Abstract Mycotoxins are secondary metabolites produced by specific filamentous fungi that are common contaminants of agricultural commodities. These compounds are toxic to humans and animals, and they are a health problem worldwide. Mycotoxin-containing feeds can cause serious diseases in farm animals and substantial economic losses. Therefore, the objective of this study was to determine the occurrence and levels of six mycotoxins in three types of forage at retail in the state of Nuevo León, México. Anti-Fusarium moniliforme activity and fumonisin biodegradation by corn and silage microflora 52

Studies were carried out to isolate microorganisms from corn and silage screened for their ability to inhibit F. moniliforme growth (strain 113F) in association with fumonisin detoxification. Among 150 isolates four Gram-positive bacilli and one yeast with inhibitory activity were selected. The inhibition zone ranged from 50 to 72.5 mm using cultures, and from 25 to 52.5mm for crude alcoholic extract. The isolates S9, S10, S69 (sporulated bacilli) and SE3071 (yeast) degraded 43, 48, 83 and 57% of the initial FB1 concentration, respectively. The pH increased gradually in the medium during incubation for biodegradation process.

Fusarium verticillioides strains isolated from corn feed: characterization by fumonisin production and RAPD fingerprinting 52

In this study a total of 16 Fusarium verticillioides strains isolated from corn feed samples were characterized by fumonisin (FB) production and random amplified polymorphic DNA (RAPD). All the strains produced FB1 and FB2 with levels ranging from 2.41 to 3996.36 µg/g, and from 1.18 to 1209.91 µg/g, respectively. From the 16 F. verticillioides strains, four were identified as low (3.59 to 1289.84 µg/g), eight as intermediate (>1289.84 to 3772.44 µg/g) and four strains as high (>3772.44 µg/g) fumonisin producers. From the total of 105 loci amplified, 60 (57.14%) were polymorphic. RAPD analysis showed very similar patterns among low, moderate and high fumonisin-producing strains. Production of aflatoxins by Aspergillus flavus and of fumonisins by Fusarium species isolated from Brazilian sorghum 58

Fifty-nine Aspergillus flavus and 35 Fusarium verticillioides strains, isolated from freshly harvested (10) and stored (130) Brazilian sorghum samples, were tested regarding their ability to produce aflatoxins (coconut milk agar) and fumonisins (rice culture), respectively. Aflatoxins B1 and B2 were detected by TLC, and fumonisins B1 and B2 were analyzed by HPLC. Thirty-eight (64.4%) A. flavus strains produced detectable levels of aflatoxins at concentrations ranging from 12.00 to 3282.50 µg/kg (AFB1 + AFB2), while thirty two (91%) F. verticillioides strains produced FB1 at concentrations ranging from 0.12 to 5.38 µg/g. It is estimated that 25 to 50% of the crops harvested worldwide are contaminated with mycotoxins. Because of the toxic and carcinogenic potential of mycotoxins, there is an urgent need to develop detection methods that are rapid and highly specific. The highly advanced physico-chemical methods for the analysis of mycotoxins in use, have the disadvantage that highly sophisticated clean-up and/or derivatization procedures must be applied. Detection of fumonisin producing Fusarium verticillioides in paddy (Oryza sativa L.) using polymerase chain reaction (PCR) 58

The study reports the occurrence of fumonisin producing Fusarium verticillioides in 90 samples of stored paddy (Oryza sativa L.)collected from different geographical regions ofKarnataka, India. Fumonisin producing F. verticillioides was identified based on micromorphological characteristics and PCR using two sets of primers. One set of primers was F. verticillioides species specific, which selectively amplified the intergenic space region of rDNA. The other set of primers was specific to fumonisin producing F. verticillioides. Eight paddy samples were positive for F. verticillioides. Eleven isolates obtained from these samples were capable of producing fumonisin.

ABSTRACT: Horse leukoencephalomalacia (ELEM) is a disease caused by the ingestion of mycotoxins (fumonisins) produced by fungi of the genus Fusarium that infect corn and/or its byproducts. This disease has been described by ingestion of mature corn with humidity above 15% at temperatures below 20°C. The aim of this paper was to report an outbreak of leukoencephalomalacia in horses fed with immature corn. Two horses out of three showed neurological signs approximately seven days after eating immature corn in its reproductive phase (R2, milky grains). Association of the severity of tongue lesions in broiler chickens with the quality of corn in feed 86

Abstract: The objective of this work was to determine the correlation of the physical attributes of corn and the mycotoxins present in its grains with the severity of tongue lesions in broiler breeder roosters. The occurrence of the aflatoxin, vomitoxin, zearalenone, T-2 toxin, and fumonisin mycotoxins was detected in the corn grains used in the feed formulation for breeder chickens. The percentages of moisture and broken grains were positively correlated (Pearson’s correlation) with the presence of aflatoxin. Milder lesions (grade 1) are positively correlated with aflatoxin and negatively correlated with fumonisin, and the most severe lesions (grade 3) are positively correlated with vomitoxin, fumonisin, and T-2 toxin.

Can Bt Maize Reduce Exposure to the Mycotoxin Fumonisin in South Africa? 31

The mycotoxin fumonisin is associated with oesophageal cancer and birth defects. The Eastern Cape province of South Africa has high levels of fumonisin in maize and oesophageal cancer. This study measures the amounts of the mycotoxin fumonisin in the grain of Bt maize, commercial hybrids, and farmer-saved, local mixtures of seed that were produced and stored by small holders in South Africa. We find that adoption of Bt maize can reduce poor rural consumers exposure to fumonisin. Greater consumption of maize meal milled in modern mills could have a similar impact.

Mycotoxin contamination of diets for beef cattle finishing in feedlot 96

ABSTRACT The objective of this survey was to identify what mycotoxins were present in ingredients used in diets offered to beef cattle in feedlots and their concentrations. The survey covered 30 Brazilian feedlots located in the five major beef-producing states. Samples of total mixed ration (TMR) and ingredients were collected and analyzed for mycotoxin using ultra-performance liquid chromatography coupled to tandem mass spectrometry. Mycotoxin concentrations were further interpreted according to known species-specific sensitivities and normalized according to the principles of toxic equivalent factors (risk equivalent quantities - REQ) expressed in µg kg−1 of aflatoxin B1-equivalent. Equine leukoencephalomalacia associated with ingestion of corn contaminated with fumonisin B1 59

This article describes clinical, etiologic and pathologic diagnosis of an outbreak of equine leukoencephalomalacia. Two samples of the corn consumed by the affected horses contained fumonisin B1 at levels of 46 and 53 µg/g and Fusarium moniliforme, a good in vitro mycotoxin producer.
