Deleterious effect of TRIS buffer on growth rates and pigment content of Gracilaria birdiae Plastino & E.C. Oliveira (Gracilariales, Rhodophyta) Acta Botanica

This work evaluated the effects of Tris (hydroxymethyl)-aminomethane (TRIS) buffer and its interaction with nutrient concentration on the development of Gracilaria birdie, a common species on the Brazilian coast that has been exploited for agar production. Responses to different conditions were assessed through growth rates and pigment content (chlorophyll a, phycoerythrin, phycocyanin and allophycocyanin). Provasoli's nutrient solution with and without TRIS addition was tested at concentrations of 12.5, 25 and 50%. The pH was also monitored. G. birdiae grew better in the absence of TRIS and at low nutrient concentrations, 12.5 and 25% (growth rates of 10.8-11.3%.day-1). This study was designed to micropropagate E. benthamii x E. dunnii, by testing chlorine concentrations for explant asepsis, the optimal concentrations of benzylaminopurine (BAP) and naphthaleneacetic acid (NAA) for bud proliferation, and the ratio between BAP and gibberellic acid (GA3) in two nutrient media for shoot elongation. Nodal segments from H12, H19 and H20 clones were disinfected with 0.5, 1.0, 1.5 and 2.0% (v v-1) of chlorine. O objetivo deste trabalho foi determinar a fonte de silício que proporcione melhor crescimento em plântulas de Cattleya loddigesii. The objective of this study is to establish an in vitro germination and cultivation protocol for murici (Byrsonima cydoniifolia A. Juss.) using zygotic embryos. Morphological and physiological photon flux influence under in vitro culture of apple shoots BABT

The aim of the present study was to evaluate the growth and development of apple rootstock shoots submitted to different photon flux densities (7, 14, 21 and 60 µmol m-2 s-1) and three culture media. The best photon flux for maximizing growth, number of shoots and leaves was 14 µmol m-2 s-1, which also resulted in the highest values of chloroplastic pigments. On the other hand, the highest photon flux (60 µmol m-2 s-1) decreased significantly these parameters. Medium with reduced sucrose concentration did not affect the analyzed parameters, enabling an efficient multiplication of the cultivar with only 1% of sucrose.

A mutant designated NC2168, which was selected from wild-type Streptococcus equisimilis CVCC55116by ultraviolet ray combined with60Co-γ ray treatment and does not produce streptolysin, was employed to produce hyaluronic acid (HA). Influence of initial L-asparagine and glycerol concentrations on the batch growth kinetics of Mycobacterium bovis BCG BJM

The influences of the L-asparagine and glycerol initial concentrations in Sauton medium on the productivities of biomass and colony forming units were studied. The submerged batch cultivations of Mycobacterium bovis were carried out in a 20 L bioreactor. The L-asparagine and glycerol initial concentrations of 4.54 g/L and 25 mL/L, respectively, corresponded to the best biomass productivity, namely 2.5 g/L.day. Meningococcal disease is an important cause of death and morbidity throughout the world. Nearly 330,000 cases and 35,000 deaths occur yearly. Neisseria meningitidis, serogroup B strain N.44/89, is prevalent in Brazil. Its outer membrane vesicles (OMV) with iron regulated proteins (IRP) are released to the culture medium and are used as antigen for vaccine production. Quality control for microbiological culture media: Is it enough to follow the NCCLS M22-A2 procedures? BJM

Stringent quality control protocols must be used in order to guaranty that a particular medium is able to recover all sort of organism that may be present in clinical samples. Our aim was to evaluate an alternative protocol that would allow us to detect medium failure to yield quantitative growth of selected pathogens, and compare with the document M22-A2 from NCCLS. The detection limit of Haemophilus influenzae was significantly different depending on media source. We conclude that for some fastidious microorganisms, quantitative verification of the growth capacity of the culture medium is advised.

Use of photocatalytic reduction to hasten preparation of culture media for saccharolytic Clostridium species BJM

Cysteine is the preferred reducing agent used in the preparation of culture media for the growth of many strictly anaerobic microorganisms; however, redox potential reduction of cysteine is very slow, making it inconvenient if the medium is needed immediately or in large quantity. The time required to reduce culture medium containing resazurin (an indicator of reducing conditions) was dramatically shortened when the medium, after being injected with the reducing agent cysteine, was irradiated with incandescent light from a halogen lamp. Sweet potato (Ipomoea batatas L.) plants were cultivated in vitro in Murashige and Skoog (MS) medium with 20 and 40 g L-1 of sucrose under two different photon flux densities (21 and 60 µmol m-2 s-1). Leaves developed in vitro mostly showed high variable to maximum fluorescence ratio (F V/F M) , indicating good development of photosynthetic apparatus. This ratio decreased during leaf aging, especially in the plants cultivated in vitro on medium with higher sucrose concentration and higher photon flux. Abstract Melanin is a natural high molecular weight pigment with the huge application value and development potential in food industry. In the present study, medium composition for melanin production by fungus Auricularia auricula was investigated. Wheat bran extract, l-tyrosine, and CuSO4 were determined to optimize medium composition by response surface methodology with Box–Behnken design. Results indicated that the optimal medium composition was 26.80% (v/v) wheat bran extract, 1.59 g/L l-tyrosine, and 0.11 g/L CuSO4, and the maximum melanin yield was 519.54 mg/L. In biotechnological processes, the culture media components are responsible for high costs and exert a strong influence on the cyanobacteria behavior. The objective of this study was to evaluate the Arthrospira platensis growth potential for biomass production under different cultivation conditions using an experimental design. Three factors that are important for cyanobacteria growth were evaluated: sodium bicarbonate (9 to 18 g/l), sodium nitrate (1.25 to 2.5 g/l), and irradiance (20 to 120 µmol photons/m2.s–1). Effect of coconut water on growth of olive embryos cultured in vitro Ciência Rural

The experiment was carried out to determine the appropriate dose of coconut water as supplement for in vitro cultivation of zygotic embryos from 19 olive genotypes. The isolated embryos of the olive seeds were immersed on culture medium containing 0 (control), 25, 50, and 100mL L-1 of fresh and sterile coconut water and kept for 45 days under controlled environment. ABSTRACT: European apple canker (EC) is caused by Neonectria ditissima, a pathogen officially registered as a quarantine pest in 2012. Thirty-five isolates of N. ditissima of different geographical regions of southern Brazil from apple branches showing symptoms of EC were identified by the specific pair primers Ch1 and Ch2 and analyzed concerning the virulence on Gala apple cultivar and morphophysiological characteristics. En esta investigación, se evaluó el efecto de las diluciones del agua de coco y la adición de biotina sobre el crecimiento de Saccharomyces cerevisiae. En este sentido, la dilución 1:1 adicionada con biotina (µ= 0.21 h-1) y biomasa (3.32 g/L) fue suficiente para dar los requerimientos nutricionales a la cepa estudiada, en comparación con el agua de coco 100% (µ= 0.13 h-1) y biomasa (3.28 g/L) y un medio cultivo sintético (µ= 0.12 h-1) con biomasa de 2.86 g/L. Los nutrientes disponibles en diluciones 1:2 (µ= 0.18 h-1) y biomasa (2.25 g/L), podrían ser considerables para estudios posteriores; no así, las diluciones 1:5 y 1:10 que parecen inadecuadas para el óptimo crecimiento de S. cerevisiae. In vitro propagation of five Alocasia species Horticultura Brasileira

The influence of cytokinin, N6-benzyladenine, on shoot proliferation of five Alocasia species (A. amazonica, A. cuprea, A. robusta, A. longiloba and A. chaii) was investigated. In vitro propagation of these species was established using shoot tip explants. Murashige & Skoog (MS) medium supplemented with different concentrations of BA (N6-benzyladenine) ranging from 0, 2, 5, 10 mg/L was then used to establish the optimum medium for shoot proliferation for all the species. MS medium supplemented with 2.0 mg/L BA was optimum for the shoot proliferation. All the tested species showed varying results for shoot number and shoot height.
