compared to fermentations carried out by smooth strains of S. cerevisiae that are composed of dispersed cells. In an attempt to delineate genetic and phenotypic differences underlying the two phenotypes, this study analysed 10 microsatellite loci of 22 S. cerevisiae strains as well as stress resistance towards high concentrations of ethanol and glucose, low pH and cell sedimentation rates. The...

Antiophidian sera sterility control: topics in perspective BJPS
Santos,Paula Gomes; Santos,Dilvani Oliveira; Seródio,Lilia Ribeiro; Marinho,Rachel de Souza; Rodrigues,Carlos Rangel; Cabral,Lucio Mendes; Pádula,Marcelo de; Sousa,Valéria Pereira de; Castro,Helena Carla.
The aim of this work is to review the most important topics about the antiophidic sera sterility, including obtaining methods, sterilization procedures and clean room control using Vital Brazil Institute (VBI) as an example. Bibliographical research was performed through Medline, Lilacs, PubMed, ISI and the Fundação Oswaldo Cruz - RJ and VBI Libraries, from 1960 to 2009. The antiophidic sera for human use are immunobiologic products produced in Brazil by three national laboratories, including VBI. Due to the parenteral use, these products should be sterile and pyrogen-free, which demands the microbiological control during the whole fabrication process. The sterility and pyrogen tests are important steps to ensure the quality and safety of these...

Risk perception related to food Ciênc. Tecnol. Aliment.
REMBISCHEVSKI,Peter; CALDAS,Eloisa Dutra.
Abstract There is no human activity that is risk free, including those most trivial and essential for survival, such as eating. Various factors impact the risk perception of a population, such as whether the risk is voluntary, known, brings some benefits or whether the information about the risk is provided by sources seen as trusted. Furthermore, regional and cultural aspects, gender and age can also have an impact on risk perception, and the level of scientific knowledge of the individual about the risks has in many cases little impact on the risk perception. In most countries, the perception of consumers to certain risks present in food, including genetically modified organisms (GMO), pesticides and food additives is high, probably due to the lack of...

Estimate of dietary intake of chloropropanols (3-MCPD and 1,3-DCP) and health risk assessment Ciênc. Tecnol. Aliment.
Arisseto,Adriana Pavesi; Vicente,Eduardo; Furlani,Regina Prado Zanes; Toledo,Maria Cecília de Figueiredo.
Chloropropanols, including 3-monochloropropane-1,2-diol (3-MCPD) and 1,3-dichloropropan-2-ol (1,3-DCP), comprise a group of chemical contaminants with carcinogenic and genotoxic properties. They have been found in a variety of processed foods and food ingredients, such as hydrolyzed vegetable protein, soy sauce, cereal-based products, malt-derived ingredients, and smoked foods. This study aimed to assess the dietary exposure to 3-MCPD and 1,3-DCP in Brazil and verify whether the presence of these substances in foods could represent health risks. The intake was calculated by combining data on food consumption, provided by the Consumer Expenditure Survey 2008-2009, with the levels of contaminant occurrence determined by gas chromatography-mass spectrometry....

Fish life-history traits are affected after chronic dietary exposure to an environmentally realistic marine mixture of PCBs and PBDEs ArchiMer
Horri, Khaled; Alfonso, Sebastien; Cousin, Xavier; Munschy, Catherine; Loizeau, Veronique; Aroua, Salima; Begout, Marie-laure; Ernande, Bruno.
Polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are persistent organic pollutants that have been shown to affect fish life-history traits such as reproductive success, growth and survival. At the individual level, their toxicity and underlying mechanisms of action have been studied through experimental exposure. However, the number of experimental studies approaching marine environmental situations is scarce, i.e., in most cases, individuals are exposed to either single congeners, or single types of molecules, or high concentrations, so that results can hardly be transposed to natural populations. In the present study, we evaluated the effect of chronic dietary exposure to an environmentally realistic marine mixture of PCB and...

Impact of contaminated sediment elutriate on coastal phytoplankton community (Thau lagoon, Mediterranean Sea, France) ArchiMer
Ben Othman, Hiba; Pringault, Olivier; Louati, Hela; Hlaili, Asma Sakka; Leboulanger, Christophe.
Effects of sediment-released contaminants and nitrogen were assessed on phytoplankton communities sampled from Thau lagoon (France, Mediterranean Sea) and one close offshore marine station. Phytoplankton was exposed to sediment elutriate (seawater containing a mix of metals, organic chemicals, and nutrients) or to ammonium enrichment for four days using immersed microcosms exposed to natural conditions of light and temperature. Functional (production - respiration balance) and structural (taxonomy and cell densities) responses of the phytoplankton community were assessed. In the lagoon, both treatments stimulated phytoplankton growth, compare to controls. Conversely in the offshore station, the phytoplankton growth was stimulated only with the sediment...

Biomarkers as tools for monitoring within the Water Framework Directive context: concept, opinions and advancement of expertise ArchiMer

The Water Framework Directive (WFD) currently supports chemical and ecological monitoring programmes in order to achieve the good water surface status. Although chemical and ecological assessments are necessary, they have some limitations. Chemical approaches focus on certain substances identified as priorities, but they do not take into account other potentially harmful substances and also ignore the hazards related to contaminant cocktails. On the other hand, while ecological approaches provide holistic information on the impairment of biological communities in ecosystems, they do not distinguish the role of contaminants in these alterations, and consequently do not allow the establishment of contaminant impact reduction plans. Consequently,...

Combined replacement of fish meal and oil in practical diets for fast growing juveniles of gilthead sea bream (Sparus aurata L.): Networking of systemic and local components of GH/IGF axis ArchiMer
Benedito Palos, L; Saera Vila, A; Calduch Giner, J; Kaushik, Sadasivam; Perez Sanchez, J.
Growth performance and growth regulatory pathways were examined in juvenile gilthead sea bream fed diets containing largely plant-based ingredients. Four isonitrogenous and isolipidic extruded diets with a low level (20%) of fish meal inclusion were formulated with graded levels of a vegetable oil mixture (17:58:25 of rapeseed: linseed: palm oils) replacing fish oil at 33, 66 and 100% (33VO, 66VO and VO diets). All diets were supplemented with lysine (0.55%) and contained soy lecithin (1%). Daily growth coefficients and feed efficiency over the course of an 11-week trial were almost identical in fish fed the FO, 33VO and 66VO diets. The VO diet reduced feed intake and growth without significant effects in proximate whole body composition, nitrogen or...

Port activities require continuous or occasional dredging of considerable quantities of sediments : approximately 50 million m3 along the French coastline. A large proportion of the dredgings is authorized for dumping at sea in geographically delimited areas and following an impact evaluation of their potential harmfulness to the marine environment. The environmental risks assessment linked to dredging operations relies on knowledge related to several scientific disciplines contributing to changes in the regulations. This work, intended both for managers and for environmental scientists and engineers, is designed to take stock of the current state-of-the-art in the field of sediment contamination and related ecotoxicological hazards, along with a review of...

Integrated chemical and biological assessment of contaminant impacts in selected european coastal and offshore marine areas ArchiMer
Hylland, Ketil; Robinson, Craig D.; Burgeot, Thierry; Martinez-gomez, Concepcion; Lang, Thomas; Svavarsson, Jorundur; Thain, John E.; Vethaak, A. Dick; Gubbins, Mattew J..
This paper reports a full assessment of results from ICON, an international workshop on marine integrated contaminant monitoring, encompassing different matrices (sediment, fish, mussels, gastropods), areas (Iceland, North Sea, Baltic, Wadden Sea, Seine estuary and the western Mediterranean) and endpoints (chemical analyses, biological effects). ICON has demonstrated the use of a framework for integrated contaminant assessment on European coastal and offshore areas. The assessment showed that chemical contamination did not always correspond with biological effects, indicating that both are required. The framework can be used to develop assessments for EU directives. If a 95% target were to be used as a regional indicator of MSFD GES, Iceland and offshore...
