Comparison of traditional microscopy and digitized image analysis to identify and delineate pelagic fish egg spatial distribution ArchiMer
Lelievre, Stephanie; Antajan, Elvire; Vaz, Sandrine.
One of the problems concerning studies of fish egg distribution is the weak spatial and temporal resolution due to the workload that examination of a large number of samples would demand. Recently, the development of a new laboratory imaging system, the ZooScan, capable of obtaining relatively good resolution images enables automated zooplankton identification using supervised learning algorithms. This new approach was applied to formalin-fixed fish egg samples collected during French winter IBTS (International Bottom Trawl Surveys) in the Eastern English Channel and the Southern North Sea. Fish egg spatial distributions of seven species based on the microscope and ZooScan identifications were compared. Abundance and distribution maps of winter-spawning...

Identification and characterisation of winter spawning ground in the English Channel and Southern North Sea ArchiMer
Lelievre, Stephanie.
A better knowledge and monitoring of principal commercial fish spawning grounds have become necessary in the North Sea. The efficiency of CUFES was proved by sampling pelagic fish eggs in winter in Eastern Channel and Southern North Sea. Fish egg taxonomic identification based on visual criteria cannot always be carried out effectively. In particular, cod (Gadus morhua), and whiting (Merlangius merlangus) or flounder (Platichthys flesus) and dab (Limanda limanda) have the same range of egg diameter and similar morphologies. Alternative identification methods using molecular techniques were developed to improve the accuracy of egg taxonomic identification. First, PCR-RFLP method, then, in order to accelerate egg identification, the use of a new laboratory...

The effectiveness of the continuous underway fish egg sampler compared with standard vertical plankton hauls for sampling the eggs of demersal species in the southern North Sea ArchiMer
Lelievre, Stephanie; Vaz, Sandrine; Fox, Christopher.
To date, the continuous underway fish egg sampler (CUFES) has been used primarily to map the spawning areas of pelagic fish in upwelling regions. To assess its utility in mapping the spawning grounds of demersal species, species composition and densities of eggs collected by CUFES were compared with results obtained from ring-net samples (VET: vertical egg trawl) from the southern North Sea over three annual cruises conducted in January/February. CUFES collected slightly fewer eggs per unit volume of water filtered when compared with vertically integrated samples but maps of egg distributions from both devices showed broadly similar patterns. Geostatistical analyses revealed that small-scale variability was greater with CUFES than with VET. CUFES may thus...

Maturation and spawning of tropical and subtropical marine finfish by environmental manipulations ArchiMer
Roberts Jr, D.
Circannual, diel, and lunal periodic mechanisms in naturally reproducing stocks of tropical and subtropical fishes are reviewed. Manipulations of biological clocks, used to control reproduction in captive broodstocks, are detailed. Experiments to induce controlled maturation and spawning in fishes either test efficacy of changing environmental conditions (regimes), or constant environmental conditions (controlled). Data have been generated in experimental/scientific studies, as well as applied research. Effects of controlled environmental regimes on serum steroids, gametogenesis, spawning, fry performance and egg quality are summarized for selected members of Centropomidae, Serranidae, and Sciaenidae.

The ZooCAM, a new in-flow imaging system for fast onboard counting, sizing and classification of fish eggs and metazooplankton ArchiMer
Colas, Florent; Tardivel, Morgan; Perchoc, Jonathan; Lunven, Michel; Forest, Bertrand; Guyader, Gerard; Danielou, Marie-madeleine; Le Mestre, Sophie; Bourriau, Paul; Antajan, Elvire; Sourisseau, Marc; Huret, Martin; Petitgas, Pierre; Romagnan, Jean-baptiste.
In this paper we present the ZooCAM, a system designed to digitize and analyse on board large volume samples of preserved and living metazooplankton (i.e. multicellular zooplankton) and fish eggs > 300 µm ESD. The ZooCAM has been specifically designed to overcome the difficulties to analyse zooplankton and fish eggs in the framework of the PELGAS survey, and provide high frequency data. The ZooCAM fish eggs counts were comparable to those done with a dissecting microscope. The ZooCAM enabled the accurate prediction and fast on board validation of staged anchovy and sardine eggs in almost real time after collection. A comparison with the ZooScan, on a more complex zooplanktonic community, provided encouraging results on the agreement between the 2...

PCR-RFLP analyses of formalin-fixed fish eggs for the mapping of spawning areas in the Eastern Channel and Southern North Sea ArchiMer
Lelievre, Stephanie; Verrez-bagnis, Veronique; Jerome, Marc; Vaz, Sandrine.
A better knowledge on spawning grounds of principal commercial fishes from North Sea is necessary for their monitoring. Fish egg taxonomic identification based on morphological characters cannot always be carried out effectively. This is particularly the case for a number of Gadidae fish species eggs (i.e. cod, haddock and whiting, and three rockling species) which have the same range of size and for which molecular-based techniques can help improving the accuracy of egg taxonomic identification, and hence the reliability of studies on spawning areas. In this study, formalin fixation, egg conservation and DNA extraction were optimized in order to develop a specific PCR-RFLP method. Based on the sequence of the cytochrome b gene of reference specimens, a...

Integrating molecular identification of pelagic eggs with geostatistical mapping to improve the delineation of North Sea fish spawning grounds ArchiMer
Lelievre, Stephanie; Jerome, Marc; Maes, Gregory E.; Vaz, Sandrine; Calaivany, Sachidhanandam; Verrez-bagnis, Veronique.
Maps of the spawning grounds of commercially important fishes are necessary when assessing the level of connectivity between life stages of fishes and for identifying ecologically valuable marine areas. A first step toward mapping the spawning grounds is a reliable and rapid species identification of pelagic fish eggs to assess the spatio-temporal distribution of spawning aggregations. As many species have similar egg sizes and morphology, the molecular validation of visually identified eggs is often essential for the use of such data in fisheries management. In the present study, we developed a rapid 16S rRNA PCR-restriction fragment length polymorphism (RFLP) assay to distinguish between formalin-fixed fish eggs of dab Limanda limanda, flounder...

Adult and juvenile fish inter and intraspecific predation may be an important source on these first development stages. In this work, adult and juvenile mackerel samples from two areas of Buenos Aires province, one south of Cape San Antonio and the other near Mar del Plata city were analyzed. An important anchovy postlarvae ingestion in both regions considered was observed. A high number of mackerel eggs in the stomach contents from adults was identified. This evidence of cannibalistic behaviour in the argentine mackerel is the first of its kind made in this species and allows to suggest that embryo mortality due to cannibalism might constitute and important percentage of the total embryonic mortality.

Abundancia y distribucion de huevos y larvas de la merluza argentina (Merluccius hubbsi Marini, 1933) en un area de desove intensivo en el litoral norpatagonico. OceanDocs
Bellizia, L.L.; Pan, J.; Venerus, L.A..
Argentine hake Merluccius hubbsi represents nowadays the main fishery resource of the Argentine Sea, in spite of the fact that the percentage of captures related to total fishery production has fallen down 30% in the last decade. The abundance and distribution of hake eggs and larvae in an intense spawning area in front of Isla Escondida (Chubut province), are analyzed. This phenomena was studied from a comparative-historical point of view allowing to apppreciate the evolution of a resource at present undergoing an overfishing pressure, capable of pushing it to collapse. Since 1973 maximum egg densities diminished and, if total egg production is considered a reliable index for recruitment prediction, no signs of overcoming stocks can be seen. On the other...
