Electrocardiographic changes during low-dose, short-term therapy of cutaneous leishmaniasis with the pentavalent antimonial meglumine BJMBR
Ribeiro,A.L.P.; Drummond,J.B.; Volpini,A.C.; Andrade,A.C.; Passos,V.M.A..
The pentavalent antimonial (Sb5+) meglumine is the drug of choice for the treatment of cutaneous leishmaniasis (CL) in Brazil. Although the cardiotoxicity of high-dose, long-term Sb5+ therapy is well known, the use of low-dose, short-term meglumine has been considered to be safe and relatively free from significant cardiac effects. In order to investigate the cardiotoxicity of low-dose, short-term therapy with meglumine in cutaneous leishmaniasis, 62 CL patients treated with meglumine were studied. A standard ECG was obtained before and immediately after the first cycle of treatment (15 mg Sb5+ kg-1 day-1). The electrocardiographic interpretation was carried out blindly by two investigators using the Minnesota Code. There were no significant differences in...

Comparison of the specificity of PCR and the histopathological detection of leishmania for the diagnosis of American cutaneous leishmaniasis BJMBR
Medeiros,A.C.R.; Rodrigues,S.S.; Roselino,A.M.F..
More precise and rapid diagnostic methods for American cutaneous leishmaniasis (ACL) are necessary because of the growing number of cases observed in Brazil, including the northeastern region of the State of São Paulo. We applied PCR to 54 skin or mucosal biopsies from patients with a clinical and/or laboratory diagnosis of ACL using primers 13A and 13B, with positive results being obtained for 82% of the samples. When the PCR results were compared to those of histopathological leishmania detection, PCR showed superior results with 81.5% sensitivity and 95% CI of 68.0-95.1%. The Montenegro skin test (MST) was positive in 88.7% of patients. Since MST cannot be used as a diagnostic tool in endemic areas, the present results strongly suggest the use of PCR...

The role of natural killer cells in the early period of infection in murine cutaneous leishmaniasis BJMBR
Laurenti,M.D.; Gidlund,M.; Ura,D.M.; Sinhorini,I.L.; Corbett,C.E.P.; Goto,H..
In order to study the role of natural killer (NK) cells during the early period of Leishmania infection, BALB/c mice were selectively and permanently depleted of NK cells by injection with 90Sr and subsequently infected with Leishmania (Leishmania) amazonensis (HSJD-1 strain). 90Sr is known to selectively deplete NK cells, leaving an intact T- and B-cell compartment and preserving the ability to produce both interferon alpha and IL-2. This method of depletion has advantages when compared with depletion using anti-NK cell monoclonal antibodies because the effect is permanent and neither activates complement nor provokes massive cell death. In the present study, after one month of treatment with 90Sr, the depletion of NK cells was shown by a more than...

The role of complement in the early phase of Leishmania (Leishmania) amazonensis infection in BALB/c mice BJMBR
Laurenti,M.D.; Örn,A.; Sinhorini,I.L.; Corbett,C.E.P..
Complement-depleted and -non-depleted BALB/c mice were inoculated with Leishmania (Leishmania) amazonensis promastigotes into the hind footpad to study the role of the complement system in cutaneous leishmaniasis. Total serum complement activity was measured by hemolytic assay and C3 fragment deposit at the inoculation site was determined by direct immunofluorescence in the early period of infection, i.e., at 3, 24, 48 h and 7 days post-infection. The inflammatory reaction and the parasite burden were evaluated in the skin lesion at 7 and 30 days post-infection. Total serum complement activity decreased in the early phase of infection, from 3 to 24 h, in non-depleted mice compared to non-infected and non-depleted mice. C3 fragment deposit at the site of...

JG,Pinto; CP,Soares; J,Mittmann.
Cutaneous leishmaniasis is an infectious disease caused by protozoans of the genus Leishmania, which is transmitted through the bite of hematophagous insects of the genus Lutzomyia. This study aimed at testing in vitro the phototoxic effect of aluminum phthalocyanine tetrasulfonate (AlPcS4) on the viability of Leishmania major and Leishmania braziliensis. Stationary phase promastigote forms were treated with AlPcS4 at 1.0 µM and 10.0 µM and incubated for one hour. Then 659 nm laser was applied at 5 and 10 J/cm². Parasite viability was determined by differential count using the trypan blue dye exclusion method and by monitoring growth curves for nine days. Trypan blue exclusion assay showed a significant reduction of viable parasites compared to controls,...

Domestic feline cutaneous leishmaniasis in the municipality of Ribas do Rio Pardo, Mato Grosso do Sul state, Brazil: a case report J. Venom. Anim. Toxins incl. Trop. Dis.
Souza,AI; Nunes,VLB; Borralho,VM; Ishikawa,EAY.
Cutaneous leishmaniases are anthropozoonoses that involve many species of Leishmania and a wide variety of wild mammalian hosts, thus presenting high importance to public health. This study reports the second case of feline leishmaniasis in Mato Grosso do Sul state, in which Leishmania (Leishmania) amazonensis was found in a domestic cat from Ribas do Rio Pardo. Clinical signs were similar to those observed in other diseases commonly diagnosed in cats, such as cryptococcosis and sporotrichosis. Cutaneous leishmaniasis should, therefore, be added to differential diagnoses by feline veterinary practitioners, and also adequate investigations should be carried out to verify the relevance of domestic cats as L. amazonensis reservoirs.

Impact of leishmaniasis on public health J. Venom. Anim. Toxins incl. Trop. Dis.
Camargo,L. B; Langoni,H..
Leishmaniasis is a parasitic zoonosis caused by protozoans of the genus Leishmania transmitted by insects known as phlebotomines, which are found in wild or urban environments. It affects domestic and wild animals and transmission to man happens by accident. The disease occurs in tropical and sub-tropical areas, mainly in Asia, Europe, Africa, and the Americas. There are two forms that affect man: American cutaneous leishmaniasis (ACL) and American visceral leishmaniasis (AVL). The latter is caused by three species of Leishmania: Leishmania (Leishmania) donovani, Leishmania (Leishmania) infantum, and Leishmania (Leishmania) chagasi, which are grouped in the Leishmania (Leishmania) donovani complex. Wild reservoir hosts of L. chagasi known so far are foxes...
