Genotoxicity of lapachol evaluated by wing spot test of Drosophila melanogaster Genet. Mol. Biol.
Costa,Wender Ferreira; Oliveira,Alaide Braga de; Nepomuceno,Júlio César.
This study investigated the genotoxicity of Lapachol (LAP) evaluated by wing spot test of Drosophila melanogaster in the descendants from standard (ST) and high bioactivation (HB) crosses. This assay detects the loss of heterozygosity of marker genes expressed phenotypically on the fly's wings. Drosophila has extensive genetic homology to mammals, which makes it a suitable model organism for genotoxic investigations. Three-day-old larvae from ST crosses (females flr³/TM3, Bd s x males mwh/mwh), with basal levels of the cytochrome P450 and larvae of high metabolic bioactivity capacity (HB cross) (females ORR; flr³/TM3, Bd s x males mwh/mwh), were used. The results showed that LAP is a promutagen, exhibiting genotoxic activity in larvae from the HB cross. In...

Cytochrome P450 detection in liver of the catfish Ancistrus multispinis (Osteichthyes, Loricariidae) BABT
Klemz,Claudio; Salvo,Ligia Maria; Bastos Neto,Jayme da Cunha; Bainy,Afonso Celso Dias; Assis,Helena Cristina da Silva de.
Sensitive biological responses to environmental contaminants are useful as early warning signals to predict the damages by long-term exposure. Protocols standardization to quantify biochemical parameters in different fish species is required to validate its use as biomarkers. Comparative studies from different fish species and its interpretation are a challenge for the validation of its use as general biomarkers, representative of environmental impact. In this study, the protocol for liver cytochrome P450 (CYP) analysis from the native Brazilian fish Ancistrus multispinis was established. The microsome contamination by hemoglobin during the analysis of CYP in liver was detected, leading to misinterpretation of the results. The spectrophotometric method for...

Hepatic Cytochrome P450 as Biomarkers of Cypermethrin Toxicity in Freshwater Teleost, Channa punctatus (Bloch) BABT
Bhutia,Dawa; Rai,Benoy Kishore; Pal,Joydeb.
In this study, Channa punctatus was treated with sub-lethal concentration of cypermethrin (6.6 µg/L) for 5, 10 and 15 days and its effect on total CYP 450 and the activity of hepatic CYP450 isoforms measured. Total CYP450 content and CYP1A mediated EROD activity was significantly induced (p<0.05) in all three treated groups compared to control whereas only 15 days treated group showed significant induction in CYP2B mediated N,Ndimethylaniline demethylase activity. CYP2E1 mediated aniline hydroxylase activity showed only a marginal increase while there was inhibition of CYP3A4 mediated erythromycin demethylase activity. Liver somatic index (LSI) also showed a marginal increase in all the treated groups. Results showed differential induction of CYP1A,...

Biochemical biomarkers of exposure to deltamethrin in freshwater fish, Ancistrus multispinis BABT
Assis,Helena Cristina da Silva de; Nicareta,Lilian; Salvo,Ligia Maria; Klemz,Claudio; Truppel,Jessé Henrique; Calegari,Rossana.
This study aimed to determine the effect of sublethal doses of deltamethrin, using biochemical biomarkers as activities of cholinesterase (ChE), ethoxyresorufin-O-deethylase (EROD) and the Na+K+- ATPase and levels of total cytochrome P450 (CYP450). Fishes received sublethal doses of deltamethrin and were sacrificed after 96 h of exposure. Samples of gills, heart, brain, liver and muscle were collected for enzymatic analyses. Deltamethrin inhibited the activity of the gills and heart Na+ K+-ATPase, induced the liver total CYP450, as well as the liver EROD activity. The activity of the ChE was not inhibited by deltamethrin. Deltamethrin altered the hepatic metabolism and the normal ionic flux in Ancistrus multispinis.

Plasma hydroxy-metronidazole/ metronidazole ratio in hepatitis C virus-induced liver disease BJMBR
Marchioretto,M.A.M.; Ecclissato,C.; Silva,C.M.F. da; Cassiano,N.M.; Calafatti,S.A.; Mendonça,S.; Ribeiro,M.L.; Bernasconi,G.C.R.; Degger,M.F.; Piovesan,H.; Pedrazzoli Jr.,J..
It has been suggested that the measurement of metronidazole clearance is a sensitive method for evaluating liver function. The aim of this study was to evaluate the usefulness of plasma hydroxy-metronidazole/metronidazole ratios as indicators of dynamic liver function to detect changes resulting from the various forms of chronic hepatitis C virus (HCV) infection. A total of 139 individuals were studied: 14 healthy volunteers, 22 healthy, asymptomatic, consecutive anti-HCV-positive HCV-RNA negative subjects, 81 patients with chronic hepatitis C (49 with moderate/severe chronic hepatitis and 34 with mild hepatitis), and 20 patients with cirrhosis of the liver. HCV status was determined by the polymerase chain reaction. Plasma concentrations of metronidazole...

Role of non-nitric oxide non-prostaglandin endothelium-derived relaxing factor(s) in bradykinin vasodilation BJMBR
Resende,A.C.; Ballejo,G.; Salgado,M.C.O..
The most conspicuous effect of bradykinin following its administration into the systemic circulation is a transient hypotension due to vasodilation. In the present study most of the available evidence regarding the mechanisms involved in bradykinin-induced arterial vasodilation is reviewed. It has become firmly established that in most species vasodilation in response to bradykinin is mediated by the release of endothelial relaxing factors following the activation of B2-receptors. Although in some cases the action of bradykinin is entirely mediated by the endothelial release of nitric oxide (NO) and/or prostacyclin (PGI2), a large amount of evidence has been accumulated during the last 10 years indicating that a non-NO/PGI2 factor accounts for...

Effects of pregnancy and protein-energy malnutrition on monooxygenase O-dealkylation activity in rat liver microsomes BJMBR
Kuriyama,S.N.; De-Oliveira,A.C.A.X.; Fidalgo-Neto,A.A.; Paumgartten,F.J.R..
Xenobiotic metabolism is influenced by a variety of physiological and environmental factors including pregnancy and nutritional status of the individual. Pregnancy has generally been reported to cause a depression of hepatic monooxygenase activities. Low-protein diets and protein-energy malnutrition have also been associated with a reduced activity of monooxygenases in nonpregnant animals. We investigated the combined effects of pregnancy and protein-energy malnutrition on liver monooxygenase O-dealkylation activity. On pregnancy day 0 rats were assigned at random to a group fed ad libitum (well-nourished, WN) or to a malnourished group (MN) which received half of the WN food intake (12 g/day). WN and MN rats were killed on days 0 (nonpregnant), 11 or 20...

Expression of CYP2A3 mRNA and its regulation by 3-methylcholanthrene, pyrazole, and ß-ionone in rat tissues BJMBR
Robottom-Ferreira,A.B.; Aquino,S.R.; Queiroga,R.; Albano,R.M.; Ribeiro Pinto,L.F..
Cytochrome P450 (CYP) 2A enzymes are involved in the metabolism of numerous drugs and hormones and activate different carcinogens. Human CYP2A6, mouse CYP2A5 and rat CYP2A3 are orthologous enzymes that present high similarity in their amino acid sequence and share substrate specificities. However, different from the human and mouse enzyme, CYP2A3 is not expressed in the rat liver. There are limited data about expression of CYP2A3 in extrahepatic tissues and its regulation by typical CYP inducers. Therefore, the objective of the present study was to analyze CYP2A3 mRNA expression in different rat tissues by RT-PCR, and to study the influence of 3-methylcholanthrene, pyrazole and ß-ionone treatment on its expression. Male Wistar rats were divided into four...
