The aims of this PhD were to determine (1) the effects of the herbicide diuron on the genome of the oyster, Crassostrea gigas, (2) the possible transmission to the offspring of damaged DNA and (3) its consequences on oyster physiology. The genotoxicity of diuron and the vertical transmission of DNA damage were highlighted after parental exposure to environmental concentrations of diuron during the gametogenesis. Primary DNA lesions were detected in both somatic cells and germ cells of exposed genitors. Detection of the oxidized base 8-oxodGuo in gonads shows that exposure to diuron led to an oxidative stress which likely explains the observed DNA strand breaks in spermatozoa. The coastal areas are subject to multiple anthropogenic pressures including chemical pollution that can pose a real risk to the sustainability of aquatic species. The Arcachon Bay, macrotidal lagoon located on the French Atlantic coast, is the important ecosystem for oyster farming. But for several years, the oyster farms face lower recruitment and high mortality of oyster spat. Chemical contamination of the environment as a factor that may contribute to the observed effects on oysters has so far not been investigated.The present thesis aimed at evaluating through different approaches, of the potential toxicity of heavy metals and pesticides representative of the Arcachon Bay contamination on the early life stages of the Pacific oyster, Crassostrea gigas....

Étude des voies de bioactivation du benzo[a]pyrène et du fluoranthène chez la sole commune (Solea solea) : Profil métabolique et génotoxicité ArchiMer

The aim of this Ph‟D was to study the biotransformation and the genotoxicity of two PAH of interest, namely the benzo[a]pyrene (BaP) and the fluoranthene (Fluo), in the common sole Solea solea, which is a benthic species sensitive to chemical pollution. The use of in vitro exposure models and the application of two different chemical analytical techniques allowed demonstrating that, as in higher vertebrates, the dihydrodiols and the phenols of BaP and Fluo are the major metabolites produced by sole liver enzymes. In vitro, the genotoxic potential of BaP and Fluo was demonstrated in sole, by the production of DNA adducts and oxidative DNA damage. The genotoxicity of Fluo appeared to be 50 fold less important than the genotoxicity of BaP. In the case of BaP,...
