Presence of the cytolytic protein enterolobin in different developmental stages of Enterolobium contortisiliquum seeds Braz. J. Plant Physiol.
Lima,Consuelo M. R. de; Zanotta,Pedro J. P.; Ricart,Carlos A. O.; Sousa,Marcelo V. de.
Enterolobin is a 52.9 kDa, cytolytic, inflammatory and insecticidal protein present in Enterolobium contortisiliquum (Vell.) Morong, a Brazilian large-sized tree. This protein was previously described to occur in different forms of association (monomeric and oligomeric forms). In the current work, we investigated the presence of enterolobin forms of association and hemolytic activity in maturing, germinating and germinated seeds. Immunoblotting analysis revealed the presence of the monomeric form of enterolobin in all phases under study, whereas the dimer was detected in developing seeds in the second half of the maturation period, during germination and in the early post-germination phase. The trimeric form of enterolobin appeared one week after the dimer...

Purification and molecular mass determination of a lipid transfer protein exuded from Vigna unguiculata seeds Braz. J. Plant Physiol.
Diz,Mariângela S. S.; Carvalho,André O.; Gomes,Valdirene M..
Plants exude a variety of substances through their surface especially of roots and germinating seeds. Some of these released compounds seem to have an inhibitory action against certain pathogens. Lipid transfer proteins (LTPs) are 9 kDa cysteine-rich cationic peptides and are thought to play a role in the protection of plants against microbial infections. The aim of this work was to isolate and determine the molecular mass of a LTP present in the exudates of imbibed cowpea seeds. For exudation induction, 50 seeds were submerged in 50 mL sterile 100 mmol.L-1 Na-acetate buffer, pH 4.5 and shaken at 30 ºC for 24 h. The resulting exudate was subjected to ammonium sulphate fractionation and the pellet formed between 0 and 70 % saturation was dialysed and...

Pathogenesis-related proteins in Brazilian wheat genotypes: protein induction and partial gene sequencing Ciência Rural
Freitas,Loreta Brandão de; Koehler-Santos,Patrícia; Salzano,Francisco Mauro.
Leaves from 14 Brazilian genotypes of Triticum aestivum L. were treated with salicylic acid to induce pathogenesis-related (PR) proteins. Inter and intracellular extracts were then obtained and investigated through polyacrilamide gel electrophoresis. Seven bands were observed. Material related to two of them (of 40 and 24 kDa) occurred in intracellular spaces only. DNA from these same genotypes was then amplified through PCR using primers developed from three sequences encoding PR proteins, and compared with previously described sequences. The fragments presented homologies to PR groups 1, 3 (chitinases), and 5 (thaumatin-like). The PR3-like sequence also showed a site characteristic of PRs induced by ethylene and a portion without homology with previous...
