Early fragmentation of polyester urethane sheet neither causes persistent oxidative stress nor alters the outcome of normal tissue healing in rat skin Anais da ABC (AABC)
SGROTT,SANDRO M.; NEVES,RODRIGO D.; D’ACAMPORA,ARMANDO J.; BERNARDES,GERALDO J.S.; BELMONTE,LUIZ; MARTINS,THIAGO C.; BOBINSKI,FRANCIANE; CARGNIN-FERREIRA,EDUARDO; HOEPERS,ANDREZA; COMIM,CLARISSA M.; MARTINS,DANIEL F.; PIOVEZAN,ANNA P..
Abstract Silicone breast implant is associated with complications inherent to the surgical procedure. Prosthesis coating with polyurethane, however, commonly reduces the incidence of such complications. In this paper, the authors evaluated the inflammatory histomorphometric profile and oxidative damage associated to the implant of polyester urethane sheets. Forty-eight Wistar rats were divided into Sham or polyester urethane groups (n = 8/group) and underwent a polyester urethane implant in the dorsal skinfold. Tissue samples were collected on days seven, 30, and 90 after surgery and subjected to histomorphometric analysis and biochemical tests. Results were analyzed by one-way ANOVA (p ≤ 0.05). Peri-implant tissue samples exhibited characteristic...

Balanced cytokine-producing pattern in mice immunized with an avirulent Trypanosoma cruzi Anais da ABC (AABC)
Soares,Milena B. P.; Gonçalves,Renata; Pyrrho,Alexandre S.; Costa,Deise A.; Paiva,Claudia N.; Gattass,Cerli R..
We have previously demonstrated that inoculation of BALB/c mice with trypomastigotes of CL-14, an avirulent Trypanosoma cruzi clone, prevents the development of parasitemia and mortality after challenge with virulent CL strain. In this report, we investigated the cytokine and antibody profiles induced by inoculation with CL-14 clone. Groups of mice were inoculated with trypomastigotes of CL-14 clone and challenged with infective CL strain. Challenged CL-14-inoculated mice had lower levels of IFN-g and higher production of IgG1 antibodies as compared to CL strain-infected mice. Previous inoculation with CL-14 clone partially prevented the suppression of IL-2 production caused by CL strain infection. No significant...

In vitro generation of human monocyte-derived dendritic cells methodological aspects in a comprehensive review Anais da ABC (AABC)
CECHIM,GIOVANA; CHIES,JOSÉ A.B..
Abstract: Dendritic cells (DCs) initiate and shape both innate and adaptive immune responses. They are specialized in antigen presentation to naive T cells, thereby orchestrating the T cell immune responses. Human peripheral blood and tissues contain several subsets of phenotypically and functionally distinct DCs, which promote interactions between the external environment and lymphoid organs. Because of the difficulty in purifying these cells, in vitro studies only became more frequent when Frederica Sallusto and Antonio Lanzavecchia developed a method to generate DCs from blood monocytes in vitro. Nowadays a wide range of biotechnological innovations has allowed the study of DCs and their precursors in the most diverse situations faced by the immune...

Abstract Our aim is to investigate the potentially preventive effects of Aliskiren in a carrageenan-induced lung pleurisy model and to compare the standard anti-inflammatory agents, indomethacin and dexamethasone. The pleurisy model was induced through the injection of carrageenan (0.2 ml-%2) into the pleural cavity. After the experiment, serum and lung tissues were collected and biochemical, molecular and pathological examinations were performed. In our study, pleural inflammation decreased superoxide dismutase activity and the glutathione level and increased the malondialdehyde level in the lung of rats, while Aliskiren increased the superoxide dismutase activity and glutathione level and decreased the malondialdehyde level. In addition,...

Understanding the mechanisms of glutamine action in critically ill patients Anais da ABC (AABC)
Oliveira,Gisele P.; Dias,Cristina M.; Pelosi,Paolo; Rocco,Patricia R.M..
Glutamine (Gln) is an important energy source and has been used as a supplementary energy substrate. Furthermore, Gln is an essential component for numerous metabolic functions, including acid-base homeostasis, gluconeogenesis, nitrogen transport and synthesis of proteins and nucleic acids. Therefore, glutamine plays a significant role in cell homeostasis and organ metabolism. This article aims to review the mechanisms of glutamine action during severe illnesses. In critically ill patients, the increase in mortality was associated with a decreased plasma Gln concentration. During catabolic stress, Gln consumption rate exceeds the supply, and both plasma and skeletal muscle pools of free Gln are severely reduced. The dose and route of Gln administration...

Clinical and laboratory aspects of common variable immunodeficiency Anais da ABC (AABC)
Kokron,Cristina M.; Errante,Paolo R.; Barros,Myrthes T.; Baracho,Gisele V.; Camargo,Maristela M.; Kalil,Jorge; Rizzo,Luiz V..
Common variable immunodeficiency (CVID) is an immunological disorder characterized by defective antibody production, recurrent infections, most notably of the respiratory tract, autoimmune phenomena and cancer. Some CVID patients may also present disturbances of the cellular immune response such as a decrease in the number and proportion of different lymphocyte populations, diminished lymphoproliferative response to mitogens and antigens, altered production of cytokines, and deficient expression of cell-surface molecules. Most Brazilian CVID patients included in this study show a decrease in T and B lymphocyte counts in the peripheral blood. Furthermore, their lymphocytes are more susceptible to apoptosis following activation than normal individuals, and...

Saccharomyces boulardii modulates and improves the immune response to Bovine Herpesvirus type 5 Vaccine Arq. Bras. Med. Vet. Zootec.
Roos,T.B.; Avila,L.F.C.; Sturbelle,R.T.; Leite,F.L.L.; Fischer,G.; Leite,F.P.L..
ABSTRACT There have been significant efforts towards the development of more efficient vaccines for animal health. A strategy that may be used to improve vaccine efficacy is the use of probiotics to enhance the immune response of the host, leading to increased immunogenicity of antigen preparations. Bovine herpesvirus 5 (BoHV-5) is an example of an important animal pathogen for which vaccines have provided only limited protection. In this study, we examined the use of the probiotic Saccharomyces boulardii (Sb) as a potential adjuvant to improve vaccine efficiency. We found that the supplemented animals exhibited an enhanced systemic IgG antibody response toward a Th1 response in favor of IgG2a and increased mRNA expression levels of the cytokines IFN-y,...

Interleukin-1β regulates metalloproteinase activity and leptin secretion in a cytotrophoblast model Biocell
Fontana,Vanina Andrea; Sanchez,Melisa; Cebral,Elisa; Calvo,Juan Carlos.
Implantation is one of the most regulated processes in human reproduction, by endocrine and immunological systems. Cytokines are involved in embryo-maternal communication and an impaired balance could result in pregnancy loss. Here we investigated the effect of interleukin 1-β on the activity of two important metalloproteinases (MMP-2 and MMP-9) that are involved in extracellular matrix remodeling as well as the secretion of leptin, one of the reproductive hormones actively regulating their activity and secretion. We found that IL-1β activates matrix metalloproteinase activity as well as increases leptin secretion. We propose that this interleukin, through the regulation of leptin, in turn activates matrix metalloproteinases which results...

Toll-like Receptors are Key Participants in Innate Immune Responses Biol. Res.
ARANCIBIA,SERGIO A; BELTRÁN,CAROLL J; AGUIRRE,ISABEL M; SILVA,PAULINA; PERALTA,ALEXIS L; MALINARICH,FRANO; HERMOSO,MARCELA A.
During an infection, one of the principal challenges for the host is to detect the pathogen and activate a rapid defensive response. The Toll-like family of receptors (TLRs), among other pattern recognition receptors (PRR), performs this detection process in vertebrate and invertebrate organisms. These type I transmembrane receptors identify microbial conserved structures or pathogen-associated molecular patterns (PAMPs). Recognition of microbial components by TLRs initiates signaling transduction pathways that induce gene expression. These gene products regulate innate immune responses and further develop an antigen-specific acquired immunity. TLR signaling pathways are regulated by intracellular adaptor molecules, such as MyD88, TIRAP/Mal, between others...

Caveolin-1-mediated post-transcriptional regulation of inducible nitric oxide synthase in human colon carcinoma cells Biol. Res.
FELLEY-BOSCO,EMANUELA; BENDER,FLORENT; QUEST,ANDREW F.G..
Reactive oxygen species are now widely recognized as important players contributing both to cell homeostasis and the development of disease. In this respect nitric oxide (NO) is no exception. The discussion here will center on regulation of the inducible form of nitric oxide synthase (iNOS) for two reasons. First, only iNOS produces micromolar NO concentrations, amounts that are high by comparison with the picomolar to nanomolar concentrations resulting from Ca2+-controlled NO production by endothelial eNOS or neuronal nNOS. Second, iNOS is not constitutively expressed in cells and regulation of this isoenzyme, in contrast to endothelial eNOS or neuronal nNOS, is widely considered to occur at the transcriptional level only. In particular, we were...

Immune cellular response to HPV: current concepts BJID
Gonçalves,Maria Alice Guimarães; Donadi,Eduardo Antonio.
Although cellular immunity is essential for the elimination of human papillomavirus (HPV), the mechanisms involved are still poorly understood. We summarize the main mechanisms involved in cellular immune response to infections caused by HPV. Immunotherapies for HPV-related cancers require the disruption of T-cell response control mechanisms, associated with the stimulation of the Th1 cytokine response.

Serum cytokines in patients with Brazilian pemphigus foliaceus (fogo selvagem) BJMBR
Zeoti,D.M.; Figueiredo,J.F.C.; Chiossi,M.P.V.; Roselino,A.M.F..
Pemphigus foliaceus (PF) is characterized by acantholysis determined by IgG4 binding to desmoglein I, a 160-kDa desmosomal glycoprotein. To investigate the immunopathological aspects of Brazilian PF, we determined levels of serum cytokines in patients with PF. Twenty-five patients with PF and a control group consisting of 10 healthy individuals were studied. Serum IL-2, IL-4, IL-5, IL-10, IL-12 and IFN-gamma were measured in the two groups by ELISA. The median concentration of IL-2 was lower in PF patients compared to the control group (0.45 and 9.50 pg/ml, respectively), as also was the concentration of IL-4 (0.26 and 10.16 pg/ml, respectively). The same was observed for IL-5 (7.94 and 15.74 pg/ml, respectively) and for IFN-gamma (5.90 and 8.58 pg/ml,...

Cytokine accumulation in osteitis fibrosa of renal osteodystrophy BJMBR
Duarte,M.E.L.; Carvalho,E.F.; Cruz,E.A.S.; Lucena,S.B.G.; Andress,D.L..
Bone marrow fibrosis occurs in association with a number of pathological states. Despite the extensive fibrosis that sometimes characterizes renal osteodystrophy, little is known about the factors that contribute to marrow accumulation of fibrous tissue. Because circulating cytokines are elevated in uremia, possibly in response to elevated parathyroid hormone levels, we have examined bone biopsies from 21 patients with end-stage renal disease and secondary hyperparathyroidism. Bone sections were stained with antibodies to human interleukin-1alpha (IL-1alpha), IL-6, IL-11, tumor necrosis factor-alpha (TNF-alpha) and transforming growth factor-ß (TGF-ß) using an undecalcified plastic embedding method. Intense staining for IL-1alpha, IL-6, TNF-alpha and TGF-ß...
