Sechium edule is an important crop in Southern Mexico and Central America, and México was the main producer, with 53% of the world production in 2012. This species presents high levels of morphological variation that have resulted in different types of varieties not clearly defned. However, morphological characteristics have not been sufficient to classify S. edule varieties, and it has been difficult to study the species genetic diversity. Because of it, there is a need to develop methods for studying the genetic diversity in S. edule. In this regard, the first step is to find a suitable DNA extraction method. Assessment of DNA extraction methods for detection of arbuscular mycorrhizal fungi in plant roots by nested-PCR Biological Sciences

DNA extraction methods were evaluated for the yield and purity of DNA recovered from mycorrhized roots and whether the recovered DNA is suitable for amplification of arbuscular mycorrhizal (AM) fungal SSU rDNA. The DNeasy Plant Mini Kit and three extraction buffers were used alone or in combination with either polyvinylpyrrolidone (PVP), polyvinylpolypyrrolidone (PVPP) and/or activated charcoal (AC). Among the extraction methods tested, those based on the CTAB buffers yielded more DNA than those based on the TE buffer and the DNeasy Plant Mini Kit. Moreover, the use of AC alone or in combination with PVPP reduced DNA yield, while it significantly improved the purity of recovered DNA, whatever the extraction buffer. Assessment of DNA extraction methods from GMO analysis for grain monitoring in Mexico: Part II: quantification by real-time PCR Agrociencia

Because of their specificity and high throughput, real-time PCR-based methods are suitable for the monitoring of genetically modified (GM) organisms at field level or in the grain trade, in order to comply with federal regulations on biosafety. In the first part of this study, DNA extracted from different maize (Zea mays L.) tissues using several commercial purification protocols available in Mexico were evaluated in terms of DNA quality as substrates for end-point PCR.
