Response to domestication and selection for growth in the European sea bass (Dicentrarchus labrax) in separate and mixed tanks ArchiMer
Vandeputte, Marc; Dupont Nivet, M; Haffray, P; Chavanne, H; Cenadelli, S; Parati, K; Vidal, Marie-odile; Vergnet, Alain; Chatain, Beatrice.
Selective breeding of European sea bass (Dicentrarchus labrax) receives a growing interest, as the estimated heritability of growth is medium to high. In this study, we compared the offspring of four groups of sea bass sires, mated with the same wild dams: wild (W), first generation of domestication (D), first generation of mass selection for length (M), first generation of PROSPER-like selection for length (P). The comparison was done both in replicated tanks (separate rearing) and in mixed tanks (mixed rearing) where sire origins were recovered by genotyping of eight microsatellite markers. Weight, length and growth rate were measured from day 238 post-fertilization (69 g mean weight) to day 611 post-fertilization (390 g mean weight). Both in mixed and...

Impact of essential fatty acid deficiency and temperature on tissues' fatty acid composition of European sea bass (Dicentrarchus labrax) ArchiMer
Skalli, Ali; Robin, Jean; Le Bayon, Nicolas; Le Delliou, Herve; Person, Jeannine.
The effects of essential fatty acid deficiency and temperature on the fatty acid profiles of polar lipids (PL) and neutral lipids (NL) from various tissues (muscle, liver, gills, eyes and brain) of European sea bass juveniles were compared in a two factorial design. Fish (60 g) were held for 84 days at 22 or 29 C (upper limit for growth) and fed either at a lower or a higher level than n-3 HUFA minimal requirement for growth (0.4% and 2.2% n-3 HUFA dry matter for diets LD and HD, respectively). Essential fatty acid deficiency had a major influence on fatty acids in NL fraction of all tissues, a more moderate influence on PL of muscle, liver and gills, while a low and very low diet influence was observed on eyes and brain PL fatty acid content,...

Human chorionic gonadotropin induces spermatogenesis and spermiation in 1-year-old European sea bass (Dicentrarchus labrax): Assessment of sperm quality ArchiMer
Schiavone, R; Zilli, L; Vilella, S; Fauvel, Christian.
The aims of the present study were (a) to compare sperm quality (percentage of motile spermatozoa, motility duration, density and fertility after cryopreservation) between precocious and normally maturing male European sea bass Dicentrarchus labrax, (b) to examine the potential of human chorionic gonadotropin (hCG) to increase spermiation in precocious males and (c) to examine the potential of hCG to induce spermatogenesis and spermiation in non-precocious 1-year-old males. One hundred precocious and 100 non-precocious fish were each randomly divided in two groups each: control (precocious saline-treated and non precocious saline-treated) and treated (precocious hCG-treated and non precocious hCG-treated). Treated groups were administered weekly with 1000...

Person, Jeannine; Skalli, Ali; Dulau, Barbara; Le Bayon, Nicolas; Le Delliou, Herve; Robin, Jean.
Triplicate groups of European sea bass juveniles (60 g) were fed for 84 days two experimental diets containing either vegetable or marine oil (0.4 and 2.2% n-3 highly unsaturated fatty acid [n-3 HUFA] dry matter in LD and HD, respectively) and held at 22 or 29 degreesC (LD-22, LD-29, HD-22 and HD-29 groups). Significant differences in mass gain between the two diets and temperatures were, observed from day 28 onwards, and on day 84, weight was 18% higher in the HD-29 group than in the LD-22 group (the highest and lowest values. respectively). Over the 84-day period-SGR was significantly affected both by temperatures and diets (P=6x10(-4) and 2x10(-4) respectively), averaging 1.01 and 1.07 in the HD-22 and HD-29 groups compared to 0.92 and 1.0 in the LD-22...

Genotype by diet interactions in European sea bass (Dicentrarchus labrax L.): Nutritional challenge with totally plant-based diets ArchiMer
Le Boucher, Richard; Vandeputte, Marc; Dupont-nivet, M.; Quillet, E.; Ruelle, Francois; Vergnet, Alain; Kaushik, Sadasivam; Allamellou, J. M.; Medale, F.; Chatain, Beatrice.
Aquaculture of carnivorous species has strongly relied on fish meal and fish oil for feed formulation; however, greater replacement by terrestrial plant-based products is occurring now. This rapid change in dietary environment has been a major revolution and has to be taken into consideration in breeding programs. The present study analyzes potential consequences of this nutritional tendency for selective breeding by estimating genetic parameters of BW and growth rates estimated by the thermal growth coefficient (TGC) over different periods with extremely different diets. European sea bass (Dicentrarchus labrax L.) from a factorial cross (1,526 fish) between 25 sires and 9 dams were used to estimate heritabilities and genotype by diet interaction. Starting...

Metabolic response to hypoxia in European sea bass (Dicentrarchus labrax) displays developmental plasticity ArchiMer
Cadiz, Laura; Zambonino-infante, Jose-luis; Quazuguel, Patrick; Madec, Lauriane; Le Delliou, Herve; Mazurais, David.
Several physiological functions in fish are shaped by environmental stimuli received during early life. In particular, early-life hypoxia has been reported to have long-lasting effects on fish metabolism, with potential consequences for fish life history traits. In the present study, we examine whether the synergistic stressors hypoxia (40% and 100% air saturation) and temperature (15° and 20 °C), encountered during early life, could condition later metabolic response in European sea bass (Dicentrarchus labrax) juveniles. Growth rate and metabolic parameters related to carbohydrate and lipid metabolism in the liver were investigated at the juvenile stage under normoxic and chronic hypoxic conditions. Juvenile growth rates were significantly lower...

Sammouth, Sophie; Roque D'Orbcastel, Emmanuelle; Gasset, Eric; Lemarie, Gilles; Breuil, Gilles; Marino, Giovanna; Coeurdacier, Jean-luc; Fivelstad, Sveinung; Blancheton, Jean-paul.
Sea bass (Dicentrarchus tabrax) (135 +/- 4 g) were reared under tank-based recirculating aquaculture system for a 63-day period at four densities: 10, 40, 70, 100 kg m(-3). Fish performance, stress indicators (plasma cortisol, proteonemia plus other blood parameters-Na+, K+, glucose, pH, total CO2- and water quality were monitored. At the end of the 63-day period, resistance to infection was also studied by a nodavirus challenge. A 25-day test was performed on fish from two extreme densities (10 and 100 kg m(3)) and one intermediate density (40 kg m(3)). With regards to the different density treatments, there was no significant difference between the daily feed intake (DFI) and the specific growth rate (SGR) up to a density of 70 kg m(-3). No significant...

Growth Performance and Adaptability of European Sea Bass (Dicentrarchus labrax) Gut Microbiota to Alternative Diets Free of Fish Products ArchiMer
Pérez-pascual, David; Estellé, Jordi; Dutto, Gilbert; Rodde, Charles; Bernardet, Jean-françois; Marchand, Yann; Duchaud, Eric; Przybyla, Cyrille; Ghigo, Jean-marc.
Innovative fish diets made of terrestrial plants supplemented with sustainable protein sources free of fish-derived proteins could contribute to reducing the environmental impact of the farmed fish industry. However, such alternative diets may influence fish gut microbial community, health, and, ultimately, growth performance. Here, we developed five fish feed formulas composed of terrestrial plant-based nutrients, in which fish-derived proteins were substituted with sustainable protein sources, including insect larvae, cyanobacteria, yeast, or recycled processed poultry protein. We then analyzed the growth performance of European sea bass (Dicentrarchus labrax L.) and the evolution of gut microbiota of fish fed the five formulations. We showed that...

What is the heritable component of spinal deformities in the European sea bass (Dicentrarchus labrax)? ArchiMer
Bardon, Agnes; Vandeputte, Marc; Dupont-nivet, Mathilde; Chavanne, Herve; Haffray, Pierrick; Vergnet, Alain; Chatain, Beatrice.
Malformations in farmed fish are a prime research topic as it has a major impact on fish welfare and on the economical value of the product. In this study, the genetic component of spine deformities was estimated in European sea bass, a major species of Mediterranean aquaculture, with the use of a posteriori reconstruction of pedigree with microsatellites. Our population exhibited a very high malformation rate (81%),probably due to intense swimming of fish during pre-growing The heritability of global spine deformities was evaluated to be 0.21 +/- 0.04 on the underlying scale (033 +/- 0.06 for lordosis, 0.13 +/- 0.04 for scoliosis). Deformities scored by an internal or external examination turned out to be genetically the same trait (genetic...

Characterising Essential Fish Habitat using spatio‐temporal analysis of fishery data: A case study of the European seabass spawning areas ArchiMer
Dambrine, Chloe; Woillez, Mathieu; Huret, Martin; De Pontual, Helene.
Fish habitats sustain essential functions for fish to complete their life cycle, such as feeding, growing and spawning. Conservation is crucial to maintain fish populations and their exploitation. Since 2013, the spawning stock biomass of the northern stock of European seabass (Dicentrarchus labrax) has been in a worrying state. A series of low recruitments with a persistently high level of fishing has been blamed, raising concerns about the processes involved in seabass reproduction and settlement in nurseries. Here, we characterise seabass spawning areas along the French Atlantic coast using vessel monitoring system (VMS) data. A non‐linear geostatistical approach was applied, from 2008 to 2014, to detect locations where seabass aggregate for spawning....

Effects of dispersant treated oil upon exploratory behaviour in juvenile European sea bass (Dicentrarchus labrax) ArchiMer
Aimon, Cassandre; Lebigre, Christophe; Le Bayon, Nicolas; Le Floch, Stephane; Claireaux, Guy.
Accidental spills are pervasive pollution in aquatic ecosystems. Resorting to chemical dispersant is one of the most implemented strategies in response to oil spills, but it results in an increase in the bio-availability of oil compounds known to disturb fish neurosensory capacities and hence fish habitat use. While it has become well established that acute oil exposure can cause a range of physiological defects, sub-lethal consequences on animal behaviour have only received recent attention. Here we investigated the effect of an exposure to a 62 h- dispersant treated oil on the exploration tendency (exploratory activity, and avoidance of unfamiliar open areas) of juvenile European sea bass. Three different concentrations of chemically dispersed oil were...

Sea bass is a major species in Mediterranean aquaculture, but has a distribution area ranging from North Atlantic to South Mediterranean, with a population structure previously revealed by population genetics. To test the farming performances of wild sea bass populations, we produced a partial diallel cross mating scheme, using sires originating from North Atlantic (NAT), South Atlantic (SAT), West Mediterranean (WEM), North-East Mediterranean (NEM) and South-East Mediterranean (SEM). Fifteen sires per origin were mated in a full-factorial design using artificial fertilization with 9 NAT dams and 17 WEM dams, producing 10 population crosses and 1950 potential full-sib families. All fish were reared together, then tagged at an average weight of 20 g and...

Effect of oxidation–reduction potential on performance of European sea bass (Dicentrarchus labrax) in recirculating aquaculture systems ArchiMer
Li, Xian; Blancheton, Jean-paul; Liu, Ying; Triplet, Sebastien; Michaud, Luigi.
The direct impact of oxidation–reduction potential (ORP) on fish welfare and water quality in marine recirculating aquaculture systems (RAS) is poorly documented. In this study, the effects of the fish size (S1, S2, S3) and ORP level (normal, four successive levels) on the performance of European sea bass (Dicentrarchus labrax) were investigated. Three size fish were distributed into two RAS (RAS and RAS O3). Ozone was injected into RAS O3 to increase the ORP level. The ORP was stabilized to four successive levels: 260–300, 300–320, 320–350, and 300–320 mV in fish tanks during four periods (P1–4). At the last day of each period, the hematological parameters, plasma protein and mortality of sea bass were analyzed. Two-way ANOVA revealed that several...

Rapid metabolic adaptation in European sea bass (Dicentrarchus labrax) juveniles fed different carbohydrate sources after heat shock stress ArchiMer
Enes, P; Panserat, S; Kaushik, Sadasivam; Oliva Teles, A.
A study was conducted to evaluate the effect of two dietary carbohydrate sources (waxy maize starch and glucose) on the metabolic adaptation of sea bass juveniles (initial weight: 24 g) to a heat shock treatment (temperature rise from 18 degrees C to 25 degrees C within 24 h). Two isonitrogenous and isolipidic diets were fon-nulated to contain 20% waxy maize starch (WS diet) or 20% glucose (GLU diet). Triplicate groups of fish were fed to near satiation for 4 weeks at both temperatures (18 degrees C and 25 degrees C). Then, fish previously maintained at 18 degrees C were submitted to a heat shock (18 degrees C to 25 degrees C) and continued to be fed with the same diets during I more week. This preliminary study assessed genotype x diet interaction in late growth of European sea bass (Dicentrarchus labrax) fed with either a fish meal (FM)- or a fish oil (FO)-based diet (M) or an all-plant-based (PB) diet. A total of 550 fish from 224 families were reared together and tagged. DNA was sampled and microsatellites were used to assign parentage. When fish weight was 192 +/- 54 g, two tanks were fed with M (FM: 100%; FO: 100%) and two others with PB (FM: 0% and FO: 0%). Body weight (BW), fork length (FL) and fillet lipid content (CorrFat) were analysed with a linear model and with REML methodology. We observed no significant differences between groups, but a slightly lower (P=0.03) daily growth coefficient in sea bass fed PB than in those fed M....

Body shape variation in meiotic gynogenetic and triploid sea bass, Dicentrarchus labrax ArchiMer
Peruzzi, Stefano; Wiborg-dahle, Stine; Haffray, Pierrick; Chatain, Beatrice; Primicerio, Raul.
A method of geometric morphometrics was used to characterize body shape variation in meiotic gynogenetic and triploid sea bass in comparison with diploid controls. A total of 13 coordinate landmark data were recorded on the profile of 34-month-old control (n = 81), gynogenetic (n = 117) and triploid (n = 131) siblings generated from two females (A and B). Three main axes of morphometric variation were consistent, and these accounted for 60.8 and 57.6% of the total variability in females A and B, respectively. There were significant shape deviations of triploid and gynogenetic fish relative to the controls, and differences were particularly evident in the progeny of one female. Overall, triploids displayed a more slender shape than diploids, whereas...

Cloning of endothelin-1 (ET-1) from European sea bass (Dicentrarchus labrax) and its gene expression analysis in larvae with retinoic acid-induced malformations ArchiMer
Geay, Florian; Darias, Maria J.; Santigosa, E.; Desbruyeres, Elisabeth; Quazuguel, Patrick; Zambonino, Jose-luis; Cahu, Chantal; Mazurais, David.
It is known in vertebrates that endothelin-1 (ET-1) plays a key role ill morphogenesis whose expression level greatly influences the development of craniofacial malformations. In the Present study, the complete cDNA fragment encoding a precursor of endothelin-1, the preproendothelin-1 (PPET1), was cloned by RACE-PCR from European sea bass. The cDNA encoded a 199 amino acid polypeptide that was composed of the "mature" and "big" ET-1. Relative ET-1 expression levels were investigated in European sea bass larvae fed microparticulate diets containing the standard amount (0.08 g retinol/kg diet: group N) or all excess of retinoic acid which induces skeletal malformations (0.5 g retinol/kg diet: group RA). Coupling spectral analysis and hidden Markov models for the segmentation of behavioural patterns ArchiMer
Heerah, Karine; Woillez, Mathieu; Fablet, Ronan; Garren, Francois; Martin, Stephane; De Pontual, Helene.
Background Movement pattern variations are reflective of behavioural switches, likely associated with different life history traits in response to the animals’ abiotic and biotic environment. Detecting these can provide rich information on the underlying processes driving animal movement patterns. However, extracting these signals from movement time series, requires tools that objectively extract, describe and quantify these behaviours. The inference of behavioural modes from movement patterns has been mainly addressed through hidden Markov models. Until now, the metrics implemented in these models did not allow to characterize cyclic patterns directly from the raw time series. To address these challenges, we developed an approach to i) extract new metrics...

New insights into behavioural ecology of European seabass off the West Coast of France: implications at local and population scales ArchiMer
De Pontual, Helene; Lalire, Maxime; Fablet, Ronan; Laspougeas, Claire; Garren, Francois; Martin, Stephane; Drogou, Mickael; Woillez, Mathieu.
From 2010 to 2012, 246 data storage tags were deployed on European seabass in the Iroise Natural Marine Park, a marine protected area (MPA) off west Brittany, France. A return rate of 14.6% associated with long time series of data provided new information on fish ecology (e.g. maximum experienced depth greater than 225 m, temperature range 6.80–21.87°C). Depth and temperature series were used to infer individual migration using an innovative hidden Markov model (HMM) especially developed for seabass geolocation. Reconstructed fish tracks revealed that seabass is a partially migratory species, as individuals exhibited either long-distance migrations towards the Bay of Biscay or the Celtic Sea, or residency behaviour in the Iroise Sea.
