A strictly anaerobic, slightly halophilic and moderately thermophilic, sporulating rod designated strain DVird3(T) was isolated from deep-sea hydrothermal vent samples collected at a depth of approximately 800 m on the Atlantic Ocean Ridge. Strain DVird3(T) possessed a few laterally inserted flagella, had a DNA G+C content of 33.1 mol% and grew optimally at pH 6.6 and at 45 degreesC. Growth was observed at temperatures between 20 and 58 degreesC and at pH values between 5.8 and 8.2. The optimum NaCl concentration for growth was 3% sea salt (30 g l(-1)); no growth was observed in the presence of 15 or 60 g sea salt l(-1). A novel hydrothermal site was discovered in March 2007, on the mid-Atlantic ridge during the cruise 'Serpentine'. At a depth of 4100 m, the site 'Ashadze' is the deepest vent field known so far. Smoker samples were collected with the ROV 'Victor 6000' and processed in the laboratory for the enrichment of anaerobic heterotrophic microorganisms under high-temperature and high-hydrostatic pressure conditions. Strain CH1 was successfully isolated and assigned to the genus Pyrococcus, within the Euryarchaeota lineage within the Archaea domain. The prokaryotic diversity of culturable thermophilic communities of deep-sea hydrothermal chimneys was analysed using a continuous enrichment culture performed in a gas-lift bioreactor, and compared to classical batch enrichment cultures in vials. Cultures were conducted at 60 degrees C and pH 6.5 using a complex medium containing carbohydrates, peptides and sulphur, and inoculated with a sample of a hydrothermal black chimney collected at the Rainbow field, Mid-Atlantic Ridge, at 2,275 m depth. Both cultivation and molecular techniques were used to investigate the microbial diversity and dynamic of a deep-sea vent chimney. The enrichment cultures performed in a gas-lift bioreactor were inoculated with a black smoker chimney sample collected on TAG site on the mid-Atlantic ridge. To mimic as close as possible environmental conditions, the cultures were performed in oligotrophic medium with nitrogen, hydrogen and carbon dioxide (N-2/H-2/CO2) gas sweeping. A thermophilic, anaerobic, chemo-organotrophic sulfur-reducing bacterium, designated MV1075(T), was isolated from a deep-sea hydrothermal chimney sample collected on the Mid-Atlantic Ridge. Cells were rod-shaped with a sheath-like outer structure, motile with polar flagella and stained Gramnegative. They appeared singly, in pairs or in short chains. The temperature range for growth was 25-65 degreesC, with an optimum at 55 degreesC. Growth was observed from ph 5 to ph 9, and the optimum ph was around 7. A thermophillic, non-spore-forming, marine, sulfate-reducing bacterium, strain SL6(T), was isolated from deep-sea hydrothermal sulfides collected at Guaymas Basin. The Gram-negative-staining cells occurred singly or in pairs as small, highly motile rods. The temperature range for growth was 50-80degreesC with an optimum at 75degreesC. The pH range for growth at 70degreesC was 6.3-6.8, with an optimum at 6.5. The NaCl concentration range for growth was 5.55 g l(-1), with an optimum at 30 g l(-1). A novel thermophilic, chemolithoautotrophic T bacterium, designated as NE1206(T), was isolated from a Juan de Fuca Ridge hydrothermal vent sample (tubes of the annelid polychaete Paralvinella sulfincola attached to small pieces of hydrothermal chimney). The cells were rod-shaped (1.2-3.5x0.4-0.7 mum), occurring as single motile rods or forming macroscopic aggregates visible as pinkish to brownish streamers. The new isolate was anaerobic. A novel thermophilic, anaerobic, strictly chemoorganoheterotrophic bacterium, designated as AM1114(T), was isolated from a deep-sea hydrothermal vent sample from the East-Pacific Rise (EPR 13degrees N). The cells were long (3-10 mum) rods, motile with peritrichous flagella, and exhibited a Gram-negative cell wall ultrastructure. A novel anaerobic, Gram-positive, sporulating and strictly chemoorganoheterotrophic bacterium, designated strain DV1 184 T, was isolated from a deep-sea hydrothermal vent sample from the Mid-Atlantic Ridge. The cells were short, straight rods (4 mum long and 0.8 mum wide) and were motile with peritrichous flagella. They grew between 35 and 55 degreesC (optimum, 45 degreesC), between pH 5.0 and 8.0 (optimum, 6.0) and at 20-60 g sea salts l(-1) (optimum sea salts concentration, 30 g l(-1)).
