Assessing the viability of cryopreserved coconut zygotic embryos by electrolytic conductivity and potassium leaching PAB

The objective of this work was to adapt the application of electrolytic conductivity and potassium leaching tests to assess the viability of cryopreserved embryos of 'Anão Verde do Brasil de Jiqui' (AVeJBr) coconut. The zygotic embryos were excised, sterilized and subjected to four cryoprotectant treatments combined with three incubation times (12, 16 and 20 hours), totaling 12 treatments. The pre-treatment of mature zygotic embryos of AVeJBr coconut using a cryoprotectant with 1.75 mol L-1 of sucrose + 15% glycerol for 12 and 16 hours promoted lower embryo humidity and increased viability in electrolytic conductivity and potassium leaching tests. Induced reproduction of dourado (Salminus brasiliensis): fertilization with sperm cryopreserved in ACP®-104 Animal Sciences

Experiments were conducted to improve the fertilization rate of cryopreserved semen of dourado, Salminus brasiliensis. The experiment tested two cryoprotectant solutions at different semen: cryoprotectant ratios (1:5, 1:15, 1:25 and 1:50). The standard solution for the species (mixture of dimethyl sulfoxide, glucose, egg yolk and distilled water) was compared to a 350 mOsm ACP®-104 solution, which is composed of powdered coconut water diluted in distilled water and methylglycol. Differences between the dilutions tested were significant only for ACP®. The fertilization potential by using the standard solution at the lowest dilution (1:5) is equated when the sperm is diluted in ACP® at 1:25 or 1:50. Abstract Basidiomycetes have several biotechnological and industrial applications such as enzyme production, bioremediation, pharmaceutical and functional food production. Due to climatic features, the preservation of several basidiomycetes is threatened, and to guarantee the preservation of this genetic resource, the development of long-term preservation techniques is necessary once there is no universal protocol for the cryopreservation of basidiomycetes. Freeze-drying of ampicillin solid lipid nanoparticles using mannitol as cryoprotectant BJPS

abstract Solid lipid nanoparticles (SLNs) are interesting colloidal drug-delivery systems, since they have all the advantages of the lipid and polymeric nanoparticles. Freeze-drying is a widely used process for improving the stability of SLNs. Cryoprotectants have been used to decrease SLN aggregations during freeze-drying. In this study Ampicillin was chosen to be loaded in a cholesterol carrier with nano size range. To support the stability of SLNs, freeze-drying was done using mannitol. Particle size, drug release profile and antibacterial effects were studied after freeze-drying in comparison with primary SLNs.
