Comparison of four extraction methods to detect hepatitis A virus RNA in serum and stool samples BJID
Paula,Vanessa Salete de; Villar,Livia Melo; Gaspar,Ana Maria Coimbra.
The efficiency of extraction methods for hepatitis A virus (HAV) RNA in clinical samples is of great importance for molecular diagnosis, especially in regions endemic for HAV, such as Brazil. We compared the efficiency of four different extraction techniques in serum and stool samples for the detection of hepatitis A virus by reverse transcription PCR (RT-PCR). We used PCR to analyse serum and stool samples of 12 patients who were referred to the Brazilian Reference Center for Viral Hepatitis (BRCVH) in Rio de Janeiro. The methods tested were Proteinase K, Silica, TRIzol and Guanidine isothiocyanate. Proteinase K extraction was the best method for serum samples; it detected the HAV-RNA in 11 of the 12 samples. The guanidine isothiocyanate method was the...

Analysis of the composition of Brazilian propolis extracts by chromatography and evaluation of their in vitro activity against gram-positive bacteria BJM
Sawaya,Alexandra C. H. F.; Souza,Kirllian S.; Marcucci,Maria C.; Cunha,Ildenize B.S.; Shimizu,Mario T..
Brazilian propolis from São Paulo state was submitted to extraction using several solvents, resulting in extracts with different composition. These extracts were submitted to Thin Layer Chromatography (TLC). Bioauthographic analysis of the TLC plates identified fractions with inhibitory activity, which were then analysed by High Performance Liquid Chromatography (HPLC). In vitro assays, commonly used to evaluate the activity of propolis against Gram-positive bacteria, were compared to determine which rendered the most consistent results. The bactericidal activity of these extracts were analysed by Serial Dilution in Tubes and Agar Plate Diffusion. Serial Dilution in Tubes obtained the most consistent results, with the Minimal Bactericidal Concentration of...

Sustainable production with obtaining glucosamine from crab exoskeletons Ciência Rural
Montiel-Montoya,Jorge; Valdez-Morales,Maribel; Reyes,Cesar; Barrales-Cureño,Hebert Jair.
ABSTRACT: Two chemical treatments, five enzymatic (pectinase, lipase, hemicellulase, hemicellulose-cellulase or lipase-pectinase) and one microbiological (Bacillus subtilis) treatment were evaluated to obtain glucosamine hydrochloride (Gluc-HCl) from the chitin obtained from crab (Callinectes bellicosus) exoskeletons. Chemical treatments were referred as Method A (HCl hydrolysis during 75 min at 90°C) and Method B (HCl hydrolysis during 20 min and 14 h of rest). Glucosamine and, in some cases, N-acetyl-D-glucosamine were identified and quantified by HPLC. Treatments with the greater concentrations of Gluc-HCl in descending order were: lipase (94.4 mg/g), microbiological (45.7 mg/g), lipase-pectinase (22.9 mg/g), hemicellulase-cellulase (20.9 mg/g),...
